To evaluate the prophylactic and therapeutic effects of an ivermectin controlled-release capsule (IVM-CRC) on the productivity of growing sheep infested with Psoroptes ovis 24 male and 24 female Merino landrace lambs, 5-6 months old and weighing 21.2-35.0 kg, were used. Sixteen replicates of three animals were formed based on sex and Day 0 body weight. Within each replicate animals were randomly allocated to one of three groups: untreated control; IVM-CRC on Day 0; IVM-CRC on Day 84. For treatment an IVM-CRC for sheep weighing 20-45 kg was used which is designed to deliver ivermectin at a minimum dose of 20 microg/kg per day for 100 days. The lambs were infested with 50-60 P. ovis mites each on Days 14 and 21. Mites in skin scrapings were counted on Days 70, 84, 98, 112 and 126. Body weight and feed consumption were measured every 2 weeks from Day 0 to 126. The animals were slaughtered on Day 127 and their carcasses evaluated. The IVM-CRC treatment on Day 0 prevented the establishment of P. ovis. All untreated lambs became infested. The lambs treated with an IVM-CRC on Day 84 became mite-free from Day 112 onwards. The lambs treated on Day 0 had significantly (p<0.05) greater body weight gain from Day 0 to 84 (13.9 kg) and Day 0 to 126 (20.9 kg) than the untreated controls (9.6 and 12.8 kg, respectively) and the sheep treated on Day 84 (8.4 and 14.9 kg, respectively). Feed consumption (Days 0-126) for sheep treated with the IVM-CRC on Day 0 was higher than for sheep treated on Day 84 (p<0.05) and for the untreated controls (p<0.1). The carcasses of sheep treated with the IVM-CRC on Day 0 had significantly (p<0.05) higher warm and cold weights, carcass yield, rib eye area and back fat thickness than the untreated control group and the sheep treated with the IVM-CRC on Day 84. The sheep treated with the ivermectin CRC on either Day 0 or 84 had significantly (p<0.05) better muscle scores and lower muscle pH 1h post-slaughter than the untreated controls. There was no significant (p>0.1) difference between warm and cold carcass weights, carcass yield and rib eye area between sheep treated on Day 84 and untreated controls.  相似文献

A collaborative study, to validate the use of SDS-PAGE and urea IEF, for the identification of fish species after cooking has been performed by nine laboratories. By following optimized standard operation procedures, 10 commercially important species (Atlantic salmon, sea trout, rainbow trout, turbot, Alaska pollock, pollack, pink salmon, Arctic char, chum salmon, and New Zealand hake) had to be identified by comparison with 22 reference samples. Some differences in the recoveries of proteins from cooked fish flesh were noted between the urea and the SDS extraction procedures used. Generally, the urea extraction procedure appears to be less efficient than the SDS extraction for protein solubilization. Except for some species belonging to the Salmonidae family (Salmo, Oncorhynchus), both of the analytical techniques tested (urea IEF, SDS-PAGE) enabled identification of the species of the samples to be established. With urea IEF, two laboratories could not differentiate Salmo salar from Salmo trutta. The same difficulties were noted for differentiation between Oncorhynchus gorbuscha and Oncorhynchus keta samples. With SDS-PAGE, three laboratories had some difficulties in identifying the S. trutta samples. However, in the contrast with the previous technique, SDS-PAGE allows the characterization of most of the Oncorhynchus species tested. Only Oncorhynchus mykiss was not clearly recognized by one laboratory. Therefore, SDS-PAGE (Excel gel homogeneous 15%) appears to be better for the identification, after cooking, of fish such as the tuna and salmon species which are characterized by neutral and basic protein bands, and urea IEF (CleanGel) is better for the gadoid species, which are characterized by acid protein bands (parvalbumins). Nevertheless, in contentious cases it is preferable to use both analytical methods.  相似文献

