Two types of transglutaminases (TGases), Ca(2+)-dependent TGase derived from guinea pig liver (GTGase) and Ca(2+)-independent TGase derived from a variant of Streptoverticillium mobaraense (MTGase), were used to study the cross-linking of soybean 11S globulin (glycinin). The effects of sulfhydryl reductant (dithiothreitol, DTT) and Ca(2+) on the conformation and TGase-catalyzed polymerization of glycinin were investigated. The conformational change of glycinin was probed by spectral methods. The degree of cross-linking and the polymer (aggregate) formation were analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and dynamic light scattering, respectively. Addition of DTT stimulated the TGase-catalyzed cross-linking reactions without destroying the secondary and tertiary structure of glycinin but did not influence the polymer or aggregate formation. It was found that Ca(2+) caused the formation of larger size polymers at lower concentrations, while it suppressed the polymerization at higher concentrations. In addition, the cross-linking behaviors of glycinin were shown to be different between MTGase- and GTGase-catalyzed systems. 相似文献

Many surface soils in Japan may experience more frequent and intense drying–rewetting (DRW) events due to future climate changes. Such DRW events negatively and positively affect microbial biomass carbon (MBC) through microbial stress and substrate supply mechanisms, respectively. To assess the MBC immediately after DRW and during the incubation with repeated DRW cycles, two laboratory experiments were conducted for a paddy soil. In the first experiment, we exposed the soil to different drying treatments and examined the MBC and hourly respiration rates immediately after the rewetting to evaluate the microbial stress. In the second experiment, we compared microbial growth rates during the incubation of the partially sterilized soil with a continuously moist condition and repeated DRW cycles to evaluate the contribution of the substrate supply from non-biomass soil organic C on MBC. First, all drying treatments caused a reduction in MBC immediately after the rewetting, and higher drying intensities induced higher reduction rates in MBC. A reduction of more than 20% in MBC induced the C-saturated conditions for surviving microbes because sufficient concentrations of labile substrate C were released from the dead MBC. Second, repeated DRW cycles caused increases in the microbial growth rates because substrate C was supplied from non-biomass organic C. In conclusion, MBC decreased immediately after DRW due to microbial stress, whereas MBC increased during repeated DRW cycles due to substrate C supplied from non-biomass organic C. 相似文献

Thelephora aurantiotincta is an edible mushroom belonging to the genus Thelephora; it grows in symbiosis with pine trees. Recently, phytochemical investigations have revealed that the genus Thelephora is an abundant source of p-terphenyl derivatives. However, their bioactivity has not yet been well characterized. In screening for natural materials with anticancer activity, a T. aurantiotincta ethanol extract (TAE) was found to decrease cell viability in human hepatocellular carcinoma cells (HepG2). In this study, a new p-terphenyl derivative, thelephantin O, and a known compound, vialinin A, were isolated as the principal bioactive components of TAE. These compounds decreased cell viability in HepG2 and human colonic carcinoma cells (Caco2), but not in noncancerous human hepatocytes. This is the first report of the isolation from T. aurantiotincta of selective cytotoxic agents against cancer cells. 相似文献

A specialized measurement system for measuring suspended sediment concentration in model experiments was developed; it utilizes general-purpose optical sensors to perform measurements. A photoelectric sensor was used for measuring the transmitted light, and a fiber optic sensor was used for measuring the backscattered light. The values of the light absorbance and intensity of backscattered light for turbid standard solutions measured with these sensors show a linear relationship with the turbidity. The calibration lines for sand particles with a large settling velocity were established by using a calibration tank containing upwelling water. The vertical profiles of the sediment concentration measured with the photoelectric sensor corresponded well to those of the sediment concentration measured with a water sampler in an artificial channel. This result suggests that the measurement system using a photoelectric sensor functions satisfactorily. The sediment concentrations measured using the fiber optic sensor correlate well with the concentrations measured through siphon sampling; however, the values measured with the fiber optic sensor are approximately 25% larger than those measured with the siphon. This disagreement may result from the differences in the optical conditions between the calibration tank and the actual channel. Thus, it may be possible to utilize fiber optic sensors calibrated on the basis of siphon sampling for the measurement of sediment concentration. 相似文献

Abstract Recently agricultural activity in the mountainous area of northern Thailand has increased and problems relating to soil fertility have arisen. In order to gain basic information about the soil properties associated with shifting cultivation, physicochemical properties of the surface soils (0–10 cm) and subsoils (30–40 cm) were investigated in selected villages in the area. The physicochemical properties of the soils studied are summarized as follows: 1) The soils were rich in organic matter, content of which ranged from 11.4 to 63.3 g C kg?1 in the surface soil. 2) The pH(H2O) of the soils mostly ranged from 5 to 7 and soil acidity was more pronounced in the deeper horizons. In the surface soils, exchangeable Ca and Mg were generally dominant, whereas exchangeable Al was often predominant in the subsoils. 3) Most of the soils showed a medium to fine texture with more than 30% clay. The clay mineral composition was characterized by various degrees of mixture of kaolin minerals and clay mica with, in some cases, a certain amount of 2:1-2:1:1 intergrades. 4) According to the ion adsorption curves, most of the B horizon soils were characterized by the predominance of permanent negative charges. On the other hand, organic matter contributed to the increase of variable negative charges in the surface soils. The content of organic matter and the percentage of the clay fraction were essential for determining the CEC of the soils of the surface 10 and 30–40 cm depths, respectively. Under the field conditions, the composition of exchangeable cations largely reflected the soil acidity. In addition, the content of organic matter also showed a significant correlation with that of available N in the surface soils. Thus, soil acidity both in the surface soils and subsoils, organic matter content in the surface soils, and clay content in the subsoils were considered to be the main factors that affected soil chemical fertility in the area. 相似文献

The pathological effects of copper deficiency (COD) are well known. However, the pathogenesis of cardiomyopathy resulting from COD remains unclear. In this study, aimed to elucidate the pathogenesis of COD-induced cardiomyopathy by examining the morphology of the cardiovascular system in copper-deficient rats using histopathology, immunohistochemistry, and scanning and transmission electron microscopy. Changes detected in the myocardium and interstitium were consistent with those reported for COD. Morphological changes included mesh-like changes in the capillary endothelial cells that appear to be a novel finding in COD-induced cardiomyopathy. These changes are hypothesized to result from abnormal vascular remodeling following damage to the basement membrane and due to the mechanical effects of myocardial contractions. Although cardiomyopathy may be associated with microcirculatory disorders arising from these lesions, further investigations are necessary to demonstrate a causal relationship between the pathogenesis of cardiomyopathy and the contribution of these lesions to disease progression. 相似文献
