The progressive morphohistopathologic changes, distribution pattern of lesions and ultrastructural characteristics in Eimeria arloingi infection were investigated in experimentally infected kids. The 18 newborn animals allocated to 3 equal groups. Two of groups,
A, B were inoculated with a single dose of 1 × 103 and1 × 105 sporulated oocysts of E. arloingi, respectively. At 7, 14, 21, 28, 35, and 42 days postinoculation (DPI), 1 kid from each group was necropsied for pathologic
and ultrastructural studies. Progressive lesions were present at 21, 28, 35 and 42 DPI in the jejunum, ileum, cecum with fewer
in the duodenum and proximal colon. The oocysts shedding begin between 16 to 18 DPI. Grossly, minimal changes were observed
at 21 DPI as few whitish plaques or nodules and advanced lesions at 42 DPI as pseudoadenomatous pattern in the mucosa and
a cerebriform pattern on the serosal surface of jejunum and ileum. Early histopathologic lesions due to schizogony phase were
including presence of intracytoplasmic developmental stages of the parasite such as trophozoites, immature to mature schizonts
and mild infiltration of inflammatory cells. In late lesions due to various stages of gametogony, the histological pattern
was mainly remarkable hyperplasia of the villi and crypts epithelial cells, eventually developed into papillary projections
of reactive epithelium. The mesenteric lymph nodes showed a few numbers of large schizonts in the cortical lacteals. This
study showed E. arloingi as a highly pathogenic species for kids, the incubation period was 16–18 days and the main target organ was jejunum with

The present study evaluates the effects of embryonic age and proteolytic enzymes on the isolation and primary culture of chicken enterocyte and to establish an effective technique for chicken intestinal epithelial cell (IEC) cultivation. Fourteen‐day‐old, 16‐day‐old and 18‐day‐old embryos (average weight: 52.23 ± 0.76 g, 50.86 ± 0.99 g, 48.98 ± 1.03 g) were the source for preparation of enterocyte culture, and trypsin‐ethylene diamine tetraacetic acid, collagenase, thermolysin and combination of collagenase and thermolysin were used for digestion medium. Optimal culture protocols were determined by qualitative assays of proliferation. Cells isolated by using 14‐day‐old embryo and collagenase obtain the best attachment and growth in culture, and the production of continuously growing IEC cultures. Thus, we conclude that the use of collagenase as a dissociating enzyme and 14‐day‐old embryo as a source can be advantageously applied to the isolation of chicken IEC and this method may be useful for various applications and basic studies of the intestinal tract concerning such objects as physiology, immunology and toxicology. 相似文献

Background:Immobilization is an approach in industry to improve stability and reusability of urease. The efficiency of this technique depends on the type of membrane and the method of stabilization. Methods:The PEI-modified egg shell membrane was used to immobilize urease by absorption and glutaraldehyde cross-linking methods. The membranes were characterized by FTIR and AFM, and Nessler method was applied to measure the kinetic of the immobilized enzymes. Finally, the storage stability (6 °C for 21 days) and reusability (until enzyme activity reached to zero) of the immobilized enzymes were investigated. Results:Based on FTIR, three new peaks were observed in both the absorption- (at 1389.7, 1230.8, and 1074.2 cm-1) and the cross-linking (at 1615-1690, 1392.7, 1450 cm-1) immobilized enzymes. The surface roughness of the native membrane was altered after PEI treatment and enzyme immobilization. The optimal pH of cross-linking immobilized enzymes was shifted to a more neutral pH, while it was alkaline in adsorption-immobilized and free enzymes. The reaction time decreased in all immobilized enzymes (100 min for free enzyme vs. 60 and 30 min after immobilizing by adsorption and cross-linking methods, respectively). The optimal temperature for all enzymes was 70 °C and they had a higher Km and a lower Vmax than free enzyme. The stability and reusability of urease were improved by both methods. Conclusion:Our findings propose these approaches as promising ways to enhance the urease efficiency for its applications in industries and medicines. Pomegranate fruit is an important source of potentially healthy bioactive compounds and mineral nutrients. Changes in total phenolic compound, concentrations, and levels of macronutrients (P, K, N, Mg, Ca and Na) and micronutrients (Zn, Cu, Mn, Fe and B) in arils and peel of pomegranate fruit were recorded from 10 days after full bloom until harvest. Total phenolics levels increased at early stage of growth both in peel and arils of fruit, but thereafter generally decreased during maturation and reached to 3.70 and 50.22 mg g−1 of dry weight in arils and peel, respectively, at harvest. The amount of total phenolics in peel was markedly higher than arils of pomegranate fruit. The concentration of most elements in arils and peel decreased during fruit growth and development. At harvest the relative order of concentration of macronutrients both in arils and peel was K > N > Ca > P > Mg > Na. The concentration of most micronutrients was greater in the arils than in the peel especially in early season. The relative order of concentration of micronutrients in arils was B > Fe > Zn > Cu > Mn. The accumulation of all the macro- and microelement within the fruit also increased during fruit growth and development. These results provide important data on total phenolics and macro- and micronutrient changes during fruit growth and development, emphasizing that pomegranate fruit can be a good source of bioactive compounds and minerals. 相似文献