Giardia duodenalis is a relevant gastrointestinal protozoan pathogen of humans and animals. This species complex consists of eight genetically different assemblages. Assemblages A and B are pathogenic to humans and pets, thus confer zoonotic potential. The risk of zoonotic transmission has been controversially discussed. The aim of this monocentric cross‐sectional pilot study was to investigate G. duodenalis assemblages in humans and pets living in common households in Berlin/Brandenburg (Germany). Samples from dogs, cats and humans sharing the same households were screened for Giardia infection by antigen‐detecting assays. All human samples were additionally analysed by a Giardia‐specific qPCR. Cyst quantification and sequences of different gene loci (triosephosphate isomerase (tpi), glutamate dehydrogenase (gdh), β‐giardin (bg) and for dogs SSUrDNA) were analysed. A total of 38 households (31 households with dogs and seven with cats) with 69 human individuals participated in the study. Initial antigen‐detecting assays revealed Giardia‐positive results for 13 (39%) canine, one (14%) feline and one human sample. Reanalysis of the human samples by qPCR revealed two more positive specimens (4%). Two of these three samples were identified as assemblage B at all tested loci. Success rate of assemblage typing for pet samples was generally low and comprised mainly the SSUrDNA locus only. Overall, six of 13 Giardia‐positive canine samples were typable (2× A, 1× co‐infection: A and B, 1× C; 2× D). One pair of samples (dog and human) from the same household had a similar but not identical assemblage B sequence at tpi locus. Assemblage A was also detected in the dog specimen, which hampered sequence analysis. In conclusion, although exhibiting limitations due to the sample size, our study highlights the need for better and standardized typing tools to distinguish G. duodenalis strains with higher resolution in order to perform proper case–control studies for a realistic estimation of zoonotic risk.  相似文献

【目的】 了解并掌握鸭坦布苏病毒(Duck Tembusu virus,DTMUV)流行特点及病毒生物学特性,为DTMUV防治提供理论依据和技术支撑。【方法】 通过细胞及鸡胚接毒试验对河北某鸭场10只具有典型临床症状的发病鸭进行病毒分离,用RT-PCR、透射电镜观察、Western blotting及间接免疫荧光试验(IFA)等方法鉴定,进行动物回归试验测定病毒毒力并对其进行囊膜蛋白遗传进化分析。【结果】 分离到的病毒可在DF-1细胞上稳定增殖产生典型细胞病变效应(CPE)并致死鸡胚;病毒纯化后经电镜观察可见直径30~60 nm的病毒粒子;RT-PCR结果显示,在约270 bp处可见单一条带,与DTMUV预期大小一致;Western blotting结果显示,在60 ku处有特异性条带,与E蛋白大小一致;IFA结果表明,接种病毒的DF-1细胞胞质中可见明亮的特异性荧光,以上结果均表明分离的病毒为DTMUV。将分离到的病毒命名为AX2020株,AX2020株经肌内注射感染北京鸭后感染率高达100%,发病鸭产生神经症状及腹泻等典型临床症状;经序列比对发现AX2020株和GA株(MK907880.1)相似性最高,与SD14毒株(MH748542.1)亲缘关系较远。与商品灭活疫苗毒株HB2010株(MN649262.1)和活疫苗毒株FX2010株(MH414568.1)相比,AX2020株第93、277和487位氨基酸发生了的突变。【结论】 成功分离得到1株DTMUV AX2020株,分离毒株对北京鸭具有较强的致病性,AX2020株的囊膜蛋白与国内疫苗毒株相比,已经发生了氨基酸位点的突变,结果为鸭坦布苏病毒病的流行病学及后续疫苗相关研究奠定了一定的基础。  相似文献

Analysis of restriction fragment length polymorphism (RFLP) profiles of a 464 bp amplicon obtained from the mitochondrial cytochrome b gene was used to differentiate between several different fish species. The method was tested by a collaborative study in which 12 European laboratories participated to ascertain whether the method was reproducible. Each laboratory was required to identify 10 unknown samples by comparison with RFLP profiles from authentic species. From a total of 120 tests performed, unknown samples were correctly identified in 96% of cases. Further work attempting to use the method to analyze mixed and processed fish samples was also performed. In all cases the species contained within mixed samples were correctly identified, indicating the efficacy of the method for detecting fraudulent substitution of fish species in food products.  相似文献

