Two experiments were conducted to determine the effects of supplementing ruminally degradable intake protein (DIP) or ruminally undegradable intake protein (UIP) on N balance (Exp. 1; n = 6 wethers; initial BW = 48.7 +/- 4.6 kg) and site and extent of digestion (Exp. 2; n = 5 wethers; initial BW = 36.9 +/- 3.1 kg) in whiteface wethers consuming (as-fed basis) 69% blue grama and 31% love grass hay (mixture = 7.5% CP, 73.0% NDF, 36.0% ADF [DM basis]). Treatments were 1) no supplement (Control), 2) a supplement (219 g/d, as-fed basis) low in UIP (70 g/d of CP; 24.8 g/d of UIP), and 3) a supplement (219 g/d, as-fed basis) high in UIP (70 g/d of CP; 37.1 g/d of UIP). Both experiments were replicated 3 x 3 Latin square designs, with identical feeding and supplementation. Wethers had ad libitum access to the forage mixture and fresh water, and received supplement once daily. In Exp.1, forage intake (percentage of BW) was greatest (P = 0.04) for control, but total DMI (g/d) was greatest (P = 0.05) for lambs consuming supplement. Apparent total-tract OM digestibility was numerically greater (P = 0.11) for supplemented wethers than for controls, whereas total-tract ADF digestibility tended (P = 0.08) to be greater for control wethers. Lambs fed supplements consumed and retained more (P < or = 0.01) N (% of N intake) compared with controls, but no difference (P = 0.22) was observed between low and high UIP treatments. Similar to Exp. 1, forage intake (percentage of BW) tended (P = 0.06) to be greater for control than for supplemented wethers in Exp. 2. Ruminal NDF digestibility was 16.3% greater (P = 0.02) for supplemented wethers than for controls. Postruminal NDF and N digestibilities were greatest (P < or = 0.03) for controls, but apparent OM digestibility did not differ among treatments at all sites. Duodenal N flow was greatest (P = 0.05) for high UIP and least for control wethers. Nonmicrobial N flow was greater (P = 0.02) for high UIP compared with low UIP or controls. Control wethers had greater (P = 0.05) microbial efficiency. Ruminal ammonia concentration tended (P = 0.08) to be greatest for wethers fed low UIP and least for controls, with high-UIP wethers having intermediate ammonia concentrations. Results from these experiments suggest that in lambs fed low-quality forage there was no difference in apparent total-tract digestion or N balance (percentage of N intake) between lambs fed supplements that had the same CP but differed in the proportion of UIP and DIP; however, supplementing protein (regardless of UIP:DIP ratio) to wethers consuming low-quality forage increased N balance.  相似文献

It is believed that milk production is determined by the number and activity of mammary secretory cells. Secretory activity, as assessed by milk volume, depends on secretion of the major osmole in milk, lactose, which is produced by lactose synthase. The amount of either of the two proteins in lactose synthase may regulate milk production. The objective of this study was to determine whether the concentrations in milk of the two components of lactose synthase, α‐lactalbumin (α‐LA) and β1,4‐galactosyltransferase (B4GALT), were related to genetic background, stage of lactation, breed or parity of dairy cows. α‐Lactalbumin and B4GALT concentrations were measured by ELISA and by enzyme assays, respectively, from single milk samples. Two herds with a total of 279 cows were used in the analysis. One herd contained Ayrshire, Brown Swiss, Holstein and Jersey cows; the second herd contained two groups of cows; Holsteins selected for high milk production and Holsteins with 1960s genetics. The α‐LA concentration in milk was greater in Jerseys and Ayrshires than in Holsteins and Brown Swiss. However, no difference in α‐LA concentration was observed in milk from high and low genetic merit cows in the Minnesota herd or among different genetic backgrounds in the Illinois herd. β1,4‐Galactosyltransferase concentrations were similar for all groups that were analyzed. α‐Lactalbumin concentrations were positively correlated with milk protein concentration, milk fat concentration and lactose concentration. β1,4‐Galactosyltransferase concentration in milk exhibited a strong positive correlation with number of days in milk. Although the concentration of B4GALT increased as lactation progressed, the values did not show any correlation with persistency of lactation or late lactation milk production. In conclusion, this survey shows that the two components of lactose synthase are each correlated to protein concentration and individually correlated to the concentration of other milk components and stage of lactation.  相似文献

A calcium-selective electrode with solid ion exchanger was prepared from a solution of the calcium salt of a dialkylphosphoric acid in collodion. Proteolytically produced carboxyl-terminal fragments of the human immunodeficiency virus type-1 (HIV-1) Tat protein that include a conserved region rich in arginine and lysine bind specifically to transactivation response RNA sequences (TAR). A chemically synthesized 14-residue peptide spanning the basic subdomain also recognizes TAR, identifying this subdomain as central for RNA interaction. TAR RNA forms a stable hairpin that includes a six-residue loop, a trinucleotide pyrimidine bulge, and extensive duplex structure. Competition and interference experiments show that the Tat-derived fragments bind to double-stranded RNA and interact specifically at the pyrimidine bulge and adjacent duplex of TAR.  相似文献

Twenty-eight dogs immunized to dinitrophenol-ascaris (DNP-ASC) at birth and then three times at two week intervals produced serum anti-DNP antibody. The IgM response was detected one week after primary immunization and lasted for up to five weeks. The IgE and IgG antibody response in general was not present until week three but persisted through the immunization schedule. Although variation in the level and duration of the antibody response was detected between individual dogs, each dog did have a response that included all three isotypes examined.  相似文献

By influencing belowground processes, streamside vegetation affects soil processes important to surface water quality. We conducted this study to compare root distributions and dynamics, and total soil respiration among six sites comprising an agricultural buffer system: poplar (Populus × euroamericana‘ Eugenei), switchgrass, cool-season pasture grasses, corn (Zea mays L.), and soybean (Glycine max (L.) Merr.). The dynamics of fine (0--2 mm) and small roots (2--5 mm) were assessed by sequentially collecting 35 cm deep, 5.4 cm diameter cores from April through November. Coarse roots were described by excavating 1 × 1 × 2 m pits and collecting all roots in 20 cm depth increments. Root distributions within the soil profile were determined by counting roots that intersected the walls of the excavated pits. Soil respiration was measured monthly from July to October using the soda-lime technique. Over the sampling period, live fine-root biomass in the top 35 cm of soil averaged over 6 Mg ha-1 for the cool-season grass, poplar, and switchgrass sites while root biomass in the crop fields was < 2.3 Mg ha-1 at its maximum. Roots of trees, cool-season grasses, and switchgrass extended to more than 1.5 m in depth, with switchgrass roots being more widely distributed in deeper horizons. Root density was significantly greater under switchgrass and cool-season grasses than under corn or soybean. Soil respiration rates, which ranged from 1.4--7.2 g C m-2 day-1, were up to twice as high under the poplar, switchgrass and cool-season grasses as in the cropped fields. Abundant fine roots, deep rooting depths, and high soil respiration rates in the multispecies riparian buffer zones suggest that these buffer systems added more organic matter to the soil profile, and therefore provided better conditions for nutrient sequestration within the riparian buffers.
