We studied seed morphometric characters, oil content and azadirachtin content in a medicinally important and insect repellent tree species Azadirachta indica A. Juss. Ten populations were selected from two central Indian states of Madhya Pradesh and Chhattisgarh and twenty candidate plus trees were selected from each population based on tree attributes. Azadirachtin content was estimated in seeds, in vitro shoots and callus cultures through HPLC. Significant variation (p < 0.05) was observed in seed morphometric characters, oil content and azadirachtin content among the ten populations. Maximum value for 100 seed weight was obtained in seeds of Raigarh (30.51 g) which was on par with seeds of Bargi (28.37 g) and Chhatarpur (26.36 g). The ratio of highest kernel to seed was also obtained in seeds of Raigarh (60.11%). Kernels from Chhatarpur population recorded the highest oil content (24.20%), which was equal to that in Sihore (23.75%) and Katni (21.75%). We recorded highly significant variation in azadirachtin content in seed samples, in vitro shoot samples and callus cultures. Maximum azadirachtin was present in the seeds of Gwalior (0.882%) which was on par with azadirachtin content in seeds of Chhatarpur, Katni, Sihore, Shahdol and Chhindwara. Maximum azadirachtin in the in vitro shoots was present in the cultures of Gwalior (0.218%) which was statistically on par with Sihore (0.176%). Similarly, in callus cultures of nodal segments maximum azadirachtin was recorded in the Gwalior population (0.033%) which was statistically on par with Sihore (0.028%). The trees of Gwalior and Sihore populations can be produced in large numbers through vegetative propagation to obtain seeds with higher azadirachtin content.  相似文献

Genetic erosion in cultivated wheat provides a good reason for studying genetic diversity in crop wild relatives. In the present study, genetic diversity of 32 accessions belonging to T. boeoticum species collected from different parts of Iran were evaluated using 13 morphological traits as well as ten inter-simple sequence repeat primers. Statistical analysis for morphological traits showed significant differences among accessions (except number of fertile tillers and total tillers per plant). In principal component analysis, the first three PCA showed 82.65 % of the total morphological variation. Based on the morphological traits, accessions were separated into two main groups by cluster analysis. In molecular analysis, polymerase chain reactions amplified 105 DNA fragments, out of which, 95 (90.47 %) were polymorphic. From geographic perspective, the accessions sampled from western and southwestern of Iran showed the highest and lowest polymorphism, respectively. However, the maximum values of effective number of alleles (Ne), Nei’s gene diversity (He) and Shannon’s information index (I) was related to accessions collected from NW regions. Also, according to cluster analysis and PCoA plot genetic diversity was not related to geographical distribution. Overall, our results revealed a remarkable level of genetic diversity among studied Iranian T. boeoticum accessions; especially accessions collected from Kermanshah and Lorestan provinces, which can be of interest for future breeding programs. So, conservation of germplasm of these areas is recommended.  相似文献

Accelerated soil erosion can impact upon agronomic productivity by reducing topsoil depth (TSD), decreasing plant available water capacity and creating nutrient imbalance in soil and within plant. Research information on soil‐specific cause – effect relationship is needed to develop management strategies for restoring productivity of eroded soils. Therefore, two field experiments were established on Alfisols in central Ohio to quantify erosion‐induced changes in soil properties and assess their effects on corn growth and yield. Experiment 1 involved studying the effects of past erosion on soil properties and corn yield on field runoff plots where soil was severely eroded and comparing it with that on adjacent slightly eroded soil. In addition, soil properties and corn grain yield in runoff plots were compared on side‐slopes with that on toe‐slopes or depositional sites. Experiment 2 involved relating corn growth and yield to topsoil depth on a sloping land. With recommended rates of fertilizer application, corn grain yield did not differ among erosional phases. Fertilizer application masked the adverse effects of erosion on corn yield. Corn grain yield on depositional sites was about 50 per cent more than that on side‐slope position. Corn plants on the side‐slope positions exhibited symptoms of nutrient deficiency, and the ear leaves contained significantly lower concentrations of P and Mg and higher concentrations of Mn and K than those grown on depositional sites. Corn grain yield in experiment 2 was positively correlated with the TSD. Soil in the depositional site contained significantly more sand and silt and less clay than that on the side‐slope position. There were also differences in soil properties among erosional phases. The soil organic carbon (SOC) content was 19\7 g kg−1 in slightly eroded compared with 15\1 g kg−1 in severely eroded sites. Aggregate stability and the mean weight diameter (MWD) were also significantly more in slightly eroded than severely eroded soils. Adverse effects of severe erosion on soil quality were related to reduction in soil water retention, and decrease in soil concentration of N and P, and increase in those of K, Ca and Mg. Severe erosion increased leaf nutrient contents of K, Mn and Fe and decreased those of Ca and Mg. Corn grain yield was positively correlated with aggregation, silt and soil N contents. Various clinical observations and experimental data from in vitro studies suggest that insulin and sex hormones interact. The main purpose of the present study was to examine the effects of testosterone and estradiol on serum insulin in rats. Seven week old male and female albino (Wistar) rats were used in our study. Testosterone enanthate (50 mg kg(-1) day(-1)) or estradiol valerate (200 microg kg(-1) day(-l)) were injected intraperiotenally or subcutaneously in orchidectomised or ovariectomised rats, respectively. In orchidectomised rats, serum insulin was decreased compared with control animals (p < 0.01), on the other hand, decreasing of serum insulin was prevented by testosterone replacement (p < 0.001). In ovariectomised rats, serum insulin was also decreased compared with control group (p < 0.01) and decreasing of serum insulin was prevented by estradiol replacement (p < 0.05). Conclusively, present findings indicated that testosterone or estradiol were serum insulin enhancer hormones in male or female rats, respectively.  相似文献