The pharmacokinetics of afoxolaner and milbemycin oxime (A3 and A4 forms) in dogs were evaluated following the oral administration of NexGard Spectra ® (Merial), a fixed combination chewable formulation of these two active pharmaceutical ingredients. Absorption of actives was rapid at levels that provide the minimum effective doses of 2.5 mg/kg and 0.5 mg/kg of afoxolaner and milbemycin oxime, respectively. The time to maximum afoxolaner plasma concentrations (tmax) was 2–4 h. The milbemycin tmax was 1–2 h. The terminal plasma half‐life (t1/2) and the oral bioavailability were 14 ± 3 days and 88.3% for afoxolaner, 1.6 ± 0.4 days and 80.5% for milbemycin oxime A3 and 3.3 ± 1.4 days and 65.1% for milbemycin oxime A4. The volume of distribution (Vd) and systemic clearance (Cls) were determined following an IV dose of afoxolaner or milbemycin oxime. The Vd was 2.6 ± 0.6, 2.7 ± 0.4 and 2.6 ± 0.6 L/kg for afoxolaner, milbemycin oxime A3 and milbemycin oxime A4, respectively. The Cls was 5.0 ± 1.2, 75 ± 22 and 41 ± 12 mL/h/kg for afoxolaner, milbemycin oxime A3 and milbemycin oxime A4, respectively. The pharmacokinetic profile for the combination of afoxolaner and milbemycin oxime supports the rapid onset and a sustained efficacy for afoxolaner against ectoparasites and the known endoparasitic activity of milbemycin oxime.  相似文献

The options for the treatment and control of sheep scab (psoroptic mange) have been increased in recent years through the introduction of the endectocides ivermectin, doramectin and moxidectin. Whilst therapeutic efficacy is good, the current injectable formulations offer limited protection against re-infestation with Psoroptes ovis. An intraruminal controlled-release formulation of ivermectin has been developed to provide therapeutic and prophylactic activity against a range of sensitive endo- and ecto-parasites of sheep for 100 days after administration. These ivermectin boluses are designed to release ivermectin at 20-40 microg/kg/day over 100 days and were developed for use in sheep of 20-90 kg bodyweight. Several controlled therapeutic and prophylactic trials against sheep scab have been conducted under a variety of protocols with such boluses in Europe and South America. The results of these studies indicate that the bolus provides 100% therapeutic efficacy against established P. ovis infestations and equivalent prophylactic efficacy against challenge infestations administered during the active life of the bolus.  相似文献

In this study, average weight, colour, soluble solids content and titratable acidity were determined as quality parameters of strawberry fruits while ascorbic acid, total anthocyanins, total phenolics, total flavonoids, minerals and trace elements content as nutritional parameters. All these attributes along with the DPPH radical scavenging activity were screened in two strawberry genotypes grown on the same site at the ripe stage. Also, the phenolic profiles of the strawberry fruits were obtained by HPLC. Except colour, results showed no significant differences between cultivars in quality parameters. Ascorbic acid found for ‘Premial’ cv. (81.62?mg/100?g) was twice the amount found in ‘Magic’ cv. (42.15?mg/100?g), total phenolics varied from 207.95 (cv. ‘Premial’) to 249.98 (cv. ‘Magic’) mg GAE/100?g while total flavonoid content in fruits of ‘Magic’ cultivar (79.56?mg/100?g) was 2?fold higher than the one of the ‘Premial’ cv. The quercetin content ranged from 2.72 to 4.66?mg/100?g, whereas free ellagic acid ranged from 6.79 to 9.03?mg/100?g. The pale shiny fruits of ‘Premial’ cv. (higher L*, ho and chroma index) had a lower total anthocyanin content (19.07?mg CG/100?g) than the bluish fruits of the ‘Magic’ cv. (23.96?mg CG/100?g). An inverse relationship between the levels of ascorbic acid and anthocyanin contents was also noticed in strawberry samples. Significant differences were found also in minerals and trace elements content of the two strawberry cultivars. The results revealed the importance of genetic background for the content of antioxidant compounds in strawberry fruits.  相似文献
