The aim of this study was to evaluate the influence of rider's skill on the plasma cortisol levels of trekking horses on two courses, walking on field and forest courses (about 4.5 to 5.1 km each). Three riders of different skills did horse trekking (HT) in a tandem line under a fixed order: advanced‐leading, beginner‐second and intermediate‐last. A total of six horses were used and they experienced all positions in both courses; a total of 12 experiments were done. Blood samples were obtained before HT, immediately after and 2 h after HT. As a control, additional blood samples were obtained from the same horses on non‐riding days. Irrespective of the course and the rider's skill, the cortisol level before HT was higher than that of control (< 0.05). In both courses, the cortisol levels immediately after HT ridden by the advanced rider were higher than that of control (< 0.05). However, in every case, the cortisol level 2 h after HT was closely similar to the level of the control. Thus, we concluded the stress of trekking horse was not sufficient to disturb the circadian rhythm of the cortisol level, irrespective of the course and the rider's skill.  相似文献

During the period from 2001 to the following year, progenital diseases had been epidemic among the draft stallions and mares pastured together in Iwate Prefecture, the northeastern district of Japan. A stallion and 8 of 31 mares were affected in 2001, and 1 of 2 stallions and 10 of 36 mares in 2002. The clinical symptoms consisted of the formation of papules, pustules, ulcers and scabs on the progenital skin and mucosa in stallions and mares. In 2002, Equine herpesvirus 3 (EHV3) was isolated from 2 mares and the glycoprotein G gene of the virus detected from a stallion and 4 mares by polymerase chain reaction. Serum neutralizing tests showed that 12 of 38 horses, 10 clinically and 2 subclinically affected, changed to be positive for the EHV3 antibody. The results suggest that the horses were affected with equine coital exanthema (ECE) through coitus. Five mares with the antibody at the pre-pastured period may have been the possible origins of EHV3 infection in 2002, although the exact origin in 2001 remains unknown. The artificial insemination was performed for the prevention of ECE spreading through coitus on the pasture in 2003. There was no epidemic of the disease in 31 mares, although 3 mares with the antibody at the pre-pastured period showed the significant increase in the titers during the pastured period.  相似文献

The E2 regions of 177 bovine viral diarrhea virus (BVDV) strains isolated in Japan between 1957 and 2006 were analyzed for genotyping. The strains were classified into 8 genotypes (1a, 1b, 1c, 1d, 1e, 1f, So and 2a) based on the phylogenetic analysis. The restriction fragment length polymorphism (RFLP) analysis of the RT-PCR products using 6 selected enzymes (Apo I, Mly I, BstAP I, Pvu II, Ear I, EcoR V) disclosed the cutting patterns classified into 11 groups (I-XI), each of that consisted of strains belonging to a single genotype. Namely, groups-I and -II were composed by genotype-1a strains, groups-III and -IV by 1b strains, and groups-V and -VI by 1c strains. Other groups-VII, -VIII, -IX, -X and -XI comprised genotypes-1d, -1e, -1f, -So and -2a strains, respectively. The results suggest that the RFLP analysis can simply and rapidly differentiate the 8 genotypes of BVDV strains.  相似文献

Satellite telemetry from 26 loggerhead (Caretta caretta) and 10 olive ridley (Lepidochelys olivacea) sea turtles captured and released from pelagic longline fishing gear provided information on the turtles’ position and movement in the central North Pacific. These data together with environmental data from satellite remote sensing are used to describe the oceanic habitat used by these turtles. The results indicate that loggerheads travel westward, move seasonally north and south primarily through the region 28–40°N, and occupy sea surface temperatures (SST) of 15–25°C. Their dive depth distribution indicated that they spend 40% of their time at the surface and 90% of their time at depths <40 m. Loggerheads are found in association with fronts, eddies, and geostrophic currents. Specifically, the Transition Zone Chlorophyll Front (TZCF) and the southern edge of the Kuroshio Extension Current (KEC) appear to be important forage and migration habitats for loggerheads. In contrast, olive ridleys were found primarily south of loggerhead habitat in the region 8–31°N latitude, occupying warmer water with SSTs of 23–28°C. They have a deeper dive pattern than loggerheads, spending only 20% of their time at the surface and 60% shallower than 40 m. However, the three olive ridleys identified from genetics to be of western Pacific origin spent some time associated with major ocean currents, specifically the southern edge of the KEC, the North Equatorial Current (NEC), and the Equatorial Counter Current (ECC). These habitats were not used by any olive ridleys of eastern Pacific origin suggesting that olive ridleys from different populations may occupy different oceanic habitats.  相似文献

