Postnatal development of leukocyte subset composition and activity in dogs   总被引:1,自引:0,他引:1  
The aim of the presentation is to summarise our data on the counts and activity of circulating canine leukocytes at birth and on their changes in the first 3 months of life. On day 1, neutrophil counts were almost three times higher than lymphocyte counts. During the first week of life, a decrease of neutrophil and an increase of lymphocyte counts, resulting in a predominance of lymphocytes, were observed. Neutrophil counts reached values comparable with those in adults in 1 month. Lymphocyte counts were higher than those in adults during the first 3 months. From birth to the age of 3 months, the phagocytic activity of neutrophils was nonsignificantly higher than in young adults. When compared with adults, the peripheral blood of new-born pups contained a lower proportion of T lymphocytes (detected by CD3 and CD5 markers), with a very low percentage of CD8(+) cells and a higher proportion of CD21(+) B lymphocytes. The counts of individual subsets levelled out during the first 3 months of life, although the proportion of CD21(+) B cells remained higher all the time. Lymphocytes of new-born pups were able to respond to nonspecific mitogen stimulation. Spontaneous proliferation in vitro was higher during the first week of life. Although in vitro stimulation of lymphocytes with Concanavalin A in some pups was comparable with that of adult dogs, mean activity was weaker. Pups with zero or very low levels of maternal antibodies were able to develop specific immune responses to a parvovirus antigen as early as at 2 weeks of age. On the basis of these data, we assume that pups are born with an immune system that can respond to external stimuli. Nevertheless its development continues in the postnatal period and some parameters differ from adult values for at least 3 months after birth.  相似文献

松树材小蠹Xyleborus festivus Eichhoff是东南亚地区为害松属植物的重要害虫,为掌握该虫在中国南方的发生情况及其共生真菌种类,本文通过检视标本馆标本和野外调查,并对采集的标本携带的真菌进行分类培养,利用分子生物学的方法对获得的真菌种类进行鉴定。本文提供了松树材小蠹的鉴别特征,记录了其在中国南方的发生情况、地理分布、寄主等,同时分离出共生真菌7种,并确定其储菌囊中的优势真菌——Raffaelea cf.arxii。松树材小蠹对寄主植物的危害极有可能引起或加速其他病害发生,应引起相关部门的重视。  相似文献

IS901 RFLP analysis of 36 Mycobacterium avium subsp. avium (MAA) isolates from 15 pheasants (Phasianus colchicus) and two goshawks (Accipiter gentilis) from four pheasant farms was performed. Using this method, six different IS901 RFLP types (E, F, G, M, Q, and V) were identified. The distribution of IS901 RFLP profiles was tightly linked to individual flocks. Matching IS901 RFLP profiles observed in the present study indicate MAA transmission between pheasants and goshawks in the same locality. In two flocks, different pheasants within a flock as well as in various organs of five individual pheasants were found to have two distinct IS901 RFLP profiles.  相似文献

The time period during which oocyte and spermatozoa retain their fertilizing ability after contacting with water was evaluated in pikeperch (Sander lucioperca). In addition, success of in vitro fertilization was examined regarding to the sperm‐to‐oocyte ratio (SOR). In the first trial, oocytes were placed in Petri dishes containing 5 ml of the hatchery water, to which freshly collected and pooled sperm were added to each sample at 0, 15, 30, 60, 90, 120, 150 and 180 s post oocyte activation. The oocytes retained their fertility for at least 30 s after contacting with water. The second trial tested the maximum time period during which spermatozoa retained fertilizability after contacting with water. Milt (50 μl) was collected from each male and added to 5 ml of water in Petri dishes. Thereafter, oocytes were added at 0, 5, 15, 30, 60 and 75 s post‐sperm activation. Delays exceeding 10 s affected negatively the fertilization success. The third trial examined the optimum SOR; in which was found that 100 × 103 spermatozoa per oocyte were the minimum ratio to ensure fertilization rates above 70%. Overall, the data clarified some biological interactions of gametes in the artificial propagation of pikeperch.  相似文献

