Spices and herbs are natural products or their blends that must be free of extraneous matter content. Conventional production of these products implicates a number of hygienic problems so spices and herbs may be exposed to a wide range of microbial contamination during pre- and post-harvest and they can present high microbial counts. In this study, we have analyzed the microbial quality of 53 samples of spices and dry herbs collected from Spanish markets detecting a contamination of samples of spices with mesophilic aerobic counts (10%) and Enterobacteriaceae (20%). The analysis from herbs showed that the percentage of contamination was 26% in both microbiological values. Pathogenic microorganisms like Staphylococcus aureus, Yersinia intermedia, Shigella spp., Enterobacter spp., Acinetobacter calcoaceticus and Hafni alvei were also isolated from spices and herbs. These unsatisfactory results showed a poor microbiological quality. Spices and dry herbs are used as ingredients in a variety of products prepared in different ways, this fact suggests the need to provide a control system to improve the quality of herbs and spices.  相似文献

This study evaluated the effect of different environmental temperatures in the physiology of Lutjanus guttatus juveniles by analysing their thermoregulatory behaviour, thermal tolerance, oxygen consumption rates and thermal metabolic scope. Jointly, the effect of acclimation and critical temperatures on heat shock protein 70 (Hsp70) and lactate dehydrogenase (Ldh‐a) gene expressions were also analysed using acclimation temperatures of 20, 23, 26, 29 and 32°C. The results showed that the final preferred temperature in juvenile snapper was 26°C with a thermal window of 336.5°C2, which was related to an optimal temperature for their physiology determined by the routine metabolic rate and thermal metabolic scope. At temperatures from 20 to 26°C, the routine metabolic rate and Hsp70 and Ldh‐a genes had the lowest values related to a basal expression level. At acclimation temperatures from 29 to 32°C and after critical thermal maximum (CTmax) limit, the relative expression of Hsp70 and Ldh‐a genes increased significantly, but the main response at CTmax was the upregulation of Hsp70 gene.  相似文献

Isoflavones are a class of compounds present in high amounts in soybean seeds, which can be used for prevention and treatment of several chronic diseases. Proteins present in soybean seeds are the basis for the high nutritional value and versatility of this leguminous species in animal and human feeding. The main goals of this work were to estimate heritabilities for isoflavone contents in soybean seeds and the correlation between isoflavone and protein contents. Commercial variety IAC-100 (high isoflavone and normal protein contents) and the line BARC-8 (low isoflavone and high protein contents) were crossed, and one single F1 plant derived 97 F2 seeds, which were used to obtain F3 seeds. A sample of F3 seeds from each F2 plant was used for isoflavone determination by HPLC and protein by the Kjeldahl method. Six isoflavone forms were detected: daidzin, genistin, glycitin, malonyldaidzin, malonylgenistin and malonylglycitin. Total isoflavone contents ranged from 427.92 to 965.89 μg per gram of dry seed and the protein content ranged from 45.17 to 34.95% in BARC-8 and IAC-100, respectively. Our results indicate that it is possible to select for high isoflavone content in early breeding generations because the broad sense heritabilities for the contents of the various isoflavone forms were higher than 90%. In addition, high correlation values among the contents of the individual isoflavone forms were observed (between 0.80 and 0.98). However, negative correlation values were obtained between isoflavone and protein contents, ranging from −0.51 to −0.37 for the different isoflavone forms. The correlation value of −0.47 between total isoflavone and protein contents confirmed the negative correlation between these two parameters, as reported by other authors. Four barley doubled haploid populations were produced by anther culture from the reciprocal crosses between two six‐row barley cultivars, ‘Plaisant’ and ‘Orria’; the doubled haploid lines (DHLs) derived from each cross were subsequently assigned to weak or vigorous populations according to the weak or vigorous nature of the originating embryos. Well‐formed embryos at day 25 on the induction medium were considered vigorous, whereas embryos maturing later were considered weak. The classification of vigorous and weak was closely associated with the ratio of green to albino plantlets regenerated. A random set of 25 DHLs from each of the four populations were selected for field testing in a replicated trial. Furthermore, a second set consisting of a total of 454 unreplicated DHLs from the four populations were also field assessed for grain yield. Distortion during in vitro culture may impede regeneration of a random array of microspores from a given cross, and may bias genetic estimates of specific trait/marker association in genetic studies. However, no significant differences were detected in this study among the four populations for days to heading, height, grain yield and thousand‐kernel‐weight when measured on the replicated trial of 100 DHLs, nor for grain yield in the second collection of 454 entries. This suggests that the likelihood of producing improved agronomic pure lines is independent of the direction of crossing and, more importantly, independent of the time when embryos matured in the induction media, at least for these particular six‐row cultivars and for the anther culture method used.  相似文献

