Earlier research with endogeic and epigeic earthworm species in loamy arable soil has shown that both earthworm groups can increase nitrous oxide (N2O) emissions, provided that crop residue placement matches the feeding strategy of the earthworm ecological group(s). However, it is not yet clear whether these effects also occur in sandy soils which typically contain less soil organic matter and have low soil aggregation levels. Here, we aimed to quantify N2O emissions as affected by endogeic and/or epigeic earthworm species, and to relate changes in N2O emissions to earthworm-induced changes in soil properties in a sandy soil. A 90 day mesocosm study was conducted with sandy soil and 15N-labeled radish (Raphanus sativus cv. Adagio L.) residue applied on top. Treatments included: (i) no earthworm addition, (ii) addition of the endogeic species Aporrectodea caliginosa (Savigny), (iii) addition of the epigeic species Lumbricus rubellus (Hoffmeister), and (iv) both species combined. An additional treatment was included without earthworms and with residue manually incorporated into the soil. L. rubellus significantly increased cumulative N2O emissions from 228 to 859 μg N2O–N kg?1 (F1,12 = 83.12, P < 0.001), whereas A. caliginosa did not affect N2O emissions. In contrast to earlier studies in loamy soil, no positive interaction between both species with regard to N2O emissions was found. This was probably related to high competition for organic resources in the relatively poor soil and a low potential for stable soil aggregate formation (and associated anaerobic microsites) by endogeic worms in sandy soil. 15N isotope analysis revealed that the activity of L. rubellus significantly increased (F1,12 = 6.20, P = 0.028) the recovery of 15N in the 250–8000 μm size fraction, indicating incorporation of crop residues into the mineral soil. When residues were manually incorporated, N2O emissions were significantly (P < 0.008) lower (509 μg N2O–N kg?1) than when incorporated by L. rubellus. The high N2O emissions in the presence of L. rubellus, when compared to manual mixing, suggest a stimulation of microbial activity and/or changes in the microbial community composition. Insights on the earthworm effects on N2O emission from such soils are discussed.  相似文献

The ostrich industry experiences a high rate of embryonic mortalities during artificial incubation of eggs. Embryonic deaths were studied from data recorded on 37,740 fertile eggs incubated artificially during the 1998-2005 breeding seasons. Roughly 10,000 eggs that sustained embryonic mortalities were classified according to the stage and nature of death, i.e. before 21 days of incubation, after 21 days of incubation, deaths after pipping and rotten eggs. Although infection may have played a role in approximately 1300 rotten eggs, no detailed knowledge of the pathogens involved was available. The remainder of deaths could not be related to pathogens and the deaths were thus generally referred to as non-infectious. The overall level of embryonic mortality in all the eggs studied was 28.5 %. Overall embryonic mortality was affected by incubator, with higher levels (57.0 %) found in eggs incubated in an African Incubator and also in eggs that were transferred between incubators during incubation (38.1%). Overall embryonic mortality also increased in eggs produced by older females. Eggs produced in the autumn had the highest level of embryonic mortality at 53.6 %, whereas eggs produced in the winter had a marginally higher level of embryonic mortalities of 29.2 % compared with eggs produced during summer (27.4 %). Eggs produced by South African (SA) Black males crossed to Zimbabwean Blue females had high levels of embryonic losses of 45.7 %. The embryonic mortality of eggs produced by SA Blacks or Zimbabwean Blue breeding birds subjected to pure breeding was similar at approximately 33-34 %, but embryonic mortality was improved in eggs produced by Zimbabwean Blue males crossed to SA Black females (27 %). Embryonic mortality was increased in eggs that were set directly (32.0 %) or subjected to longer than 6 days of storage (43.5 %). Embryonic mortality was affected by year. The results that were obtained will assist in determining non-infectious factors that have a negative effect on hatching success. Steps can thus be taken to eliminate such factors that may compromise hatching success.  相似文献

Summary When using two restriction enzymes and DNA probes from different sources, high levels of restriction fragment length polymorphism (RFLP) were observed in Southern blot hybridisation experiments, among 6 di(ha)ploid genotypes ofSolanum tuberosum and some closely related species. In 3 F1 sample populations of 14 individuals each, both heterozygosity and segregation of hybridisation patterns were observed. The non-radioactive biotin-dUTP/SA-AP method for DNA detection on Southern blots proved to be satisfactory and reliable.  相似文献