Based on cluster groups from the first-round analyses of the Third International Swine CD Workshop, 38 monoclonal antibodies (MAbs) including eight internal controls were analysed by flow cytometry (FCM) and immunohistochemistry (IH) in the second-round analysis of the B-cell section of this workshop. Targets in this section included peripheral blood lymphocytes and cells isolated from ileal Peyer's patches (PP), mesenteric lymph nodes (MLN) of adult animals, bone marrow cells from newborn piglets and thymus cells isolated from foetuses at day 105 of gestation.Immunohistochemistry of these 38 MAbs identified four sets, whose ligands were co-expressed with CD21, which showed a tissue distribution compatible with specificity for cells including those of the B-cell lineage. Another group of miscellaneous antibodies appeared to identify other cells, several antibodies were negative. Two-colour flow cytometry (2C-FCM) was carried out by pairing each antibody of interest with antibodies to SWC7, CD21, sIgM and a polyclonal rabbit anti-swine immunoglobulin antiserum (RaSwIg).The anti-CD21 MAb BB6-11C9 (no. 20) and IAH-CC51 (no. 19), established in previous workshops, as well as the cross-reactive anti-human CD21 B-1y4 (no. 146), clustered together in FCM analyses of the first round and showed similar cellular distribution in IH. A further cluster was formed by the standard CC55 (no. 55) and 2A10/8 (no. 102) submitted as SWC7 specific. The second SWC7 standard 2F6/8 (no. 100) clustered separately, but IH showed an identical pattern of reactivity to the other SWC7 MAb.Unfortunately, this work could not identify any other novel clusters with specificity for B-cells, as the statistical clustering of other MAbs could not be substantiated by IH or subsequent two-colour-FCM work. However, we could identify MAb with similar cellular distribution. The ligands for the cross-reactive anti-human CD40 G28.5 (no. 25) and STH224 (no. 153) were expressed on very similar targets, similarly the ligands for the MAb pair JM1H1 (no. 139) with BB6-10A10 (no. 142) and the MAb pair 3F7/11 (no. 115) with 1C2F10 (no. 187).  相似文献

The demise of traditional woodland management techniques, such as coppicing or woodland pasture, is causing a gradual closure of formerly sparse lowland woods across Central Europe. It is established that these processes threaten such organisms as butterflies and higher plants. Effects on other groups, such as epigeic invertebrates, are little known, hindering rational conservation decisions. We investigated the effects of stand openness on three epigeic groups, carabids (Carabidae), arachnids (Araneae + Opiliones) and myriapods-isopods (Chilopoda + Diplopoda + Oniscidea), in a lowland deciduous wood in the Czech Republic. Situating some of the traps in an intensive deer park allowed a simultaneous assessment of effects of high vs. low ungulate densities. Carabids reached the highest species richness in either sparse stands with low game or dense stands, high game. More arachnids occurred under low game and in sparse stands than under high game density and in dense stands. The highest richness of myriapods-isopods was in sparse stands with low game. Ordinations revealed that species of conservation concern (‘relic species’ according to C. European authors) tended to be associated with sparse stands and low game. A considerable proportion of epigeic woodland invertebrates, including many species of conservation concern, depends on preserving highly heterogeneous sparse canopy conditions. Restoring such conditions in selected areas will benefit these sensitive open woodland specialists while causing minimum harm to specialists of dense stands, that likely prosper in commercially managed high forests.  相似文献

Helicobacter (H.) suis colonizes the stomach of pigs and is the most prevalent gastric non-H. pylori Helicobacter species in humans. Limited information is available on host immune responses after infection with this agent and it is unknown if variation in virulence exists between different H. suis strains. Therefore, BALB/c and C57BL/6 mice were used to compare colonization ability and gene expression of various inflammatory cytokines, as determined by real-time PCR, after experimental infection with 9 different H. suis strains. All strains were able to persist in the stomach of mice, but the number of colonizing bacteria at 59 days post inoculation was higher in stomachs of C57BL/6 mice compared to BALB/c mice. All H. suis strains caused an upregulation of interleukin (IL)-17, which was more pronounced in BALB/c mice. This upregulation was inversely correlated with the number of colonizing bacteria. Most strains also caused an upregulation of regulatory IL-10, positively correlating with colonization in BALB/c mice. Only in C57BL/6 mice, upregulation of IL-1β was observed. Increased levels of IFN-γ mRNA were never detected, whereas most H. suis strains caused an upregulation of the Th2 signature cytokine IL-4, mainly in BALB/c mice. In conclusion, the genetic background of the murine strain has a clear impact on the colonization ability of different H. suis strains and the immune response they evoke. A predominant Th17 response was observed, accompanied by a mild Th2 response, which is different from the Th17/Th1 response evoked by H. pylori infection.  相似文献

