Summary Variation in the capacity to regenerate shoots and morphogenetic potential were investigated by cyclic culture and regeneration of Nicotiana tabacum var. BEL W3. Explants were excised from different shoots formed by the same callus line. Differences in the organogenetic in vitro response among regenerated plantlets was observed in two subsequent regenerate generations. A progressive decrease of the regenerative capacity was observed across callus line generations.  相似文献

BACKGROUND: Pesticide residues remaining on food represent a potential risk to consumer's health. Determination of these pesticide residues involves tedious procedures of analysis with regard to time and laboratory work. Near‐infrared spectroscopy (NIRS) is a possible alternative to these methods. The aim of this research was to evaluate the ability of NIRS to classify two pesticides used for controlling apple fruit pests according to their concentration. Different solutions were prepared, based on the dose recommended by the pesticide producers for apple pest treatments. Spectra were acquired on a spectrophotometer from liquid samples belonging to these solutions. RESULTS: Calibration models were developed from liquid samples, following the soft independent modelling of class analogy (SIMCA) analysis method. These models classified between 99 and 100% of the validation samples belonging to different pesticide concentration solutions even at the maximum residue limit level of these products in apple fruit. CONCLUSIONS: NIRS technology shows a high potential for identifying pesticides in liquid samples, according to their concentration, at the levels required by the legislation. © 2012 Society of Chemical Industry  相似文献

The 5-n-alkylresorcinol (AR) contents of thirty different cultivars of durum wheat grown in two years (2009 and 2010) in two Italian locations were determined and related to the total soluble phenolic content (TPC) and antiradical activity (AA). On average, AR and TPC ranged from 161.3 to 405.8 μg/g (dry matter, DM) and from 19.0 to 192.4 μg/g (DM), respectively. AA (EC50 values) ranged from 70.9 to 289.2 mg of dry whole milled wheat grain (DM). The results showed that the environment (E) and the genotype (G), as well as their interactions (G × E), significantly influenced the phytochemical profiles of the samples. The contribution of G × E to the total variance was much lower than that due to the separate effects (G and E). Principal component analysis identified genotypes that were richer in ARs and more stable across environments. There were significant negative correlations between ARs and TPC (p < 0.05) and between TPC and AA (p < 0.01), but not between ARs and AA. Graphical representation was efficient in summarizing the overall antiradical profiles of the durum wheat grain in each environment.  相似文献

Seven cultivated Triticum species or subspecies (Triticum monococcum, Triticum turgidum ssp dicoccum, T. turgidum ssp. durum, T. turgidum ssp turanicum, Triticum timopheevii, Triticum aestivum and Triticum zhukovskyi) were compared for their contents of bioactive compounds, mainly 5-n-alkylresorcinols (ARs). Multivariate analysis of variance and principal component analysis were used to evaluate the differences in the phytochemical profiles and to establish the relationships among variables. Significant differences were observed for both total phenol (TP) and AR content. The highest AR level (377 μg/g) was observed in Triticum dicoccum, which also exhibited the highest variability for these compounds (298–436 μg/g). By contrast, the lowest AR content (286 μg/g) was found in Triticum durum. C21:0 was the main homologue chain in all the samples, its value ranging between 54.5% in T. durum and 41.2% in T. aestivum. The T. dicoccum and T. monococcum wheats had relatively low amounts of TP, whereas Triticum turanicum (215 mg/kg), T. timopheevii (250 mg/kg) and T. zhukovskyi (286 mg/kg) had approximately 3-fold higher TP levels. These results suggest that there are opportunities for breeding wheat varieties with superior health benefits and for promoting the use of ancient Triticum crops as novel sources of healthy food.  相似文献

