OBJECTIVE: To describe onset and duration of neuromuscular blockade induced by mivacurium chloride and its associated hemodynamic effects at 3 dosages in healthy dogs. ANIMALS: 7 Labrador Retrievers. PROCEDURE: Anesthesia was induced with thiopental and maintained with halothane in oxygen, and dogs were mechanically ventilated to end-tidal P(CO)2 between 35 and 40 mm Hg. Core temperature, end-tidal P(CO)2, and halothane concentration were kept constant throughout the experiment. Neuromuscular function was assessed by evaluation of the train-of-four response to a supramaximal electrical stimulus of 2 Hz applied to the ulnar nerve every 10 seconds. Blood for determination of plasma cholinesterase activity was obtained prior to administration of mivacurium, a bolus of which was administered IV, using a randomized Latin-square design for dosages of 0.01, 0.02, and 0.05 mg/kg of body weight. RESULTS: All dogs had typical plasma cholinesterase activity. After administration of mivacurium, differences were not evident between groups in heart rate, systolic, mean, or diastolic blood pressure, change at any time in heart rate, systolic, mean, or diastolic blood pressure, or pH. Interval from onset to 100% neuromuscular blockade was 3.92+/-1.70, 2.42+/-0.53, and 1.63+/-0.25 minutes at dosages of 0.01, 0.02, and 0.05 mg/kg, respectively. Duration of measurable neuromuscular blockade was 33.72+/-12.73, 65.38+/-12.82, and 151.0+/-38.50 minutes, respectively. Time of onset and duration of effect differed significantly among dosages. CONCLUSIONS AND CLINICAL RELEVANCE: Mivacurium provides good hemodynamic stability at the dosages tested. In dogs, this drug has a rapid onset and long duration of effect.  相似文献

The effects of extensive sward management and patch size on the persistence and colonization of gaps in sown swards was examined by creating gaps of five different sizes (2·3, 7, 10, 14 and 19 cm in diameter) in four different sward treatments: a fertilized sward grazed to 4 cm, i.e. relatively intensive management, and three extensively managed unfertilized swards, which were not grazed or grazed to 4 cm or 8 cm. The swards were originally sown with ryegrass (Lolium perenne L.) and white clover (Trifolium repens L.), but had developed differences in species composition as a result of the management treatments imposed 2 years before and during the experiment. Light quality measurements, i.e. red-far red (R/FR) ratio, were used to determine when the light environment in the gaps no longer differed from that in control, uncut patches and this was used as an estimate of gap persistence. Persistence of gaps depended on both sward management and gap size. Gaps disappeared most rapidly in the ungrazed sward and fertilized 4-cm sward, and most slowly in the unfertilized 8-cm sward. Small gaps persisted for up to 2 weeks in all but 8-cm swards, whereas larger gaps were estimated to persist for up to 20–25 weeks in unfertilized, grazed swards. There was no evidence that the number of grass or dicotyledonous species increased in the gaps compared with the control areas. There were significant positive linear relationships between the vegetation that developed in gaps and that in the control, uncut patches, reflecting the different species composition of the established sward of the grazed (grass-dominant) and ungrazed (Ranunculus repens-dominant) treatments. For total grass dry matter and tiller numbers, as well as L. perenne tiller numbers, there was a small, but significant, effect of both patch size and sward management on the slopes of the regressions between the controls and gaps. The results are discussed in relation to the potential for species composition of sown swards to change as a result of gap creation.  相似文献

A 5-year-old female spayed Shetland Sheepdog Mix dog was evaluated for a history of recent seizure activity, progressive hind limb ataxia, polyuria, and polydipsia and no history of gastrointestinal signs. Physical examination findings included conscious proprioceptive deficits, ataxia, and anterior uveitis along with a hypermature cataract in the right eye. Results of a CBC, serum biochemical profile, urinalysis, and computed tomography scan of the brain were unremarkable. Cerebrospinal fluid (CSF) analysis revealed marked eosinophilic pleocytosis and rare organisms consistent with Prototheca spp within neutrophils and macrophages. On postmortem histologic examination, mononuclear inflammation and numerous intralesional algal organisms, similar to those seen on the cytologic preparation of CSF, were found in the brain, eyes, kidneys, and heart. Abnormalities were not detected on gross and histologic examination of the gastrointestinal tract. Cultures of CSF and subdural/olfactory bulb, but not intestinal tract, yielded growth of Prototheca spp, and PCR analysis and DNA sequencing confirmed the organism as Prototheca zopfii genotype 2. We have reported a rare case of disseminated protothecosis that was diagnosed by evaluation of CSF in a dog presented with neurologic signs and no overt enteric disease. Protothecosis should be considered as a rare cause of seizures, even in the absence of obvious enteric signs, and should be included in the differential diagnosis of eosinophilic pleocytosis.  相似文献

