Population structure and genetic diversity in the Portuguese native breeds of pigs Alentejano (AL), Bísaro (BI), and Malhado de Alcoba?a (MA) and the exotic breeds Duroc (DU), Landrace (LR), Large White (LW), and Pietrain were analyzed by typing 22 microsatellite markers in 249 individuals. In general, the markers used were greatly polymorphic, with mean total and effective number of alleles per locus of 10.68 and 4.33, respectively, and an expected heterozygosity of 0.667 across loci. The effective number of alleles per locus and expected heterozygosity were greatest in BI, LR, and AL, and least in DU. Private alleles were found in 9 of the 22 markers analyzed, mostly in AL, but also in the other breeds, with the exception of LW. The proportion of loci not in Hardy-Weinberg equilibrium in each breed analyzed ranged between 0.23 (AL) and 0.41 (BI, LW, and Pietrain), mostly because of a less than expected number of heterozygotes in those loci. With the exception of MA, all breeds showed a significant deficit in heterozygosity (F(IS); P < 0.05), which was more pronounced in BI (F(IS) = 0.175) and AL (F(IS) = 0.139), suggesting that inbreeding is a major concern, especially in these breeds that have gone through a genetic bottleneck in the recent past. The analysis of relationships among breeds, assessed by different methods, indicates that DU and AL are the more distanced breeds relative to the others, with the closest relationship being observed between LR and MA. The degree of differentiation between subpopulations (F(ST)) indicates that 0.184 of the total genetic variability can be attributed to differences among breeds. The analysis of individual distances based on allele sharing indicates that animals of the same breed generally cluster together, but subdivision is observed in the BI and LR breeds. Furthermore, the analysis of population structure indicates there is very little admixture among breeds, with each one being identified with a single ancestral population. The results of this study confirm that native breeds of pigs represent a very interesting reservoir of allelic diversity, even though the current levels of inbreeding raise concerns. Therefore, appropriate conservation efforts should be undertaken, such as adopting strategies aimed at minimizing inbreeding, to avoid further losses of genetic diversity.  相似文献

The uterus plays an essential role in mammalian reproduction and is a target of several hormonal protocols used to improve fertility in cattle. Many studies highlighted the importance of eCG treatment following fixed‐time artificial insemination in improving follicular growth, ovulation and pregnancy rates in cattle. Moreover, eCG has been implicated in angiogenesis, leading to important changes in uterine blood flow and vascularisation. However, there is still a lack of information regarding the specific alterations induced by eCG upon glandular and vascular characteristics of bovine uterus. To investigate the influence of eCG on: uterine thickness and area; uterine artery diameter and area; uterine vascular and gland density; and the expression of the VEGFA‐system, the uteri of crossbred beef cows were collected. All cows were submitted to follicular wave emergence synchronization. On day four of protocol, cows submitted to superovulation (n = 6) received 2000 IU eCG, on day eight, after expected follicular deviation, cows submitted to stimulatory treatment (n = 5) received 400 IU eCG. Control cows (n = 5) did not receive eCG. On day five po cows were subjected to ultrassonographic evaluation and slaughtered for uterine tissue sampling on day six po. Uterine vessels and glands were quantified by the counting point stereological method. The VEGFA‐system was localized in different cellular types, showing no qualitative or quantitative differences in the site of expression or the intensity of the positive signal among the groups. Vascular density was decreased in the endometrium of stimulated and myometrium of superovulated cows compared with the control ones, which showed higher vascular density in the myometrium and endometrium of the ipsilateral uterine horn. The uterine gland density was higher in superovulated compared with stimulated and control cows. Thus, we can infer that stimulatory or superovulatory treatments with eCG influence the vascular density in the endometrium and myometrium in cattle.  相似文献

The present study sought to determine: (i) the effects of Neospora caninum infection and twin pregnancy on plasma pregnancy‐associated glycoprotein‐2 (PAG‐2) concentrations throughout pregnancy and (ii) whether plasma PAG‐2 concentrations could predict abortion in N. caninum‐infected cows. The study was performed on a commercial Holstein‐Friesian dairy herd in northeastern Spain and the final data included those recorded in 53 non‐aborting and 19 aborting animals. Blood samples were collected immediately before pregnancy diagnosis (on Days 40, 90, 120, 150, 180 and 210 post‐insemination) in non‐aborting cows or until the time of abortion detection in aborting cows. General lineal models (GLM) repeated measures anova revealed the different behaviour of PAG‐1 and PAG‐2, and significant effects of Neospora seropositivity, cool season and twin pregnancy on plasma PAG‐2 concentrations throughout gestation (between‐subject effects). In addition, based on the odds ratios, the likelihood of abortion increased in Neospora‐seropositive cows (by a factor of 7.0) compared to seronegative animals and decreased in cows with a high plasma PAG‐2 concentration (>4.5 ng/ml) on Day 120 of pregnancy (by a factor of 0.24), compared to the remaining cows. In conclusion, there is a relationship between plasma PAG‐2 concentrations and the risk of abortion in Neospora‐infected dairy cows. Thus, plasma PAG concentrations measured using anti‐boPAG‐2 antiserum on Day 120 of gestation could serve as an indicator of the abortion risk in N. caninum infected animals; values <4.5 ng/ml indicating a high risk of abortion in chronically infected animals.  相似文献

Abstract

Results are given of a trial in which trifluralin, incorporated in the soil, at rates of 0.5 and 0.75 kg/ha, was combined with fluometuron at 1.6 kg/ha, and with prometryne at 1.0 kg/ha, both incorporated, pre- and post-em. and with noruron + MSMA (1.0:2.2) at 1.7 kg/ha post-em.