In this study, an isolate of Magnaporthe oryzae expressing the green fluorescent protein gene (gfp) was used to monitor early events in the interaction of M. oryzae with resistant rice cultivars harbouring a blast resistance (R) gene. In the resistant cultivars Saber and TeQing (Pib gene), M. oryzae spores germinated normally on the leaf surface but produced morphologically abnormal germ tubes. Germling growth and development were markedly and adversely affected in leaves of these resistant cultivars. Penetration of host cells was never seen, supporting the idea that disruption of germling development on the leaf surface might be one of the resistance mechanisms associated with Pib function. Thus, this particular R gene appeared to function in the absence of host penetration by the fungal pathogen. Confocal laser scanning microscopy of Moryzae‐infected susceptible rice cultivars showed the dimorphic growth pattern that is typically observed during the biotrophic and necrotrophic stages of leaf colonization in susceptible cultivars. The suitability of the gfp‐expressing M. oryzae isolate for further research on R‐gene function and identification of resistant genotypes in rice germplasm collections is discussed.  相似文献

The effect of maize and wheat cultivation on a Johnson grass (Sorghum halepense (L.) Pers.) seed population, and its vertical distribution in a 20-cm soil profile was evaluated from autumn to early spring. The experiment was carried out in plots with 2 different types of seed-banks:
1. (A) with 1 generation of seeds distributed on the surface of the soil;
2. (B) with 2 generations of seeds, 1 buried and the other distributed on the surface of the soil.

The wheat system had one tilling operation in April with a mouldboard plough and rotary cultivator and another one in August, before sowing, with the rotary cultivator only. The maize system had 2 tilling-operations with a mouldboard plough and rotary cultivator; the first one in April and the second one in October, before sowing.

Treatments were a combination of cultivation systems and seed-bank types.

A significant seed loss was detected only in those plots under the maize cultivation system with 2 generations of Johnson grass seeds.

Tilling with the mouldboard plough and rotary cultivator significantly changed the vertical pattern of seed distribution. No appreciable modification was observed after cultivation with the rotary cultivator alone.

The mouldboard plough appeared to be more efficient at burying seeds (ca. 80%) than at bringing them to the surface (ca. 38%).

In the maize crop system, after sowing, there was a significantly greater number of full florets (25%) in the 0–4-cm layer, in relation to the total in the soil profile, than in the wheat crop system (7%).

Ecological implications of these cultivation methods on Johnson grass seed dynamics are discussed.  相似文献

Popcorn inbred lines with more efficient nitrogen use are better able to uptake nitrogen from the soil and convert it into higher grain yield, resulting in lower environmental and economic impacts caused by nitrogen fertilization. The objectives of this study were to (i) identify inbred lines superior in nitrogen use (ii) assess the genetic diversity between popcorn inbred lines under high and low N conditions and (iii) investigate the causal effects of several traits in nitrogen use efficiency (NUE). We evaluated 25 popcorn inbred lines under high and low N, and several traits related to NUE and its components were measured. Efficient and inefficient inbred lines under both N levels were identified and can be useful for generating a segregating population for quantitative trait loci mapping. The genetic diversity assessment based on phenotypic traits grouped the inbred lines into four clusters under both N levels, and the efficient inbred lines were grouped together, as were the inefficient lines. The most divergent inbred lines under high N were classified as efficient and intermediate and can be useful to generate a divergent breeding population with a high frequency of favorable genes for NUE. Nitrogen uptake efficiency (NUpE) was considered the most important trait for NUE. To improve the accuracy of selection for NUE, a selection index involving the total root length (TRL), daily growth and NUpE traits under both N levels is recommended, as these traits had high correlations with and direct effects on NUE.  相似文献
