Transgenerational effects of environmental toxins require either a chromosomal or epigenetic alteration in the germ line. Transient exposure of a gestating female rat during the period of gonadal sex determination to the endocrine disruptors vinclozolin (an antiandrogenic compound) or methoxychlor (an estrogenic compound) induced an adult phenotype in the F1 generation of decreased spermatogenic capacity (cell number and viability) and increased incidence of male infertility. These effects were transferred through the male germ line to nearly all males of all subsequent generations examined (that is, F1 to F4). The effects on reproduction correlate with altered DNA methylation patterns in the germ line. The ability of an environmental factor (for example, endocrine disruptor) to reprogram the germ line and to promote a transgenerational disease state has significant implications for evolutionary biology and disease etiology. 相似文献

Seventeen of eighteen orders of living placental mammals are not known before 65 million years ago. The monophyly of each order is well established, but interrelations have been less certain. A superordinal grouping of up to seven extant orders plus a variety of extinct orders, all included within Ungulata ("hoofed" mammals), can be linked to Late Cretaceous mammals from the 85-million-year-old Bissekty Formation, Uzbekistan (and, less certainly, North America and Europe), thus pushing the origin of this major clade back by 20 million years. Whole-farm simulation provides a tool for predicting the effects of farm management strategies on farm productivity and profitability. One such model, the Integrated Farm System Model (IFSM), was modified to allow representation of up to four forage species coexisting in a pasture mixture. The model was calibrated to simulate net herbage accumulation (NHA) observed during six periods of a 2002 experiment in a 3-species pasture in Pennsylvania, USA, composed of orchardgrass (Dactylis glomerata L.), white clover (Trifolium repens L.), and chicory (Cichorium intybus L.). The model also predicted sward botanical composition, total annual NHA, crude protein (CP), and neutral detergent fiber (NDF). Sensitivity analysis showed that predictions of NHA were most sensitive to both chicory and orchardgrass specific leaf area and partitioning of photosynthate to the shoot, as well as chicory photosynthetic efficiency. The model was evaluated against data from the same 3-species pasture in 2003 as well as a 2-species pasture (lacking chicory) from the same experiment in 2002 and 2003. Predictions of total annual NHA in 2- and 3-species pastures were within ±18% of observed values, though predictions of within-season NHA were less accurate. Predictions of botanical composition tended to remain within ±15% of observed values by species. Predictions of within-season CP and NDF concentrations in the whole sward tended to remain within ±22% and ±15%, respectively. Given the generality and realism required of IFSM, the degree of precision in the modified pasture submodel is acceptable for achieving IFSM’s primary goal of comparing the effects of different management scenarios on forage productivity and the long-term profitability and environmental impact of farms. 相似文献

Information on critical area required to ensure the long-term survival of plant populations is lacking for the development of conservation policy. Here we examine the relative importance of critical area and environmental factors to the distribution of a threatened perennial Penstemon cobaea, biennial Stenosiphon linifolius, and annual Centarium texense on limestone glades in southern Missouri. Two of three plants show a glade-size dependent distribution: P. cobaea colonised large and S. linifolius small glades. C. texense successfully colonised glades of all sizes. Environmental requirements of populations of the three species differed, but overlap in range was evident. Diffuse competition may account for the observed patterns in distribution but further study is required. In the interim, we suggest that a useful approach for developing a conservation policy for natural plant communities is the intensive study of a few plant species with contrasting life-history strategies. 相似文献

Culture filtrate protein (CFP) vaccines have been shown to be effective in small animal models for protecting against tuberculosis while immunisation with these types of vaccines in cattle has been less successful. A study was conducted in cattle to evaluate the ability of selected adjuvants and immunomodulators to stimulate protective immune responses to tuberculosis in animals vaccinated with Mycobacterium bovis CFP. Seven groups of cattle (n=5) were vaccinated with M. bovis CFP formulated with either Emulsigen or Polygen adjuvant alone or in combination with a specific oligodeoxynucleotides (ODN), polyinosinic acid: polycytidylic acid (poly I:C) or poly I:C and recombinant granulocyte-macrophage colony stimulating factor. Two additional groups were vaccinated subcutaneously with BCG or non-vaccinated. In contrast to the strong interferon-gamma (IFN-gamma) responses induced by BCG, the CFP vaccines induced strong antibody responses but weak IFN-gamma responses. The addition of CpG ODN to CFP significantly enhanced cell-mediated responses and elevated antibody responses to mycobacterial antigens. Of the CFP vaccinated groups, the strongest IFN-gamma responses to CFP vaccines were measured in animals vaccinated with CFP/Emulsigen+CpG or CFP/Polygen+CpG. The animals in these two groups, together with those in the BCG and non-vaccinated groups were challenged intratracheally with virulent M. bovis at 13 weeks after the first vaccination and protection was assessed, by examination for presence of tuberculous lesions in the lungs and lymph nodes, 13 weeks later at postmortem. While BCG gave the best overall protection against tuberculosis, significant protection was also seen in animals vaccinated with CFP/Emulsigen+CpG. These results establish an important role for CpG ODN in stimulating protective Th1 responses to tuberculosis in cattle and indicate that a sub-unit protein vaccine can protect these animals against tuberculosis. 相似文献
