Message transmission: receptor controlled adenylate cyclase system   总被引:23,自引:0,他引:23  
The adenylate cyclase system is composed of an activating hormone or neurotransmitter (H), its receptor (R), the guanosine triphosphate (GTP) binding protein (Gs), and the catalytic unit (C). The activation of the receptor R involves a transient change in conformation, from a loose binding of the neurotransmitter H to an extremely tight interaction, termed locking. The system is regulated in the activation steps and also by three deactivation processes. A guanosine triphosphatase activity is built into the Gs protein so that the active GsGTP has only a limited lifetime during which it is able to activate C. In addition, the continued occupation of R by H causes desensitization of R. Finally, there are inhibitory receptors, such as alpha-adrenergic and opiate receptors, which inhibit the adenylate cyclase by way of a specific GTP binding protein (Gi). Yet to be determined are the conformational transformations of pure R on binding of an agonist or a partial agonist; the genes that code for the many different receptors that activate the adenylate cyclase, and the possibility that the G components interact with systems in the cell other than the adenylate cyclase.  相似文献

Epinephrine caused amylase secretion and K(+) release in rat parotid slices. Propranolol, which blocks beta-receptors, inhibited amylase secretion; phentolamine, which blocks alpha-receptors, inhibited K(+) release. Since enzyme secretion was associated with fusion of secretory granules to the cell membrane and K(+) release was associated with vacuole formation, it could be shown that both alpha- and beta-receptors are present in the same exocrine cell. The findings appear to exclude cyclic 3',5'-adenosine monophosphate as an intermediate in the alpha-receptor response.  相似文献

Straws of sex‐sorted sperm are usually packaged at a low concentration (e.g., ~2.1 × 106 sperm/ml) and cost significantly more than unsorted conventional semen from the same sire. In order to maximize the efficiency of using sex‐sorted sperm under in vitro fertilization conditions, the selection of an appropriate sperm separation technique is essential. In this study, the effect of using different silane‐coated silica colloid dilutions and layering configurations during centrifugation of sex‐sorted sperm was examined over an extended period of incubation time. Sperm recovery and viability after centrifugation using the colloid separation technique were measured along with several sperm motility parameters using CASA. For this purpose, frozen and thawed sex‐sorted sperm samples were centrifuged using mini‐volume single‐layer (40%, 60% and 80%) and mini‐volume two‐layer (45%/90%, 40%/80% and 30%/60%) separation configurations using PureSperm®. A single layer of 40% PureSperm® recovered significantly more sex‐sorted sperm (78.07% ± 2.28%) followed by a single layer of 80% PureSperm® (68.43% ± 2.33%). The lowest sperm recovery was obtained using a two‐layer PureSperm® dilution of 45%/90% (47.57% ± 2.33%). Single‐layer centrifugation recovered more sorted sperm (68.67% ± 1.74%) than two layer (53.74% ± 1.74%) (< .0001). A single layer of 80% PureSperm® exhibited the highest sorted sperm viability (72.01% ± 2.90%) after centrifugation (< .05). The mini‐volume single layer of 80% PureSperm® was determined to be an effective alternative to a two‐layer centrifugation configuration for sex‐sorted sperm selection. In addition, single‐layer colloid dilution of 80% performed either as well as or significantly outperformed the other treatments, as well as the control, with regard to motility (MOT) for all time periods of analysis.  相似文献

A graphic method was worked out for representing the statistical processed results of the determination of blood pH, pCO2, and base excess in a group of animals subjected to a metabolic test. The method is based on the determination of a selective mean and 95% confidence interval of the selective mean of the above-mentioned parameters in a group of animals; these mean values and confidence intervals are then plotted in a compensation graph. The area delimited in the graph by the upper and lower limits of the 95% confidence intervals of the selective means includes the mean values of the basic population with the probability of 94.7375%. The position of the plotted individual acid-base balance values of 422 animals, compared with the 60 areas delimited by the 95% confidence intervals of the selective means for the groups of animals, showed a very good correspondence in 91.23% of cases and partial correspondence in 8.74% of cases.  相似文献

Molecular self-assembly is of key importance for the rational design of advanced materials. To investigate the causal relation between molecular structure and the consequent self-assembled microstructure, self-assembled tubules of diacetylenic lipids were studied. Circular-dichroism studies give experimental evidence that the formation of tubules is driven by chiral molecular packing, in agreement with recent theories of tubules. On the basis of these results, a molecular mechanism for the formation of tubules is proposed.  相似文献

X-ray lasers (XRLs) have experimental average gains that are significantly less than calculated values and a persistently low level of spatial coherence. An XRL has been used both as an injected signal to a short XRL amplifier and as an interferometer beam to measure two-dimensional local gain and density profiles of the XRL plasma with a resolution near 1 micrometer. The measured local gain is in agreement with atomic models but is unexpectedly spatially inhomogeneous. This inhomogeneity is responsible for the low level of spatial coherence observed and helps explain the disparity between observed and simulated gains.  相似文献

The potential development of potato (Solanum tuberosum) as a low-cost eukaryotic system for the production of a commercially valuable enzyme feed supplement was examined. AFibrobacter succinogenes 1,3-1,4-β-glucanase [1,3-1,4-β-D-glucan 4-glucanohydro-lase] gene under the control of the constitutive cauliflower mosaic virus 35S promoter was transferred into the potato cultivar, Desiree. The presence of the β-glucanase cDNA in the plant genome of independent transgenic potato lines was confirmed by PCR and Southern analysis. Northern analysis identified the presence of the β-glucanase mRNA in the leaf tissue of transgenic plants. Furthermore, western analysis showedF. succinogenes β-glucanase accumulations of 0.1% and 0.05% of total soluble protein in the leaves and tubers, respectively. Specific activities of the enzyme in leaves (1693 units mg-1 β-glucanase) and tubers (2978 units mg-1 β-glucanase) were comparable to that previously reported for the enzyme produced in bacteria. Lyophilization of leaves had no effect on the specific activity of the β-glucanase, and only marginally influenced the specific activity of the enzyme expressed in tubers. Relative to the control line (cv. Desiree), tuber yields were significantly reduced by 28%-72% in all lines expressing theF. succinogenes β-glucanase, and microscopy showed that expression of the β-glucanase caused changes in cell wall structure. Results of this study demonstrate that a 1,3-1,4-β-glucanase can be expressed in potato tissues, and that potato plants have the potential to be used for the commercial production of heterologous enzymes.  相似文献

A controlled period of submaximal exercise on a treadmill was used as a standardised stress test in 6 young horses to monitor the effects of training. Circulating plasma concentrations of immunoreactive beta-endorphin (IR beta-EP) were measured before, during and after the exercise period. The stress test was conducted on 3 occasions during an intensive training program lasting 14 weeks. In week 3 a marked increase in plasma IR beta-EP (P = 0.003) was demonstrated as a result of training, but by the last exercise test performed in week 9 no significant increase in plasma IR beta-EP concentrations could be detected. During the training period the basal concentrations of plasma IR beta-EP significantly decreased (P = 0.0059). Plasma adrenocorticotrophin (ACTH) did not increase during exercise, although there was a trend of decreasing basal plasma ACTH by the end of the training period. It was concluded that a standardised work test acted as a mild stress to unfit horses, but as the horses' fitness increased the hormonal response to exercise diminished. Basal plasma beta-EP concentrations were decreased with increasing fitness.  相似文献