This research was carried out in Kashan Fajre Sepahan Galvanizing mills (KFSGM) for evaluation of water corrosivity during 2005-2006. A total of 18 samples were taken from various points of the water supply system for testing the specific parameters and calculation Langelier Index (LI), Ryznar Index (RI) and Pukorious Index (PI). This research showed that in raw water (sand filter effluent) LI were positive as well as RI and PI were lower than 7 which means that mentioned water is not corrosive. Also LI in treated water by reverse osmosis process was negative and RI and PI were higher than 7, so, this water has corrosive properties. Finally, calculated indexes indicate that according to LI, conditioned water is not corrosive but based on RI and P. this water tend to corrosivity which this findings is compatible with literature review statement. So it is recommended that, for water conditioning addition of preservative chemicals to be continued but at the same time another alternatives such as pH adjustment, air stripping and deoxygenating, control of carbonate concentration and split flow treatment should be studied.  相似文献

Safflower (Carthamus tinctorius L.) is mainly grown as an oil seed crop in the semiarid and temperate regions of the world. In this study, microsatellite markers were used to evaluate and characterize genetic relationships among 20 safflower genotypes including six Iranian and 14 exotic ones. Nine primer pairs generated clear scoring bands and yielded 22 alleles ranging from 100 to 400 bp with 2-4 alleles per locus. Polymorphic information content (PIC) values ranged from 0.11 to 0.62 with an average of 0.33. The Dice similarity coefficients among the genotypes ranged from 0.06 (between Soviet Union 2 and USA) to 0.95 (between Local Isfahan and Indian 2), indicating a relatively high genetic variation among the safflower genotypes studied. Both cluster and principal components analysis (PCA) clearly classified safflower genotypes into four groups. Results indicated that in most cases safflower genotypes were divided into the groups consistent with their country of origin. Clustering some foreign genotypes into the Iranian group was due to their similar genetic basis and it was suggested that Iran may be the origin of these. The findings suggested that microsatellite markers could be efficiently utilized to assess genetic diversity and relationships among safflower genotypes.  相似文献

Despite the success in fertilization and hatching of fish eggs with cryopreserved sperm, report on growth and survival of larvae produced from frozen‐thawed sperm is inadequate. The study evaluates the applicability of cryopreserved sperm for mass seed production by comparing the growth and survival of a popular food‐fish olive barb, Puntius sarana (Hamilton 1822) larvae produced from cryopreserved and fresh sperm. The eggs were artificially fertilized with cryopreserved and freshly collected sperm, and the growth and survival of produced larvae from both group recorded up to 12 weeks. The independent sample t‐test statistic showed the difference in lengths, t(718) = 0.241; P = 0.810 and weights, t(718) = 0.412; P = 0.680 were insignificant between two groups. There was also no significant difference, t(718) = ?0.758, P = 0.448 in survival of larvae produced from cryopreserved and freshly collected sperm. The study indicates that larvae of olive barb produced from cryopreserved sperm are equally compatible in growth and survival as the larvae produced from fresh sperm. Therefore, cryopreserved sperm can be applied for artificial fertilization of P. sarana to supply quality seed for aquaculture.  相似文献

