Foot-and-mouth disease (FMD) is an economically important disease of cloven-hoofed animals that is primarily controlled by vaccination of susceptible animals and movement restrictions for animals and animal-derived products in South Africa. Vaccination using aluminium hydroxide gel-saponin (AS) adjuvanted vaccines containing the South African Territories (SAT) serotypes has been shown to be effective both in ensuring that disease does not spread from the endemic to the free zone and in controlling outbreaks in the free zone. Various vaccine formulations containing antigens derived from the SAT serotypes were tested in cattle that were challenged 1 year later. Both the AS and ISA 206B vaccines adjuvanted with saponin protected cattle against virulent virus challenge. The oil-based ISA 206B-adjuvanted vaccine with and without stimulators was evaluated in a field trial and both elicited antibody responses that lasted for 1 year. Furthermore, the ISA 206 adjuvanted FMD vaccine protected groups of cattle against homologous virus challenge at very low payloads, while pigs vaccinated with an emergency ISA 206B-based FMD vaccine containing the SAT 1 vaccine strains were protected against the heterologous SAT 1 outbreak strain. 相似文献

Background
Recently, a Risk Assessment methodology was applied to animal welfare issues in a report of the European Food Safety Authority (EFSA) on intensively housed calves.Methods
Because this is a new and potentially influential approach to derive conclusions on animal welfare issues, a so-called semantic-modelling type ''validation'' study was conducted by asking expert scientists, who had been involved or quoted in the report, to give welfare scores for housing systems and for welfare hazards.Results
Kendall''s coefficient of concordance among experts (n = 24) was highly significant (P < 0.001), but low (0.29 and 0.18 for housing systems and hazards respectively). Overall correlations with EFSA scores were significant only for experts with a veterinary or mixed (veterinary and applied ethological) background. Significant differences in welfare scores were found between housing systems, between hazards, and between experts with different backgrounds. For example, veterinarians gave higher overall welfare scores for housing systems than ethologists did, probably reflecting a difference in their perception of animal welfare.Systems with the lowest scores were veal calves kept individually in so-called "baby boxes" (veal crates) or in small groups, and feedlots. A suckler herd on pasture was rated as the best for calf welfare. The main hazards were related to underfeeding, inadequate colostrum intake, poor stockperson education, insufficient space, inadequate roughage, iron deficiency, inadequate ventilation, poor floor conditions and no bedding. Points for improvement of the Risk Assessment applied to animal welfare include linking information, reporting uncertainty and transparency about underlying values.Conclusion

In recent years it has become clear that cell-mediated immunity is playing a role in the control of lentivirus infections. In particular, cytotoxic T lymphocyte responses have been associated with improved outcome of infection, especially those directed against the regulatory proteins like Rev and Tat, which are expressed early after infection. Therefore, there is considerable interest in lentiviral vaccine candidates that can induce these types of immune responses. In the present study, we describe the construction and characterisation of expression vectors based on recombinant Semliki Forest virus system and modified vaccinia virus Ankara for the expression of feline immunodeficiency virus (FIV) accessory proteins Rev and OrfA. The Netherlands has enjoyed a relatively free state of vector-borne diseases of economic importance for more than one century. Emerging infectious diseases may change this situation, threatening the health of humans, domestic livestock and wildlife. In order to be prepared for the potential outbreak of vector-borne diseases, a study was undertaken to investigate the distribution and seasonal dynamics of candidate vectors of infectious diseases with emphasis on bluetongue vectors (Culicoides spp.). The study focused primarily on the relationship between characteristic ecosystems suitable for bluetongue vectors and climate, as well as on the phenology and population dynamics of these vectors.Twelve locations were selected, distributed over four distinct habitats: a wetland area, three riverine systems, four peat land areas and four livestock farms. Culicoides populations were sampled continuously using CO2-baited counterflow traps from July 2005 until August 2006, with an interruption from November 2005 to March 2006. All vectors were identified to species level. Meteorological and environmental data were collected at each location.Culicoides species were found in all four different habitat types studied. Wetland areas and peat bogs were rich in Culicoides spp. The taxonomic groups Culicoides obsoletus (Meigen) and Culicoides pulicaris (Linnaeus) were strongly associated with farms. Eighty-eight percent of all Culicoides consisted of the taxon C. obsoletus/Culicoides scoticus. On the livestock farms, 3% of Culicoides existed of the alleged bluetongue vector Culicoides dewulfi Goetghebuer. Culicoides impunctatus Goetghebuer was strongly associated with wetland and peat bog. Many Culicoides species were found until late in the phenological season and their activity was strongly associated with climate throughout the year. High annual variations in population dynamics were observed within the same study areas, which were probably caused by annual variations in environmental conditions.The study demonstrates that candidate vectors of bluetongue virus are present in natural and livestock-farm habitats in the Netherlands, distributed widely across the country. Under favourable climatic conditions, following virus introduction, bluetongue can spread among livestock (cattle, sheep and goats), depending on the nature of the viral serotype. The question now arises whether the virus can survive the winter conditions in north-western Europe and whether measures can be taken that effectively halt further spread of the disease. 相似文献