Most temperate terrestrial orchids are endangered species. Attempts to produce plantlets from plants of the genus Epipactis by asexual methods have totally failed. This study was conducted using somatic embryogenesis as a rapid vegetative propagation technique for conservation of E. veratrifolia. For these purposes, effects of different types of plant growth regulators (PGRs), different types of explants, light and dark conditions, and the effect of gibberellic acid (GA3) and Paclobutrazol (PBZ) were investigated on somatic embryogenesis induction. Abscisic acid (ABA) pre-treatment effectiveness on inducting somatic embryogenesis and increasing the number of embryos per explant were investigated. Subsequently, 16 media were tested to find the best medium for plant regeneration and shoot and root proliferation. BA (3 mg L?1) resulted in a better response than the other PGRs by supporting the development of 17 embryos per protocorm. PBZ, which resulted in 11 embryos per explant, was better than GA3. FAST medium supplemented with organic substances was recognized as the best medium for plant regeneration and shoot and root proliferation. ABA pre-treatment had positive effect on somatic embryogenesis initiation. This study, for the first time, succeeded in finding a rapid and suitable protocol for propagation and genetic resource conservation of E. veratrifolia. 相似文献

Microbial indices and their spatial patterns are strongly affected by environmental factors. Spatial variability of soil properties is one of the most important causes of variability in soil microbial indices. This research was conducted in the Caspian forest to assess spatial variabilities and frequency distributions of microbial properties.Ninety soil samples were taken using a grid sampling design 40 9 40 m. Microbial indices, organic carbon,nitrogen and pH were determined. Soil variable distributions showed that microbial indices had abnormal distributions. Logarithmic transformation produced normal distribution. Spatial continuity using geostatistical(variogram) was studied and maps obtained by point kriging.The variograms revealed the presence of spatial autocorrelation. The results indicate that spatial dependence of soil microbial indices was affected by non-intrinsic factors and forest management procedures. The maps show that soil microbial indices and soil properties have spatial variability. The spatial pattern of microbial indices was correlated to organic carbon and nitrogen. 相似文献

Tolerance to flooding is crucial when thinking in promissory species for restoration of ecosystems prone to suffer soil water excess. In this study, we tested the flooding tolerance of two-years-old seedlings of Chestnut-leaved oak (Quercus castaneifolia C.A.Mey.) to determine whether it can be recommended for use in wetland restoration programs. Seedlings of Q. castaneifolia were subjected to three treatments: (1) control (C), (2) flooding for 60 days followed by a 42-day recovery period (F?+?R) and (3) continuous flooding for 102 days (F). Physiological performance, plant morphological changes and biomass accumulation were assessed. Results showed that, although net photosynthetic rates, stomatal conductance and transpiration decreased with prolonged flooding, when flood waters were removed, plants were able to recover their physiological activity (49–80% compared to controls). By contrast, when plants were continually flooded, their physiological activity decreased as well as the leaves experienced precocious senescence and wilting. Biomass responses paralleled physiological responses: leaf and root biomass were 42–49% higher under F?+?R treatment than under continuous flooding, and all plants under the F?+?R survived. Therefore, Q. castaneifolia appears as a promising species to be further studied when thinking re-vegetation of riverine areas and other temporarily flooded wetlands. 相似文献

Charcoal canker of oak is a common disease in the western regions of Iran, which has widely spread due to climate change and increasing drought in recent years. Biscogniauxia mediterranea and Obolarina persica are known as two agents of oak charcoal canker in Zagros forests causing the dieback of oak trees. The effects of charcoal disease agent pathogens on the growth and physiological response of two‐year‐old seedlings of Quercus brantii were evaluated under drought stress in greenhouse over a period of nine months. Survival was 21.7% lower in seedlings inoculated with B. mediterranea subjected to drought stress compared with control treatment. The length of lesions on stems was 68% more under charcoal pathogens in drought stressed compared with the length of lesions caused by charcoal pathogens only. On the other hand, stem lesion length caused by B. mediterranea was 25% greater than those caused by O. persica. Some morphophysiological characteristics were affected by charcoal pathogens alone and/or in combination with drought. Diameter, height and biomass decreased the most in seedlings inoculated with charcoal pathogens under drought stress. Pathogenic agents alone and in combination with drought stress reduced photosynthetic rates, stomatal conductance, transpiration, the maximum Rubisco activity (Vcmax), maximum photochemical efficiency of photosystem II (Fv/Fm) and xylem water potential. In general, drought increased the aggressiveness of fungus and intensification of their destructive effects in Quercus brantii. Moreover, there was no significant difference between the effect of B. mediterranea and O. persica on the morphophysiological parameters studied. 相似文献
