Genetic analysis of resistance of plant introduction (PI) 438489B to soybean cyst nematode (SCN) have shown that this PI is
highly resistant to many SCN HG types. However, validation of the previously detected quantitative trait loci (QTL) has not
been done. In this study, 250 F2:3 progeny of a Magellan (susceptible) × PI 438489B (resistant) cross were used for primary genetic mapping to detect putative
QTL for resistance to five SCN HG types. QTL confirmation study was subsequently conducted using F6:7 recombinant inbred lines (RILs) derived from the same cross. Simple sequence repeat (SSR) and single nucleotide polymorphism
(SNP) markers were employed for molecular genotyping. Interval mapping (IM), permutation tests, cofactor selection, and composite
interval mapping (CIM) were performed to identify and map QTL. Results showed that five QTL intervals were associated with
resistance to either multiple- or single-HG types of SCN. Among these, two major QTL for resistance to multiple-SCN HG types
were mapped to chromosomes (Chr.) 8 and 18, consistent with the known rhg1 and Rhg4 locations. The other QTL were mapped to Chr. 4. The results of our study confirmed earlier reported SCN resistance QTL in
this PI. Moreover, SSR and SNP molecular markers tightly linked to these QTL can be useful for the near-isogenic lines (NILs)

Effect of supplementation frequency and supplemental urea level on forage use (Exp. 1) and performance (Exp. 2 and 3) of beef cattle consuming low-quality tallgrass-prairie were evaluated. For Exp. 1 and 2, a 2 x 2 factorial treatment structure was used, such that two supplements (30% CP) containing 0 or 30% of supplemental degradable intake protein (DIP) from urea were fed daily or on alternate days. In Exp. 1 and 2, supplement was fed at 0.41% BW daily or at 0.83% BW (DM basis) on alternate days. For Exp. 3, a 2 x 4 factorial treatment structure was used, such that four supplements (40% CP) containing 0, 15, 30, or 45% of supplemental DIP from urea were fed daily or 3 d/wk. Supplements were group-fed at 0.32% BW daily or at 0.73% BW (DM basis) 3 d/wk. In Exp. 1, 16 Angus x Hereford steers (initial BW = 252 kg) were blocked by BW and assigned to treatment. Urea level x supplementation frequency interactions were not evident for forage intake, digestion, or rate of passage. Forage OM intake (OMI) and total digestible OMI (TDOMI) were not significantly affected by treatment. Total-tract digestion of OM (P = 0.03) and NDF (P = 0.06) were greater for steers supplemented daily. In Exp. 2, 48 Angus x Hereford cows (initial BW = 490 kg) grazing winter tallgrass prairie were used. Significant frequency x urea interactions were not evident for BW and body condition (BC) change; similarly, the main effects were not substantive for these variables. In Exp. 3, 160 Angus x Hereford cows (initial BW = 525 kg) grazing dormant, tallgrass prairie were used. Supplement refusal occurred for cows fed the highest urea levels, particularly for cows fed the supplement with 45% of the DIP from urea 3 d/wk, and supplement refusal increased closer to calving. A frequency x urea interaction (P = 0.02) was observed for prepartum BW changes. As supplemental urea level increased, prepartum BW loss increased quadratically (P = 0.02); however, a greater magnitude of loss occurred when feeding supplements containing > or = 30% of DIP from urea 3 d/ wk. Cumulative BC change followed a similar trend. In conclusion, moderate protein (< or = 30% CP) supplements with < or = 30% of supplemental DIP from urea can be fed on alternate days without a substantive performance penalty. However, infrequent feeding of higher protein (> 30% CP) supplements with significant urea levels (> 15% of DIP from urea) may result in decreased performance compared with lower urea levels. 相似文献

REASONS FOR PERFORMING STUDY: The mediators and signalling cascades important in the initiation of laminitis remain unclear. We therefore wanted to explore the genes and overall signalling mechanisms that play an important role in the developmental stage of laminitis. OBJECTIVE: To use a broad genomic screening technique to identify novel genes that are differentially regulated in the equine lamellae during the developmental period of laminitis. METHODS: Differential mRNA display (DRD) was performed to discover regulated genes, and real-time quantitative polymerase chain reaction (RT-qPCR) was then used to evaluate lamellar mRNA levels of a regulated gene (MAIL) and mediators related to that gene (IL-1beta and IL-6) in control horses (n = 5) and horses administered black walnut extract (BWE; n = 5). RESULTS: Using DRD, MAIL was identified as a regulated gene. RT-qPCR indicated a 4-fold increase in expression of the MAIL mRNA in BWE lamellae compared to controls. A 30-fold increase in IL-1beta, and a 160-fold difference in IL-6 mRNA expression was present in BWE lamellae. Differences in MAIL, IL-1beta and IL-6 mRNA expression were statistically significant between groups (P < 0.05). CONCLUSIONS AND POTENTIAL RELEVANCE: The data strongly support a role for inflammatory cytokines in the developmental stages of laminitis, possibly inducing the vascular and metabolic alterations reported to occur in the affected digit. These results potentially support the use of anti-inflammatory drugs in horses at risk of laminitis, and warrant further investigation of the link between systemic disease processes associated with laminitis and the reported digital inflammation. 相似文献