The influence of succinylation on lysozyme is studied using circular dichroism, fluorescence spectroscopy, and differential scanning calorimetry. The spectroscopic data reveal that at room temperature the structures of succinylated lysozyme and native lysozyme are similar. However, the calorimetric results show that the thermal stability of succinylated lysozyme is lower than that of native lysozyme. For succinylated lysozyme, the denaturation temperature (Td) varies in the range of 325-333 K (52-60 degrees C) and the associated denaturation enthalpy (DeltadenH) varies between 225 and 410 kJ/mol. For lysozyme, Td is 342-349 K (69-76 degrees C) and DeltadenH is 440-500 kJ/mol. From these data, the change in the heat capacity (DeltadenCp) upon thermal denaturation is derived. For lysozyme, DeltadenCp is 7.5 kJ/mol/K, and for succinylated lysozyme, it is 16.7 kJ/mol/K. The value of DeltadenCp for lysozyme is comparable to previously reported values. The high value of DeltadenCp for succinylated lysozyme is explained in terms of an extended degree of unfolding of the secondary structure and exposure of the apolar parts of the succinyl groups. Furthermore, the Gibbs energy of denaturation, as a function of temperature, derived from the thermodynamic analysis of the calorimetric data, indicates a cold-denaturated state of succinylated lysozyme below 20 degrees C. However, because a denatured state at low temperatures could not be detected by CD or fluorescence measurements, the native state may be considered to be metastable at those conditions.  相似文献

The molecular structures determine the physical properties of milk proteins and are important for the texture of many dairy-based foods. Bovine alpha-lactalbumin (alpha-LA) is a globular 123 amino acid Ca(2+) binding milk protein. Modification with microbial Ca(2+) independent transglutaminase (MTGase) was used to modify lysines and glutamines in holo and apo alpha-LA. At 30 degrees C no lysines or glutamines are modified in holo alpha-LA, whereas in apo alpha-LA lysines 13, 16, 108, and 114, and glutamines 39 and 43, are modified. At 50 degrees C lysines 13, 16, 108, and 114, but no glutamines, are modified in holo alpha-LA, whereas in apo alpha-LA lysines 5, 13, 16, 108, and 114, and glutamines 39, 43, 54, 65, and 117, are modified. The methods presented here offer the possibility to manipulate the availabilities of residues in alpha-LA to the MTGase reaction and enable the preparation of alpha-LA species with different degrees of modification and hence with different physical properties.  相似文献

Sequential sampling of rain on a volume basis presents fewer problems than sampling on a time basis. The optimum diameter of the collector opening depends on the rainfall statistics, the size of the sample needed for chemical analysis and the total number of samples that has to be collected. The sampler described here consists of a square polyethylene funnel of 0.64 m2 and a fraction collector with 250 polypropylene tubes of 39 ml, which is activated by a rain detector. Each fraction corresponds to about 0.05 mm of rainfall. The funnel is covered by a roller blind that automatically rolls up when it starts to rain. Evaluation of the system leads to the conclusion that the collector opening can be made smaller (by 0.20 m2), that the measured rainfall rate depends on the averaging time, and that all other surfaces of the sampling set-up should be covered with inert material.  相似文献

After a 35 days growth on nutrient solutions with NO 3 NH4NO3 and NH+ 4 as nitrogen source (pH 4.2) dry matter yield of the sorghum genotype SC0283 was much less affected by Al (1.5 mg‐1) than that of the genotype NB9040. With NO 3 as the sole nitrogen source only growth of the NB9040 plants was significantly reduced. Since OH efflux, shoot Al content and concentrations of all major nutrients of both genotypes were almost equal, a higher sensitivity to Al may underlie the lower Al tolerance of the NB9040 genotype. In the presence of NH.‐N Al again lowered d.m. yield of the NB plants. With SCO283 significant Al effects on d.m. yield were observed only with NH4NO3. Aluminum drastically increased the amount of protons released per unit of root surface area, especially with the NB9040 line. This shift in proton flux density was partly the result of a decrease of the specific root surface area and partly due to enhanced excess of catlonic nutrients taken up. With NH4NO3‐fed plants the latter could almost completely be attributed to a changed N preference brought about by inhibited uptake of NO 3 and a simultaneous enhanced NH, absorption. Although both proton efflux and NH+ 4 preference of the NB plants were severely increased by Al, relative yields of this genotype were not lowered by NH+ 4. This can probably be explained by (1) the high NH, sensitivity of this cultivar through which Al effects can be masked and (2) the continuous adjustment of the solution pH through which rhizosphere conditions were prevented.  相似文献

In situ enrichment of bread with arabinoxylan‐oligosaccharides (AXOS) through enzymic degradation of wheat flour arabinoxylan (AX) by the hyperthermophilic xylanase B from Thermotoga maritima (rXTMB) was studied. The xylanolytic activity of rXTMB during breadmaking was essentially restricted to the baking phase. This prevented problems with dough processability and bread quality that generally are associated with thorough hydrolysis of the flour AX during dough mixing and fermentation. rXTMB action did not affect loaf volume. Bread with a dry matter AXOS content of 1.5% was obtained. Further increase in bread AXOS levels was achieved by combining rXTMB with xylanases from Pseudoalteromonas haloplanktis or Bacillus subtilis. Remarkably, such a combination synergistically increased the specific bread loaf volume. Assuming an average daily consumption of 180 g of fresh bread, the bread AXOS levels suffice to provide a substantial part of the AXOS intake leading to desired physiological effects in humans.  相似文献