SUMMARY: Tissue type transglutaminase (TGase) was purified from scallop striated adductor muscle with successive chromatographies of DE-52 cellulose, Sephacryl S-300, and Mono Q columns. The yield and purification of the enzymatic activity was 16.6% and 101.9-fold, respectively. The molecular mass of purified enzyme was estimated to be 95 kDa by sodium dodecylsulfate–polyacrylamide gel electrophoresis analysis. Scallop TGase was Ca2+-dependent and strongly inactivated by ρ-chloromercuribenzoic acid, N -ethylmaleimide, Cu2+, and Zn2+, meaning it belongs to the thiol group of enzymes as well as being a mammalian enzyme. When scallop TGase was incubated in 0.5 M NaCl without substrate for 2 h at 20°C and pH 7.5, enzymatic activity decreased to 14.4% of its original. However, a conformational change in the TGase molecule was not detected by either fluorescent, ultraviolet, and circular dichroism spectra analyses compared to the enzyme incubated without NaCl. In addition, the enzyme inactivated by NaCl was partially recovered by the dilution of salt concentration, which means that the NaCl-induced inactivation process is reversible to some extent. These results suggest that NaCl-induced modulation of the TGase molecule occurs via a small conformational change.  相似文献

The ability of sugi wood carbonized at 1000°C to adsorb mercury was examined using aqueous solutions of mercuric chloride. Parameters studied include contact time, pH, adsorption temperature, and initial concentration of mercury in solution. Results showed that sugi wood carbonized at 1000°C could effectively remove mercury from aqueous solutions. The carbonized wood showed high adsorption ability for mercury at a wide pH range (pH 3–9), but its ability drastically decreased at pH 11. Adsorption decreased with increases in adsorption temperatures, indicating that the processes were exothermic in nature. Adsorption was found to follow the Freundlich isotherm model. The adsorption capacity of carbonized sugi wood was comparable to that of commercial activated carbon.  相似文献

Hyperlipidemic ocular lesions are described for Watanabe heritable hyperlipidemic (WHHL) rabbits. Male WHHL rabbits 8 months old exhibited serum hyperlipidemia and ophthalmoscopically yellowish-white lesions along the corneoscleral junction and in the iris. Histopathologically, foamy macrophages aggregated in the stroma of the cornea, iris, and ciliary body were observed. These findings have been interpreted as lipid keratopathy. In addition, multiple clusters of a large number of foamy macrophages occurred throughout the choroid and sclera in association with the blood vessels. The lesions in the choroid and sclera could not be detected ophthalmoscopy, yet were much more prominent than those in the cornea, iris, and ciliary body, suggesting greater involvement and earlier onset of lipidosis at these sites associated with hyperlipidemia in WHHL rabbits.  相似文献

The aim of this study was to establish a method for estimating loading capacity for Japanese native horses by gait analysis using an accelerometer. Six mares of Japanese native horses were used. The acceleration of each horse was recorded during walking and trotting along a straight course at a sampling frequency of 200 Hz. Each horse performed 12 tests: one test with a loaded weight of 80 kg (First 80 kg) followed by 10 tests with random loaded weights between 85 kg and 130 kg and a final test with a loaded weight of 80 kg again. The time series of acceleration was subjected to fast Fourier transformation, and the autocorrelation coefficient was calculated. The first two peaks of the autocorrelation were defined as symmetry and regularity of the gait. At trot, symmetries in the 100, 110, and 125 kg tests were significantly lower than that in First 80 kg (P < 0.05, by analysis of covariance and Sidak's test). These results imply that the maximum permissible load weight is less than 100 kg, which is 29% of the body weight of Japanese native horses. Our method is a widely applicable and welfare‐friendly method for estimating maximum permissible load weights of horses.  相似文献