Ruminally fistulated steers (n = 13; 263 kg) were used in an incomplete Latin square with 13 treatments and four periods to evaluate the effects of level and source of supplemental carbohydrate (CHO) and level of degradable intake protein (DIP) on the utilization of low-quality, tallgrass-prairie hay. Steers were given ad libitum access to forage (5.7% CP, 2.6% DIP, and 74.9% NDF). The supplementation treatments were fashioned as a 2x3x2 factorial arrangement plus a negative control (NC; no supplement). The factors included two DIP levels (.031 and .122% BW) and three CHO sources (starch, glucose, and fiber) fed at two levels (.15 and .30% BW) within each level of DIP supplementation. The effect of supplementation on forage OM intake (FOMI) was dependent (P<.01) on level and source of CHO and level of DIP fed. When DIP was low, forage, total, and digestible OM intakes were generally greater for the starch treatment than for the nonstarch treatments. However, when the DIP level was high, intakes were greater for the nonstarch (i.e., fiber and glucose) treatments. Generally, FOMI decreased (P<.01) when more supplemental CHO was provided. Supplementation typically increased fiber digestion, but the response was dependent (P<.01) on level and source of CHO and level of DIP. Generally, supplements with low levels of CHO improved NDF digestion (NDFD). However, supplements with the high level of CHO decreased NDFD, except for fiber at the high level of DIP. Organic matter digestion was increased by supplementation, but the impact of increasing CHO was dependent (P<.01) on source of CHO and level of DIP. Supplementation treatments had significant impact on ruminal pH, NH3 N, and the total concentration of organic acids as well as their relative proportions. In conclusion, supplemental DIP enhanced the use of low-quality forage; however, the impact of supplemental CHO on low-quality forage use was dependent on source and level of CHO offered, as well as the level of DIP provided. 相似文献

Inaccurate or incomplete diagnosis of the root causes of overfishing can lead to misguided and ineffective fisheries policies and programmes. The “Malthusian overfishing narrative” suggests that overfishing is driven by too many fishers chasing too few fish and that fishing effort grows proportionately to human population growth, requiring policy interventions that reduce fisher access, the number of fishers, or the human population. By neglecting other drivers of overfishing that may be more directly related to fishing pressure and provide more tangible policy levers for achieving fisheries sustainability, Malthusian overfishing relegates blame to regions of the world with high population growth rates, while consumers, corporations and political systems responsible for these other mediating drivers remain unexamined. While social–ecological systems literature has provided alternatives to the Malthusian paradigm, its focus on institutions and organized social units often fails to address fundamental issues of power and politics that have inhibited the design and implementation of effective fisheries policy. Here, we apply a political ecology lens to unpack Malthusian overfishing and, relying upon insights derived from the social sciences, reconstruct the narrative incorporating four exemplar mediating drivers: technology and innovation, resource demand and distribution, marginalization and equity, and governance and management. We argue that a more nuanced understanding of such factors will lead to effective and equitable fisheries policies and programmes, by identifying a suite of policy levers designed to address the root causes of overfishing in diverse contexts. 相似文献

A nondestructive protocol for maize kernel starch sampling was developed, enabling starch preparation from a single kernel for analysis of starch structure while also maintaining the vitality of the seed. To develop the single kernel sampling (SKS) method, maize genotypes varying in starch structure including ae, wx, su2, du and normal in the W64A inbred line were used. Crude endosperm material was removed from the kernel crown, soaked, ground, washed, and dissolved in 90% DMSO. The sample represented ≈10% of the total kernel. Endosperm starch was also isolated from the same genotypes by a standard multikernel isolation (MKI) method. Starches isolated by the two methods were debranched and analyzed by high‐performance size‐exclusion chromatography (HPSEC) and fluorophore‐assisted carbohydrate electrophoresis (FACE). HPSEC and FACE showed similar results for the two sampling methods for degree of polymerization (DP) ≤ 50. We concluded that the material obtained by SKS could be used for identifying amylopectin structural differences among genotypes. Kernel sampling for SKS had no effect on germination, thus plants could be grown for subsequent genetic crosses and analysis. The SKS method may be useful for the screening of populations of maize kernels from genotypes producing novel amylopectin structure, and allow the growth of novel genotypes for further analysis. 相似文献