Artificial hybridization was performed between Rutilus frisii kutum and Abramis brama orientalis of the Caspian Sea. Synchronization of spawning of female broodstock of both species was induced by injection of carp pituitary extract. Reciprocal crossings between R. frisii kutum ♀×A. brama orientalis ♂ (RA) and A. brama orientalis ♀×R. frisii kutum ♂ (AR) produced viable hybrid larvae without any clear particular pre‐ or postzygotic isolation phenomena. RA and AR hybrid larvae were reared to fingerling stage with survival rates of 22.5 and 28% and average weight of 6.8 ± 0.17 g and 9.0 ± 0.79 g, respectively. A heterosis of 45% was calculated for weight at fingerling stage. RA and AR hybrid fingerlings were cultured in polyculture along with Chinese carps for 6–7 mo and reached an average weight of 190–195 g and 235–255 g, respectively. Karyotyping of these hybrids revealed a modal diploid number of 2n = 50 for both groups, which is similar to those of the parental species. Discriminant function analysis on 28 morphometric and meristic characteristics of two parental species as well as their hybrids could separate these groups at highly significant level (P < 0.001). These results indicated an overall intermediate inheritance of the studied characters.  相似文献

Gas exchange was measured in potatoes (cv. Folva) grown in lysimeters (4.32 m2) in coarse sand, loamy sand, and sandy loam and subjected to full (FI), deficit (DI), and partial root-zone drying (PRD) irrigation strategies. PRD and DI as water-saving irrigation treatments received 65% of FI and started after tuber bulking and lasted for six weeks until final harvest. Midday photosynthesis rate (An) and stomatal conductance (gs) of fully irrigated (FI) plants were lowest in coarse sand and mean An of diurnal measurements in FI, PRD and DI tended to be lower in this soil as compared with the loamy sand and sandy loam. The results revealed that diurnal values of An and gs in PRD and DI were consistently lower than FI without reaching significant differences in accordance with findings that xylem [ABA] in PRD was significantly higher than FI, and tended to be higher than in DI. Diurnal measurements showed that An reached peak values during mid-morning and midday, while gs were highest during the morning. Intrinsic water use efficiency (An/gs) correlated linearly well with the leaf to air vapor pressure deficit (VPD) and the slope of the line revealed the rate of An/gs increase per each kPa increase in VPD, i.e. approximately 10 μmol mol−1. Transpiration efficiency (An/T) of PRD was higher than DI, which shows slightly better efficient water use than DI. The slope of the linear relationship between transpiration efficiency and VPD decreased from −2.03 to −1.04 during the time course of the growing season, indicating the negative effect of leaf ageing on photosynthesis and thus on plant water use efficiency. This fact shows the possibility to save water during last growth stages through applying water-saving irrigations without much effect on transpiration efficiency.  相似文献

The objective of this study was to determine the effect of the presence of recombinant ovine growth hormone either alone or together with follicle stimulating hormone (FSH) during ovine oocyte in vitro maturation (IVM) on nuclear maturation and subsequent embryo development. Moreover, the effect of growth hormine (GH) on embryo development whether influenced by the presence of foetal bovine serum (FBS) was assessed. The abattoir‐derived oocytes were randomly divided into four treatment groups and cultured in maturation medium supplemented with: (i) 0.05 IU/ml FSH; (ii) 300 ng/ml roGH; (iii) FSH + roGH; and (iv) no FSH and GH (control). The percentages of germinal vesicle‐stage oocytes in GH‐treated group after 8 h of culture was significantly higher than the FSH and FSH + GH groups and lower than control (22.4%, 8.7%, 9.1%, and 32% respectively). The percentage of MII‐stage oocytes was significantly increased in the presence of GH after 16 and 24 h of culture compared to the control (44.7% and 83.1% vs 32.6% and 73.6% respectively). There was no significant synergism between GH and FSH in terms of nuclear maturation. The blastocyst rates in serum‐supplemented groups were enhanced by the presence of FSH and GH compared to the control (35.4% and 31.3 vs 11.4% respectively). Compared with either GH or FSH alone, the subsequent embryo development (blastocyst rate), however, was negatively influenced by co‐presence of both hormones (22.8%). In contrast, the corresponding values were not affected in the absence of serum. In conclusion, GH had positive effect on nuclear maturation of sheep oocytes. Moreover, the pattern of the effect of GH on embryo development was influenced by the presence of FBS during IVM.  相似文献
