Variant Creutzfeldt-Jakob disease (vCJD) is a unique and highly distinctive clinicopathological and molecular phenotype of human prion disease associated with infection with bovine spongiform encephalopathy (BSE)-like prions. Here, we found that generation of this phenotype in transgenic mice required expression of human prion protein (PrP) with methionine 129. Expression of human PrP with valine 129 resulted in a distinct phenotype and, remarkably, persistence of a barrier to transmission of BSE-derived prions on subpassage. Polymorphic residue 129 of human PrP dictated propagation of distinct prion strains after BSE prion infection. Thus, primary and secondary human infection with BSE-derived prions may result in sporadic CJD-like or novel phenotypes in addition to vCJD, depending on the genotype of the prion source and the recipient.  相似文献

We have studied the ordering dynamics of the striped patterns of a single layer of cylindrical block copolymer microdomains in a thin film. By tracking disclinations during annealing with time-lapse atomic force microscopy, we observe a dominant mechanism of disclination annihilation involving three or four disclinations (quadrupoles). Pairwise disclination annihilation events are suppressed as a result of the topological constraints in this system. The kinetic scaling laws with exponents observed here are consistent with topologically allowed annihilation events involving multiple disclinations. The results provide insight into two-dimensional pattern formation and may lead to the successful application of block copolymer lithography.  相似文献

Purpose

The purpose was to develop a netbag technique suitable for burying biochar in a compost or composting medium, followed by its collection in a clean state (i.e., free of compost debris) without loss or damage, for physicochemical analyses. Collection in a clean state is required to ensure that the analytical data of various biochars are representative and comparable. Five criteria were identified to evaluate the suitability of netbag.

Materials and methods

A small netbag (3.5?×?3.5 cm) was developed using nylon fabric (30-μm mesh) to contain ~1 g of biochar. A production system was developed to make 15 netbags per batch. Unlike commonly used litterbags, polypropylene was used to seal netbags. Two experiments were conducted in which three biochars, made from macadamia nutshell, hardwood shaving, and chicken litter, were co-composted with chicken manure and sawdust and also incubated with a chicken litter-based commercial compost. Biochars were added at the rates of 5 or 10 % in the co-composting and 10 or 20 % in the incubation experiments. The biochar-containing netbags were buried in the co-composting and incubation mediums for 133 days. Various physicochemical analyses were conducted with netbag-biochars and their compost mediums.

Results and discussion

The netbags collected after both experiments showed no visible sign of degradation. The weight of netbag-biochars from co-composting and incubation systems did not reduce significantly over the experimental period, thereby indicating no loss of biochar. No visible evidence of entry of solid particles from compost medium was found on the netbag-biochars. Pretests indicated that the netbag and biochars absorbed pore solution from the medium. Findings showed that elements translocated between the netbag-biochar and compost medium. A colony of coccus bacteria was found on the surface of composted chicken litter biochar, denoting probable entry of bacteria from compost medium. Unlike conventional litterbags, the netbags were suitable for burying and extracting biochar in compost/composting mediums due to smaller size, smaller mesh, and strong sealing with polypropylene.

Conclusions

The netbags addressed all the five criteria. Therefore, it was concluded that, in the co-compost or incubated-compost medium, the biochar retained in the netbag and the biochar mixed with the medium were exposed to a similar bio-oxidative environment, and netbag-biochar represents the biochar in the medium. This means that these netbags can be used as a convenient means to examine the effects of the composting process or incubation in compost on biochar.

Training populations for pedigree and genomic prediction in plant breeding programmes are largely updated with superior genotypes from multi-environment trials, where they are tested with the goal of variety development. Such a selective phenotyping has, however, a negative impact on prediction abilities, especially when only a subset of breeding lines can be tested, as for laborious and costly to phenotype traits. This study focused, thus, on investigating the impact of selective phenotyping in the training population of an applied wheat breeding programme, and assessing the potential to mitigate this impact by pedigree and genomic multi-trait prediction models as well as bi-directionally selected training populations for several baking quality parameters. Combining both pedigree and genomic information in multi-trait prediction models with pre-existing phenotypic information for protein content and sedimentation value compensated for the observed loss in prediction ability, while entering few inferior breeding lines into the training population further mitigated the impact of selective phenotyping and even led to a slight increase of prediction ability in comparison to a randomly chosen training population.  相似文献

Bark beetles (Coleoptera: Curculionidae, Scolytinae) are commonly recognised as important agents of tree mortality in coniferous forests of the Western Carpathians. They, together with weevils, are consistently associated with ophiostomatoid fungi. Information regarding conifer beetle-associated fungi in the Western Carpathians remains incomplete and unreliable, particularly with respect to fir-infesting bark beetles. This study aims to clarify associations between fungi in the genera Graphilbum, Leptographium, Ophiostoma and Sporothrix (Ophiostomatales) and their beetle vectors in Norway spruce (Picea abies), European larch (Larix decidua) and silver fir (Abies alba). Samples associated with 20 bark beetle species and weevils were collected from nine stands in Poland and the Czech Republic. Fungi were isolated from adult beetles and galleries. Isolates were identified based on morphology, DNA sequence comparisons for four gene regions (ITS, LSU, ß-tubulin, TEF 1-α) and phylogenetic analyses. In total, 46 distinct taxa were identified, including 25 known and 21 currently unknown species. Several associations between fungi and subcortical insects were recorded for the first time. In addition, O. borealis and O. quercus were detected from A. alba for the first time. The composition of the fungal communities varied among the studied tree species and to a lesser degree among the beetle species. The spruce-infesting bark beetles were commonly associated with species of Leptographium s. l. and Ophiostoma s. str.; the larch-infesting bark beetles were often associated with Ophiostoma s. str. and Sporothrix, while the fir-infesting bark beetles were commonly associated with Ophiostoma s. str. and Graphilbum. The most commonly encountered fungal associates of the examined insects were (a) Grosmannia cucullata, G. piceiperda, Grosmannia sp. 1, Ophiostoma macroclavatum and O. piceae with the spruce-infesting bark beetles; (b) O. pseudocatenulatum and Sporothrix sp. 1 with the larch-infesting bark beetles; and (c) O. piceae, Ophiostoma sp. 2 and Graphilbum sp. 2 with the fir-infesting bark beetles. The differences in fungal associates among the bark beetle species occurring on P. abies, L. decidua and A. alba could be linked to the different habitats that these beetles occupy.  相似文献