Development of efficient cost‐effective diets is a critical component in the refinement of production technologies for the largemouth bass, Micropterus salmoides (LMB). One of the first steps in reducing feed costs can be to decrease the amount of fish meal in the diet. The objective of this study was to evaluate reduced levels of fish meal, and a least‐cost formulation diet, for second year growout of LMB under practical pond conditions. Twelve 0.04‐ha ponds were stocked with juvenile LMB (210.1 ± 3.3 g) at a stocking density of 8650 fish/ha (350 fish/pond). Each pond was randomly assigned one of the four dietary treatments with three replicate ponds per treatment. The three experimental diets contained varying levels of fish meal. Diets FM‐45, FM‐24, and FM‐8 contained 45, 23.5, and 8% fish meal, respectively. In diets FM‐24 and FM‐8, fish meal was replaced by varying levels of poultry by‐product meal, soybean meal, and blood meal. The fourth diet was a commercial salmonid diet widely used as a LMB growout feed (Nelson and Sons, Inc., Silvercup TM , Steelhead, Murray, UT, USA). This diet served as a commercial control (CC) and contained 46% crude protein. The experimental diets were formulated to be isonitrogenous and isocaloric with the CC diet and were fed once daily to apparent satiation for 180 d. At harvest, there were no significant differences between treatments ( P > 0.05 ) in terms of survival, which averaged 95% overall. Mean weights of fish fed the three experimental diets FM‐45, FM‐24 and FM‐8 were not significantly different ( P > 0.05 ) and averaged 518, 546, and 529 g, respectively, but were all significantly greater ( P≤ 0.05 ) than those fed the CC (488 g). Feed conversion ratio (FCR) of fish fed the FM‐45 and FM‐8 diets (1.43 and 1.46, respectively) was significantly greater ( P≤ 0.05 ) than those fed the FM‐24 diet (1.34). The FCR of fish fed the CC diet (1.39) was not significantly different ( P > 0.05 ) from fish fed other diets. Feed cost per unit of weight gain ($US/kg) was significantly lower ( P≤ 0.05 ) in fish fed the FM‐24 and FM‐8 diets ($0.73 and $0.72/kg, respectively) than in fish fed other diets. Feed cost per unit gain of fish fed the FM‐45 diet ($0.83/kg) was significantly lower ( P≤ 0.05 ) than those fed the CC diet ($1.04/kg). There were no significant differences ( P > 0.05 ) in dress‐out percentages or proximate composition among fish fed the four diets. This study indicates that fish meal levels in feeds used for the second year growout of LMB can be reduced to ≥ 8% of the formulation without reducing survival or growth and without negatively impacting body composition. 相似文献

Organic acid analysis plays a fundamental role in the testing of authenticity of fruit juices. Analytical methods used routinely for organic acids suffer from poor reproducibility, often give false positives/negatives for tartaric acid, and do not offer the possibility of analyte confirmation. There are conflicting reports in the literature on the presence/absence of tartaric acid in pomegranate juice, a potential indicator of adulteration with grape juice. In this work, a method based on stable isotope dilution liquid chromatography-tandem mass spectrometry is described for citric, malic, quinic, and tartaric acid in fruit juices. Validation data including precision and recovery in six types of juice are presented. Tartaric and quinic acids were confirmed in pomegranate juice at concentrations of 1-5 and ~1 mg/L, respectively. These concentrations are much lower than those resulting from adulteration with grape juice and apple juice, respectively, at the 5% level. A separate method for isocitric acid in orange juice based on the single standard addition method is also described. 相似文献

OBJECTIVE: To compare the validity of a modified Block food-frequency questionnaire (FFQ), a picture-sort administration of the FFQ (PSFFQ) and a meal pattern-based questionnaire (MPQ) in a multi-ethnic population of low socio-economic status (SES). DESIGN: Participants completed six 24-hour dietary recalls (24HR) over six months; the FFQ, PSFFQ and MPQ were completed in random order in the subsequent month. Instruments were interviewer-administered. The PSFFQ and MPQ were developed in formative research concerning difficulties for older adults in responding to standard food-frequency instruments. SETTING: Rural North Carolina, USA.Subjects One hundred and twenty-two African American, Native American and white adults aged > or = 65 years, with approximately one-third in each ethnic group. Inclusion criteria included education < or = 12 years and income < or = 150% of national poverty level or Medicaid recipient. RESULTS: Comparing median intakes from the average of the 24HR with the three diet assessment instruments, the MPQ tended to overestimate intakes compared with the FFQ and PSFFQ. Correlations among nutrients obtained by the 24HR and the other three instruments were generally statistically significant and positive. Across nutrients, the PSFFQ was most highly correlated with the 24HR for women, while the FFQ was most highly correlated with the 24HR for men. CONCLUSIONS: Dietary assessments using 24HR and FFQ were similar to results reported elsewhere, although correlations between 24HR and FFQ were somewhat lower. Interviewer-administered dietary assessments should be used with caution to evaluate dietary intake among older adults with low SES. Gender differences and the lower correlations should be investigated more thoroughly to assist in choosing dietary assessment instruments for this population. 相似文献