Cassava is an important crop with great economic and social significance in many countries. Most of its biomass is usable: storage roots can be destined for industry or fresh consumption, leaves are a source of protein, vitamins, and minerals, and stems can be used as propagation material. Due to its vegetative propagation, cassava yield is greatly constrained by viral diseases. Cassava common mosaic virus (CsCMV), one of the most widely spread viruses in Latin America, is the only cassava-infecting potexvirus able to cause disease in a single infection: cassava common mosaic disease (CCMD). Here, we evaluated the effect of an Argentinian strain of CsCMV on several agronomic traits. Field trials were established in north-eastern Argentina for three successive cropping seasons, 2016/17, 2017/18, and 2018/19. CsCMV presence was confirmed by PCR or ELISA tests in mechanically inoculated cassava plants. The evaluated traits were plant height (PH), total fresh weight (TFW), shoot fresh weight (SFW), diameter and FW of tuberous root per plant (TRD and FWTR), FW of each individual tuberous root (FWiTR), length and number of tuberous roots (TRL and TRN), harvest index (HI), and estimated yield of tuberous root (EY) in both infected and noninfected plants. Significant yield losses were recorded in infected plants. The most affected traits were EY and FWTR (both showing 44.3% of losses), followed by TFW, SFW, TRN, and FWiTR (37.8%, 33.5%, 29.9%, and 24.8% of losses, respectively). These losses are extremely high for Argentina, where this crop provides food security and demand for the raw material is unsatisfied.  相似文献

Purpose

Green areas in urban parks are special ecosystems because of social and ecological benefits.

Materials and methods

We investigated the ecological impacts of green spaces by comparing the properties of soils, with emphasis on metals, in green spaces and bare soil playgrounds in four urban parks in Murcia City (SE Spain).

Results and discussion

Organic matter (OM) contents in green areas are higher than playground soils due to litter decomposition from grass. Cation exchange capacity in green areas ranged from 10 to 14 compared to 2 cmol+kg?1 in playground soils, showing the effect of OM (and clay) contents in green areas due to the presence of vegetation. Soil total metal contents in urban parks are below the environmental threshold values for soils in Spain and other European countries, and come from atmospheric deposition and fertilizers application. Diethylenetriaminepentaacetic acid (DTPA)-extractable soil Cd in green areas (4% of total Cd) is lower than in playground soils (58% of total Cd). We attributed the reduced DTPA- and water-extractable Cd, Co, Cr, Mn, and Mo to the formation of stable complexes between metals and OM produced from the continued decomposition of roots of grasses in green spaces.

Conclusions

Grasses accumulate less metal than soils except in roots where Cr and Zn are higher than soils by as much as 31 and 17%, respectively. We conclude that grasses increased (up to 8×) soil OM content in green areas to significantly increase the ability of soils to accumulate nutrients for plants, and to retain and reduce the mobility and toxicity of metals in soils in urban parks.  相似文献

The spindle checkpoint delays sister chromatid separation until all chromosomes have undergone bipolar spindle attachment. Checkpoint failure may result in chromosome mis-segregation and may contribute to tumorigenesis. We showed that the human protein Hec1 was required for the recruitment of Mps1 kinase and Mad1/Mad2 complexes to kinetochores. Depletion of Hec1 impaired chromosome congression and caused persistent activation of the spindle checkpoint, indicating that high steady-state levels of Mad1/Mad2 complexes at kinetochores were not essential for checkpoint signaling. Simultaneous depletion of Hec1 and Mad2 caused catastrophic mitotic exit, making Hec1 an attractive target for the selective elimination of spindle checkpoint-deficient cells.  相似文献

Ehrlichia canis has a worldwide distribution, but clinical manifestations may vary geographically. We selected 129 dogs to determine prevalence of ehrlichiosis in dogs with anemia, thrombocytopenia, or ticks presented to a Veterinary Teaching Hospital in South Brazil. Of the 129 dogs, 68 carried the brown dog tick (Rhipicephalus sanguineus), 61 had thrombocytopenia (platelet count <150,000/microl), and 19 had anemia (PCV < 22%). Twenty dogs fulfilled more than one inclusion criteria. Ehrlichiosis was diagnosed by positive amplification of ehrlichial DNA by PCR using primers ECC and ECB that amplify a sequence of the 16S rRNA gene. Presence of E. canis was confirmed by cleavage of the amplified DNA using endonucleases HaeIII and AvaI. Fourteen of 68 (21%) dogs with ticks had ehrlichiosis, whereas 12 of 61 (20%) dogs presented with thrombocytopenia and 4 of 19 (21%) anemic dogs had ehrlichiosis. Similar results were obtained in dogs with thrombocytopenia and anemia (one of eight positive) and in dogs with thrombocytopenia and ticks (two of seven positive). All four dogs with anemia and ticks, and the dog that fulfilled all inclusion criteria yield no amplification of ehrlichial DNA by PCR. Based on our results, one in each five dogs infested by the brown dog tick, with anemia or thrombocytopenia had ehrlichosis. Contrary to widespread believe, ehrlichiosis was not the main cause for thrombocytopenia in our region.  相似文献