Diurnal and seasonal tree water storage was studied in three large Douglas-fir (Pseudotsuga menziesii [Mirb.] Franco) trees at the Wind River Canopy Crane Research site. Changes in water storage were based on measurements of sap flow and changes in stem volume and tissue water content at different heights in the stem and branches. We measured sap flow by two variants of the heat balance method (with internal heating in stems and external heating in branches), stem volume with electronic dendrometers, and tissue water content gravimetrically. Water storage was calculated from the differences in diurnal courses of sap flow at different heights and their integration. Old-growth Douglas-fir trees contained large amounts of free water: stem sapwood was the most important storage site, followed by stem phloem, branch sapwood, branch phloem and needles. There were significant time shifts (minutes to hours) between sap flow measured at different positions within the transport system (i.e., stem base to shoot tip), suggesting a highly elastic transport system. On selected fine days between late July and early October, when daily transpiration ranged from 150 to 300 liters, the quantity of stored water used daily ranged from 25 to 55 liters, i.e., about 20% of daily total sap flow. The greatest amount of this stored water came from the lower stem; however, proportionally more water was removed from the upper parts of the tree relative to their water storage capacity. In addition to lags in sap flow from one point in the hydrolic pathway to another, the withdrawal and replacement of stored water was reflected in changes in stem volume. When point-to-point lags in sap flow (minutes to hours near the top and stem base, respectively) were considered, there was a strong linear relationship between stem volume changes and transpiration. Volume changes of the whole tree were small (equivalent to 14% of the total daily use of stored water) indicating that most stored water came from the stem and from its inelastic (sapwood) tissues. Whole tree transpiration can be maintained with stored water for about a week, but it can be maintained with stored water from the upper crown alone for no more than a few hours.  相似文献

Dogs represent both an important veterinary species and a convenient model for allogeneic hematopoietic stem cell transplantation. Even though anti-canine CD34 antibodies have recently become available, little is known about hematopoietic lineages in dogs, partially because CD34- cells have been ignored in all analyses performed so far. In this study, we have focused on the bone marrow mononuclear compartment to provide an additional piece of information on the phenotype of CD34+ progenitors and to identify the dominant CD34- population. We have shown that, in contrast to the adults, mature lymphocytes are scarce in neonatal dog bone marrow. Using cross-reactive antibodies against CD79alpha we have shown that the B lineage of hematopoiesis strongly prevails. CD34+ cells were shown to be positive for MHC class II and SWC3, a member of the signal regulatory protein family.  相似文献

The developing porcine fetus offers an excellent opportunity for the study of lymphocyte development. Studies on B cell, alphabeta T cells and gammadelta T cells in the last decade have expanded our knowledge of lymphocyte development in pigs. These studies have revealed several interesting differences between swine, mice and humans. For example, porcine peripheral lymphocytes include CD4+CD8+ alphabeta T cells and an abundance of gammadelta T cells that may even prevail over the alphabeta population. There are numerous CD2- gammadelta T cells in the blood and a large number of CD8alphaalpha-bearing cells that include NK cells, conventional gammadelta and alphabeta T cells. All porcine B lymphocytes are CD25(lo) and sIgM+ B cells may differ in the expression of CD2 antigen. Unlike mice, porcine B cells appear approximately 2 weeks before T cells and progenitors undergo VDJH rearrangement at 20th day of gestation (DG20) in the yolk sac and DG30 in the fetal liver before consummating high level lymphogenesis in the bone marrow after DG45. Early B cells show an unexpectedly high proportion of in-frame rearrangements, undergo switch recombination in thymus on DG60 and use N-region insertion from the time of the earliest VDJ rearrangement. The genomic repertoire of VH, DH and JH genes is small compared to mice and humans and swine appear to depend on junctional diversity for the majority of their repertoire. The limited VH repertoire of swine contrasts sharply with the porcine TCRbeta repertoire, which is extensive, extraordinarily conserved and nearly identical to that in humans. Therefore, swine present an example of two highly related receptor systems that have diverged in the same species.  相似文献
