Rice genetic diversity partitioning between farms, varieties and, within-variety diversity, were analysed in two villages of Maritime Guinea with contrasted agroecological conditions. One thousand and two hundred individual plants belonging to 45 accessions collected in eight farms were genotyped using 10 SSR markers. The molecular variance was evenly shared between and within accessions, while the farm effect was almost nil. Local varieties had a multi-line genetic structure. The number of multilocus genotypes was proportional to the utilisation rate of the variety in the village. The F ST values between different accessions of each variety were significant which indicated low genetic consistency in the variety names. This varietal structure could mainly be explained by the migration phenomenon and the high varietal turnover. Compared to allelic diversity, multilocus genotypic diversity seemed to be the most suitable indicator of the quantitative distribution of diversity at different management scales (accession, farm and village). The within- and between-farm F ST values were in the same order of magnitude. The within-farm diversity was not farm-specific but quantitatively high, i.e. up to 50% of the total genotypic diversity of a given village. Given the relative importance of the within-variety diversity, the in situ approach stands out as the most effective solution. As farms do not host specific diversity the in situ approach could be implemented by working with a small number of farms.  相似文献

Ex situ collections offer the potential to reduce extinction risks, affording option to society in maintaining future breeding opportunities for productivity and heritage traits. However, how much should we be seeking to collect and conserve in gene banks, and where? We developed a mathematical model to optimize logistical decisions of breed conservation choices and to evaluate alternative scenarios for efficiently re‐allocating genetic materials currently stored in different European gene banks, allowing for cross‐country collections, cost and cryogenic capacity differentials. We show how alternative allocations for the breeds that are currently stored in 11 European gene banks could reduce overall conservation costs by around 20% by selecting cryogenic banks that have relatively lower combination of fixed and collection costs, and are geographically closer to collection regions. Our results show that centralizing collections in one gene bank would double the costs, relative to collective European collections approaches. We also calculate marginal costs of collections and show that increasing diversity within the gene banks implies in higher costs per conserved breed.  相似文献

Yield and water productivity of potatoes grown in 4.32 m2 lysimeters were measured in coarse sand, loamy sand, and sandy loam and imposed to full (FI), deficit (DI), and partial root-zone drying (PRD) irrigation strategies. PRD and DI as water-saving irrigation treatments received 65% of FI after tuber bulking and lasted for 6 weeks until final harvest. Analysis across the soil textures showed that fresh yields were not significant between the irrigation treatments. However, the same analysis across the irrigation treatments revealed that the effect of soil texture was significant on the fresh yield and loamy sand produced significantly higher fresh yield than the other two soils, probably because of higher leaf area index, higher photosynthesis rates, and “stay-green” effect late in the growing season. More analysis showed that there was a significant interaction between the irrigation treatments and soil textures that the highest fresh yield was obtained under FI in loamy sand. Furthermore, analysis across the soil textures showed that water productivities, WP (kg ha−1 fresh tuber yield mm−1 ET) were not significantly different between the irrigation treatments. However, across the irrigation treatments, the soil textures were significantly different. This showed that the interaction between irrigation treatments and soil textures was significant that the highest significant WP was obtained under DI in sandy loam. While PRD and DI treatments increased WP by, respectively, 11 and 5% in coarse sand and 28 and 36% in sandy loam relative to FI, they decreased WP in loamy sand by 15 and 13%. The reduced WP in loamy sand was due to nearly 28% fresh tuber yield loss in PRD and DI relative to FI even though ET was reduced by 9 and 11% in these irrigation treatments. This study showed that different soils will affect water-saving irrigation strategies that are worth knowing for suitable agricultural water management. So, under non-limited water resources conditions, loamy sand produces the highest yield under full irrigation but water-saving irrigations (PRD and DI) are not recommended due to considerable loss (28%) in yield. However, under restricted water resources, it is recommended to apply water-saving irrigations in sandy loam and coarse sand to achieve the highest water productivity.  相似文献

We studied seed morphometric characters, oil content and azadirachtin content in a medicinally important and insect repellent tree species Azadirachta indica A.Juss. Ten populations were selected from two central Indian states of Madhya Pradesh and Chhattisgarh and twenty candidate plus trees were selected from each population based on tree attributes. Azadirachtin content was estimated in seeds, in vitro shoots and callus cultures through HPLC. Significant variation(p \ 0.05) was observed in seed morphometric characters, oil content and azadirachtin content among the ten populations. Maximum value for 100 seed weight was obtained in seeds of Raigarh(30.51 g) which was on par with seeds of Bargi(28.37 g)and Chhatarpur(26.36 g). The ratio of highest kernel to seed was also obtained in seeds of Raigarh(60.11%).Kernels from Chhatarpur population recorded the highest oil content(24.20%), which was equal to that in Sihore(23.75%) and Katni(21.75%). We recorded highly significant variation in azadirachtin content in seed samples,in vitro shoot samples and callus cultures. Maximum azadirachtin was present in the seeds of Gwalior(0.882%)which was on par with azadirachtin content in seeds of Chhatarpur, Katni, Sihore, Shahdol and Chhindwara.Maximum azadirachtin in the in vitro shoots was present in the cultures of Gwalior(0.218%) which was statistically on par with Sihore(0.176%). Similarly, in callus cultures of nodal segments maximum azadirachtin was recorded in the Gwalior population(0.033%) which was statistically on par with Sihore(0.028%). The trees of Gwalior and Sihore populations can be produced in large numbers through vegetative propagation to obtain seeds with higher azadirachtin content.  相似文献

Mammalian expression systems, due to their capacity in post-translational modification, are preferred systems for biopharmaceutical protein production. Several recombinant protein systems have been introduced to the market, most of which are under clinical development. In spite of significant improvements such as cell line engineering, introducing novel expression methods, gene silencing and process development, expression level is unpredictable and unstable because of the random location of integration in the genome. Site-specific recombination techniques are capable of producing stable and high producer clonal cells; therefore, they are gaining more importance in the biopharmaceutical production. Site-specific recombination methods increase the recombinant protein production by specifically inserting a vector at a locus with specific expression trait. The present review focused on the latest developments in site-specific recombination techniques, their specific features and comparisons.  相似文献

In this research, results of an experimental interaction effect of operating parameters on tensile strength carbon fibers from a commercial PAN-based precursor are investigated. Ten parameters at two and four levels (L32=21×49) were investigated: stabilization temperature at first stage (STFIS), stabilization duration time at first stage (SDTFIS), stabilization temperature at second stage (STSS), stabilization duration time at second stage (SDTSS), stabilization temperature at third stage (STTS), stabilization duration time at third stage (SDTTS), stabilization temperature at fourth stage (STFOS), stabilization duration time at fourth stage (SDTFOS), carbonization temperature (CT), and carbonization duration time (CDT). In this study, Taguchi method was used initially to plan a minimum number of experiments. Statistical analysis, analysis of variance (ANOVA), was also employed to determine the relationship between experimental conditions and yield levels. ANOVA was applied to calculate sum of square, variance, ratio of factor variance to error variance and contribution percentage of each factor on response. The results show that increasing all of parameters improves tensile strength performance. The optimum levels of influential factors, determined for tensile strength are STFIS 200 °C, SDTFIS 120 min, STSS 225 °C, SDTSS 120 min, STTS 240 °C, SDTTS 120 min, STFOS 260 °C, SDTFOS 60 min, CT 1400 °C and CDT 10 min. The results showed that CT and ODTFIS are the most and the less effective factors on response, respectively.  相似文献