The modification of natural habitats requires behavioural plasticity, which may be challenging for ‘specialist’ species. Quantifying habitat requirements and behavioural responses of specialists to landscape transformation is thus a priority for baseline data to inform conservation practices. Using camera-trap surveys of the forest-dependent Lemon Dove Aplopelia larvata in conjunction with detailed microhabitat-scale covariates, we assessed habitat use during two periods in the year: autumn–winter and spring–summer (which largely encompassed peak breeding). We used occupancy modelling of forest-structural covariates to produce models of the probability of occupancy and detection of Lemon Doves in patches of the Indian Ocean Coastal Belt Forest of South Africa. The average occupancy and detection probability as indicated by the top-performing models was 0.39 ± 0.08 and 0.26 ± 0.05, respectively, during autumn–winter, and 0.37 ± 0.08 and 0.25 ± 0.04, respectively, during spring–summer. Although occupancy and probability of detection remained relatively constant between seasons, there was seasonal variation in the influence of individual covariates for both measures. The overall trend of positive influences on Lemon Dove occupancy was that of complex and diverse habitat structures and high plant species richness. The specific covariates that influenced occupancy positively during spring–summer may reflect the ecological requirements for nestling provisioning for both dietary needs and an avoidance of potential disturbance. Thus, while Lemon Doves may be less habitat-specific during autumn–winter, conservation management plans for safeguarding the breeding success of the species are advised to ensure adequate protection of large forest patches with complex and diverse interior structures and minimal disturbance.  相似文献

To study adherence of Actinobacillus pleuropneumoniae to porcine lower respiratory epithelium, a cell culture model was developed using primary cultures of porcine lung epithelial cells (LEC). Adherence assays were performed and results were compared with data obtained with swine kidney cells (SK6). A. pleuropneumoniae efficiently adhered to LEC with up to 62 bacteria per cell after 2h of incubation. Reference strain of serotype 3 (R3) adhered better to LEC than reference strains of serotypes 1 (R1), 7 (R7) and 8 (R8). Overall the adherence to LEC was more rapid and up to 30-fold more efficient than adherence to SK6 cells. In search for the mechanism involved in the adherence event, we tested the effect of LPS which has previously been demonstrated to cause adherence of the pathogen to upper respiratory epithelium. Adherence assays with LPS transposon mutants demonstrated unaltered (mutant with modification in core/lipid A moiety) or even three-fold more adherence (mutants lacking O antigen) compared to the parent micro-organisms. Purified LPS of strains R1, R3, R7 and R8 did not inhibit adherence of R8 to LEC either, suggesting that LPS and particularly the O-antigen are not essential for adherence of A. pleuropneumoniae to LEC. The efficient, LPS-independent adherence of A. pleuropneumoniae to LEC cells indicates that A. pleuropneumoniae may carry different, cell type-specific adhesins and that primary cultures of lower respiratory epithelium are valuable infection models in studying A. pleuropneumoniae pathogenesis.  相似文献

Traditional methods of teaching intracellular biological processes and pathways use figures or flowcharts with the names of molecules linked with arrows. Many veterinary students, presented with such material, simply memorize the names or chemical structures of the molecules and are then likely to forget the material once the examination is completed. To address this problem, the authors designed, created, and field-tested new teaching media that incorporate realistic three-dimensional (3D) animations depicting the dynamic changes that occur in intracellular molecules during cellular activation. Testing found that veterinary students taught using traditional teaching media (e.g., lectures, handouts, textbooks) are proficient in memorizing the names and order of intracellular molecules but unable to appreciate the interactions between these elements or their spatial relationships within cells. In contrast, more than 90% of veterinary students taught using 3D animations not only recall the facts about the intracellular elements but also develop accurate mental images of the interactions among these molecules and their spatial relationships. These findings strongly suggest that the comprehension of complex biological processes by veterinary students can be enhanced by the use of dynamic 3D depictions of these processes in the classroom.  相似文献

This study investigated the association between presence of respiratory pathogens and development of Canine Infectious Respiratory Disease Complex (CIRDC) in dogs in 5 Canadian small animal clinics. In total, 86 dogs were tested using a commercial PCR respiratory panel; 64 dogs were considered as cases and 22 were control dogs matched by veterinary clinic. No control animals (0/22) were positive for canine parainfluenza virus (CPIV), whereas 27/64 (42%) CIRDC cases were positive. Furthermore, 81% of case dogs tested positive for Mycoplasma cynos, compared with 73% of control dogs. Canine respiratory corona virus (CRCoV) was detected in no control dogs compared with 9.4% of clinical dogs. No animals were positive for any influenza virus type A present in the diagnostic panel. Presence of CPIV was associated (P < 0.01) with the occurrence of CIRDC after adjustment for demographic factors and presence of CRCoV (P = 0.09).  相似文献
