The higher-order assembly of chromatin imposes structural organization on the genetic information of eukaryotes and is thought to be largely determined by posttranslational modification of histone tails. Here, we study a 20-kilobase silent domain at the mating-type region of fission yeast as a model for heterochromatin formation. We find that, although histone H3 methylated at lysine 9 (H3 Lys9) directly recruits heterochromatin protein Swi6/HP1, the critical determinant for H3 Lys9 methylation to spread in cis and to be inherited through mitosis and meiosis is Swi6 itself. We demonstrate that a centromere-homologous repeat (cenH) present at the silent mating-type region is sufficient for heterochromatin formation at an ectopic site, and that its repressive capacity is mediated by components of the RNA interference (RNAi) machinery. Moreover, cenH and the RNAi machinery cooperate to nucleate heterochromatin assembly at the endogenous mat locus but are dispensable for its subsequent inheritance. This work defines sequential requirements for the initiation and propagation of regional heterochromatic domains.  相似文献

Climate change, as a result of increase in the concentration of greenhouse gases, influences growth and productivity of leguminous crops. A study was carried out to analyse the impacts of elevated carbon dioxide (CO2) and cyanobacterial inoculation on growth, N2 fixation and N availability and uptake in cowpea crop, under different doses of phosphorus. Cowpea crop was grown under ambient (400 µmol mol?1) and elevated (550 ± 20 µmol mol?1) CO2 levels using Free-Air Carbon dioxide Enrichment facility. Elevated CO2 level increased chlorophyll content in leaves, improved nodulation and nitrogen fixation by the crop. Increase in P dose up to 16 mg kg?1 soil enhanced nodule development and N2 fixation under high CO2 condition. Cyanobacterial inoculation increased nodule weight, leghaemoglobin content in nodules and total nitrogenase activity. Although nitrogen concentration in cowpea seeds decreased in high CO2 treatment, higher N uptake was recorded. Under elevated CO2 condition, cyanobacterial inoculation and higher P doses led to enhanced root growth and N2 fixation and availability of soil nitrogen. The study illustrated the synergistic effect of high CO2 and cyanobacterial inoculation in enhancing crop growth and availability of soil N, mediated by biological N2 fixation in cowpea under different levels of P.  相似文献

Responsive genotypes, timing and mode of nitrogen application are important for realizing potential yield of winter baby corn. Soil application of nitrogen is a common practice. Foliar application enhances absorption and utilization of nitrogen particularly after anthesis. We investigated combined approach in management of nitrogen for the first time including soil applications followed by foliar urea spray to enhance baby corn yield and profitability. To determine these, 2-year study conducted with three genotypes and six schedules of recommended dose of nitrogen (RDN). Growth characters, productivity traits, harvest period and yields recorded. Nitrogen content and uptake, protein content and harvest were determined. Genotype HM-4 produced 4.6% and 4.1% more cobs and corn weight over HQPM-1. Combined approach resulted higher yield attributes, yields, N uptake, protein harvest and monetary returns. RDN in 4 splits with more basal (B) dose increased cob and corn yield by 4.8% and 5.1% than 3 splits (50% B). Results suggest that HM-4 be grown using RDN 50% as B, 25% at knee height stage, 20% at tassel emergence followed by 5% foliar spray after first picking as urea solution (3%) for achieving higher yield and net returns. More studies needed under diverse conditions.  相似文献