Best results were given by incorporated treatments of trifluralin + fluometuron (0.5 + 1.6) kg/ha, or by trifluralin incorporated at 0.5 or 0.75 kg/ha + fluometuron applied pre-em. at 1.6 kg/ha, or by trifluralin incorported at 0.75 kg/ha + prometryne applied pre-em. at 1.0 kg/ha.

It is recommended that noruron + MSMA (1.0:2.2) at 1.7 kg/ha be applied post-em. as a complementary treatment to trifluralin at 0.5 kg/ha in fields where Cyperus spp. are a problem.  相似文献

Action thresholds as decision tools for insecticide application were developed and tested against the major insect pests of rice at four sites in the Philippines over a 13-year period. Action threshold treatments were compared to the farmers' practice, prophylactic insecticide usage, and an untreated check. Yield loss data using the insecticide check method partitioned yield losses over three crop growth stages in the same test fields. Chronic pests that exceeded action thresholds in 79% of fields were whorl maggot Hydrellia philippina Ferino (Diptera: Ephydridae), defoliators Naranga aenescens Moore and Rivula atimeta (Swinhoe) (Lepidoptera: Noctuidae), leaffolders Cnaphalocrocis medinalis (Guenée) and Marasmia patnalis Bradley (Lepidoptera: Pyralidae), and stemborers Scirpophaga incertulas (Walker) and S. innotata (Walker) (Lepidoptera: Pyralidae). Minor chronic pests reached threshold levels in only one site each: rice bug Leptocorisa oratorius (F.) (Koronadal), whitebacked planthopper Sogatella furcifera (Horvath) (Zaragoza) and green leafhopper Nephotettix virescens (Distant) (Guimba); brown planthopper Nilaparvata lugens (Stål) did not exceed a threshold in any field. Stemborers were the most important pest group in terms of yield loss. Despite the insecticide check method underestimating losses, a mean crop loss of 0.62 t/ha was measured which showed ample scope for corrective action. But loss was evenly distributed across crop growth stages (0.15?–?0.24 t/ha) reducing the impact of insecticides. Action threshold treatments overall outyielded the untreated check, more so in the two sites with highest pest density. The benefit of thresholds was to reduce insecticide usage, as a cost saving. However all the practices showed poor economic returns including the farmers' practice. Farmers' practice employed low insecticide dosages and timing was not consistent with pest damage, but yields were often similar to threshold treatments. Farmers appear to use insecticide more for risk aversion than for profit. The best threshold characters when evaluated against resulting pest density and yield loss criteria showed accuracies >?90% correct decisions. Future work is needed to improve the insecticide response rather than monitoring tools. Thresholds need to be incorporated into improved crop management, which was often found suboptimal by farmers, to take advantage of the high levels of tolerance in modern high tillering cultivars. Crop husbandry practices which improve yield potential such as selection of longer maturing varieties and nitrogen fertilizer may be a more effective pest management strategy than insecticides.  相似文献

The use of optimization techniques is well established in forest short-term planning and decision-making. Yet, existing techniques may pose some limitations for tackling with stochastic factors impacting in the execution of forest operations, such as delays, equipment breakdowns and other unexpected events. This paper explores the potential of using optimization techniques in combination with discrete-event simulation (DES) models for planning harvesting and logistics operations acknowledging uncertainty. DES models may be useful for assessing the performance and identifying bottlenecks associated with the execution of the deterministic plans retrieved with optimization techniques, when such stochastic events occur. This paper further presents an approach for the combination of a heuristic and a DES model developed in SIMIO. This approach was used to solve the raw material reception problem (RMRP) at a Portuguese pulp mill. This paper concludes with the analysis of the performance of deterministic schedules for the wood trucks considering uncertainty in their arrival at the mill.  相似文献

This study was conducted to determine the impact of antioxidant (α-tocopherol) supplementation in the cryopreservation extender and three different freezing rates (FRs) on quality of post-thaw semen to elaborate a new protocol for stallion semen cryopreservation. Six ejaculates from each of four stallions were subjected to cryopreservation with a commercial extender (Gent, Minitub Iberia, Spain), without any supplementation (control) or supplemented with 2 mM α-tocopherol. The semen was exposed to three different freezing rates between 5°C and −15°C: slow (5°C/min), moderate (10°C/min), and fast (20°C/min). After thawing, the sperm viability (Sybr-14 and propidium iodide [PI]), mitochondrial membrane potential (MMP) (5,5′,6,6′-tetrachloro-1,1′,3,3′tetraethylbenzimidazolyl carbocyanine iodine), lipid membrane peroxidation (LPO; C11-BODIPY581/591), and apoptosis status (fluorescein isothiocyanate–conjugated annexin V and PI) of the plasmatic membrane of each sample were determined by flow cytometry. For both extenders, the percentage of viable cells was higher for spermatozoa cooled at 5°C/min than at 10°C/min and 20°C/min (P ≤ .05). The FR of 20°C/min demonstrated a lower value of MMP than the FR of 5°C/min and 10°C/min (P ≤ .05). The α-tocopherol extender improved (P ≤ .05) post-thaw membrane LPO; however, it did not improved viability and the apoptosis status of the sperm plasmatic membrane after thawing. In conclusion, results clearly indicate that the cryosurvival of stallion spermatozoa is improved when FR of 5°C/min is used from 5°C to −15°C.  相似文献
