The incidence of rotaviruses in calves, foals, dogs and cats in the Dunedin urban and rural areas was investigated using electron microscopy and enzyme-linked immunosorbent assays. Of the 283 faecal specimens examined, 26% were positive for rotavirus. Comparison of the genetic electropherotypes was made by separating the viral dsRNA segments using polyacrylamide gel electrophoresis. It is possible that rotavirus infection is a zoonotic disease.  相似文献

Yellow cypress (Chamaecyparis obtusa (Siebold & Zucc.) Endl. var. formosana (Hayata) Rehder) is the predominant tree species of Taiwan's nutrient-poor, mountain fog forests. Little is known about the potential contribution of solute uptake from fog to the overall nutrition of these trees. Shoots of yellow cypress seedlings were misted with artificial fog containing the tracer rubidium (Rb) in laboratory and field experiments to determine if there is solute uptake from the fog. After misting shoots for six weeks, substantial amounts of tracer were detected in unexposed roots by inductively coupled plasma mass spectroscopy bulk analysis. Possible routes of entry were examined by element imaging with energy dispersive X-ray analysis. Direct uptake of the tracer into leaves across the cuticle and epidermis was small, excluding this as the major uptake path. Accumulations of Rb were found on leaf surfaces along the edges of the leaves. The almost daily changes in fog coverage and air humidity may enhance the accumulation of fog solutes at leaf edges. Accumulation of Rb was also found in narrow clefts between opposite leaves and between the outermost and underlying alternating stacked leaves. The clefts provide a direct passage from the leaf surface to the space beneath the imbricate leaves and the underlying alternate leaves, possibly facilitating solute uptake from fog, which in turn may contribute to the nutrition of yellow cypress.  相似文献

beta-Secretase plays a critical role in beta-amyloid formation and thus provides a therapeutic target for Alzheimer's disease. Inhibitor design has usually focused on active-site binding, neglecting the subcellular localization of active enzyme. We have addressed this issue by synthesizing a membrane-anchored version of a beta-secretase transition-state inhibitor by linking it to a sterol moiety. Thus, we targeted the inhibitor to active beta-secretase found in endosomes and also reduced the dimensionality of the inhibitor, increasing its local membrane concentration. This inhibitor reduced enzyme activity much more efficiently than did the free inhibitor in cultured cells and in vivo. In addition to effectively targeting beta-secretase, this strategy could also be used in designing potent drugs against other membrane protein targets.  相似文献

Aerosol and total vapor-phase Hg concentrations in air have been measured at Walker Branch Watershed, Tennessee for ≈ 2 yr. Airborne Hg at this site is dominated by vapor forms which exhibit a strong seasonal cycle, with summer maxima that correspond to elevated air temperature. Concentrations in this forest are near background levels; however, concentrations at a site within 3 km are significantly elevated due to emissions from Hg-contaminated soils. The concentration data have been combined with a recently modified dry deposition model to estimate dry deposition fluxes to the deciduous forest at Walker Branch. Weekly mean modeled Vd values for Hg° ranged from <0.01 (winter) to > 0.1 (summer) cm s1. Weekly dry deposition fluxes ranged from <0.1 μg m−2 during winter to > 1.0 μgg m−2 in the summer. Our dry deposition estimates plus limited measurements of wet deposition in this area indicate that dry deposition may be the dominant input process in this forest, at least during the summer.  相似文献

Mercury is released into the atmosphere from natural and anthropogenic sources. Once in the air, Hg species will be dispersed, chemically and physically transformed, and transported over varying distances (regional, continental and global scales) while concurrently being subjected to removal processes. Modeling of the aerial transport and deposition of Hg to receptor sites offers an attractive approach for integrating existing experimental data and represents an important tool for advancing our understanding of environmental Hg pollution. Comprehensive modeling efforts of the atmospheric pathways and fate of this heavy metal require knowledge of its atmospheric chemistry. To update and consolidate available information relevant to the atmospheric chemistry of Hg, a computer-assisted search was undertaken of the primary scientific literature published during the last ten years.