Effects of Al toxicity and interaction of Al and Mg on growth of twelve sorghum (Sorghum bicolor (L.) Moench) genotypes have been studied in nutrient solutions (pH 4.2). Aluminium at 30 μM decreased biomass (dry matter yield) of the individual genotypes by factors between 1.27 and 7.36, with identical sensitivity grouping of genotypes as obtained in an earlier pot experiment with an acid soil. Resembling acid-soil stress, Al toxicity was simultaneously expressed in two independent ways, i.e. impairment of root development and induced Mg deficiency. The effect of Al on total dry matter production of the genotypes was correlated more closely with changes in specific root length (m g−1 dry root) than with changes in internal Mg status. Increased Mg concentrations in the solutions (2.5 and 7.5 instead of 0.25 mM) not only decreased Al-induced Mg deficiency but also reduced the concentrations of Al in/on the roots and its damaging effect on root development. Therefore, the sorghum genotypes were less sensitive to Al at the higher Mg levels. At a high Mg concentration in the solution (7.5 mM) dry matter yield of two genotypes was even stimulated by Al. 相似文献

In vivo experiments were conducted to verify whether arabinoxylooligosaccharides (AXOS) obtained as low molecular mass compounds by enzymic hydrolysis from wheat bran arabinoxylan (AX) can exert nutritional effects. Two feeding trials were performed on chickens fed diets with either wheat or maize as the main component. Supplementation of bran AXOS at either 0.5% (w/w) to the wheat‐based diet or at 0.25% (w/w) to the maize‐based diet diets significantly (P < 0.05) improved the feed conversion rate without increasing the body weight of the animals, thus pointing to improved nutrient utilization efficiency. The positive effect of bran AXOS supplementation on feed utilization efficiency was similar to that obtained by adding an AX‐degrading xylanase directly to the wheat‐based diet. No significant effect on feed utilization efficiency was obtained with another type of nondigestible oligosaccharide such as fructooligosaccharides (FOS) derived from chicory roots. Bran AXOS significantly increased the level of bifidobacteria but not total bacteria in the caeca of the chickens, an effect not observed with either xylanase or FOS addition. These data suggest that bran AXOS have beneficial nutritional effects and may act as prebiotics. 相似文献

When assessing hypotheses, the possibility and consequences of false-positive conclusions should be considered along with the avoidance of false-negative ones. A recent assessment of the system of rice intensification (SRI) by McDonald et al. [McDonald, A.J., Hobbs, P.R., Riha, S.J., 2006. Does the system of rice intensification outperform conventional best management? A synopsis of the empirical record. Field Crops Res. 96, 31–36] provides a good example where this was not done as it was preoccupied with avoiding false-positives only. It concluded, based on a desk study using secondary data assembled selectively from diverse sources and with a 95% level of confidence, that ‘best management practices’ (BMPs) on average produce 11% higher rice yields than SRI methods, and that, therefore, SRI has little to offer beyond what is already known by scientists. 相似文献

The accuracy of a single sensor is often low because all proximal soil sensors respond to more than one soil property of interest. Sensor data fusion can potentially overcome this inability of a single sensor and can best extract useful and complementary information from multiple sensors or sources. In this study, a data fusion was performed of a Vis?CNIR spectrometer and an EM38 sensor for multiple soil properties. Stepwise multiple linear regression (SMLR), partial least squares regression (PLSR) and principal components analysis combined with stepwise multiple linear regression (PCA?+?SMLR) methods were used in three different fields. Soil properties investigated for data fusion included soil texture (clay, silt and sand), EC, pH, total organic carbon (TOC), total nitrogen (TN) and carbon to nitrogen ratio (CN). It was found that soil property models based on fusion methods significantly improved the accuracy of predictions of soil properties measureable by both sensors, such as clay, silt, sand, EC and pH from those based on either of the individual sensors. The accuracy of predictions of TOC, TN and CN was also improved in some cases, but was not consistent in all fields. Among data fusion methods, PLSR outperformed both SMLR and PCA?+?SMLR methods because it proved to have a better ability to deal with the multi-collinearity among the predictor variables of both sensors. The best data fusion results were found in a clayey field and the worst in a sandy field. It is concluded that sensor data fusion can enhance the quality of soil sensing in precision agriculture once a proper set of sensors has been selected for fusion to estimate desired soil properties. More efficient statistical data analysis methods are needed to handle a large volume of data effectively from multiple sensors for sensor data fusion. 相似文献
