This paper provides reviews of the most commonly used methods to detect plant pests belonging to groups of invasive organisms with high economic relevance, including Coleoptera (bark beetles, flathead borers, leaf beetles, longhorn beetles, weevils), Diptera (cone and seed flies, fruit flies), Homoptera (aphids, leafhoppers and psyllids, whiteflies), Lepidoptera (moths and butterflies), Thysanoptera (thrips), bacteria (potato brown rot Ralstonia solanacearum) and fungi (pitch canker disease Gibberella circinata, brown rot disease Monilinia fructicola). Future perspectives in detection methods are discussed, with particular reference to the considerable increase in the volume, commodity type and origins of trade in plant material from third countries, the introduction of new crops, the continuous expansion of the EU with new border countries being added, and the impact of climate change affecting the geographical boundaries of pests and their vectors. 相似文献

Summary Hybridization between radish and oilseed rape has been cumbersome, requiring elaborate embryo rescue techniques. With a modified flower culture method, we have achieved successful hybridization between radish and (transgenic) oilseed rape without the laborious and technically demandingin vitro ovule or embryo rescue techniques.The hybrid nature of the intergeneric hybrids was demonstrated using morphological traits, and DNA analyses. The described method will facilitate the generation ofRaphanobrassica hybrids useful for biosafety studies of the potential for transgenes to spread in weedyCruciferae as well as for breeding programs aimed at introducing useful radish genes, e.g. nematode resistance genes, into oilseed rape. 相似文献

Pedochronology is defined as the sequence of soil changes over time in a given landscape. Integrated stratigraphical and paleogeomorphic analysis of soil-landscape systems can offer information on landscape history that provides a rational basis for further pedological and soil-geomorphological studies. To connect these aspects of research, a new scheme, called chronogram, is introduced in which geogenic and pedogenic events can be summarized.Soil-landscape chronograms have time on one axis and depict erosion, deposition and soil formation across a given landscape along the other axis. Using open and enclosed hillslope systems as examples, the construction of chronograms and their relations to other summarizing schemes are demonstrated. These schemes include Butler's K-cycle system, Bos and Sevink's site history scheme, the catena, the toposequence, the Nine Unit Land-surface Model, and Huggett's simulation model. Opportunities for further pedochronological analysis, using paleosols, authigenic and allogenic pedorelicts, are identified. 相似文献

The high resistance of Brazil nut 2S albumin, previously identified as an allergen, against proteolysis by pepsin was examined in this work. Although the denaturation temperature of this protein exceeds the 110 degrees C at neutral pH, at low pH a fully reversible thermal denaturation was observed at approximately 82 degrees C. The poor digestibility of the protein by pepsin illustrates the tight globular packing. Chemical processing (i.e., subsequent reduction and alkylation of the protein) was used to destabilize the globular fold. Far-UV circular dichroism and infrared spectroscopy showed that the reduced and alkylated form had lost its beta-structures, whereas the alpha-helix content was conserved. The free energy of stabilization of the globular fold of the processed protein as assessed by a guanidine titration study was only 30-40% of that of the native form. Size exclusion chromatography indicated that the heavy chain lost its globular character once separated from the native 2S albumin. The consequences of these changes in structural stability for degradation by pepsin were analyzed using gel electrophoresis and mass spectrometry. Whereas native 2S albumin was digested slowly in 1 h, the reduced and alkylated protein was digested completely within 30 s. These results are discussed in view of the potential allergenicity of Brazil nut 2S albumin. 相似文献

This article presents a model to evaluate the accuracy of diagnostic tests. Data from three tests for the detection of EF-positive
Streptococcus suis serotype 2 strains in sows were analyzed. The data were collected in a field study in the absence of a gold standard, that
is, the true disease status (noninfected or infected) of the tested animals was unknown. Two tests were based on a polymerase
chain reaction (PCR); one test was applied to a tonsil swab (taken from the live animal), and the other test was applied to
the whole tonsil (collected at slaughter). The third test was based on a bacterial examination (BE) of the whole tonsil. To
reduce experimental cost BE was performed only for a subset of the animals in the sample. The model allows for dependence
between tests, conditional upon the unknown true disease status of the animals. Accuracy was expressed in terms of sensitivity
and specificity of the tests. A Bayesian analysis was performed that incorporated prior information about the accuracy of
the tests. The model parameters have a simple interpretation and specification of priors is straightforward. Posterior inference
was carried out with Markov chain Monte Carlo (MCMC) methods, employing the Gibbs sampler, as implemented in the WinBUGS program.
Different parameterizations to allow for selection and missing values, use of different priors, practical problems in the
analysis, and some interesting issues in a joint analysis of the binary (positive or negative) results of PCR and BE and two

Knowledge of the effects of species diversity within taxonomic groups on nutrient cycling is important for understanding the role of soil biota in sustainable agriculture. We hypothesized that earthworm species specifically affect nitrogen mineralization, characteristically for their ecological group classifications, and that earthworm species interactions would affect mineralization through competition and facilitation effects. A mesocosm experiment was conducted to investigate the effect of three earthworm species, representative of different ecological groups (epigeic: Lumbricus rubellus; endogeic: Aporrectodea caliginosa tuberculata; and anecic: Lumbricus terrestris), and their interactions on the bacterial community, and on nitrogen mineralization from 15N-labelled crop residue and from soil organic matter.Our results indicate that L. rubellus and L. terrestris enhanced mineralization of the applied crop residue whereas A. caliginosa had no effect. On the other hand, L. rubellus and A. caliginosa enhanced mineralization of the soil organic matter, whereas L. terrestris had no effect. The interactions between different earthworm species affected the bacterial community and the net mineralization of soil organic matter. The two-species interactions between L. rubellus and A. caliginosa, and L. rubellus and L. terrestris, resulted in reduced mineral N concentrations derived from soil organic matter, probably through increased immobilization in the bacterial biomass. In contrast, the interaction between A. caliginosa and L. terrestris resulted in increased bacterial growth rate and reduced total soil C. When all three species were combined, the interaction between A. caliginosa and L. terrestris was dominant. We conclude that the effects of earthworms on nitrogen mineralization depend on the ecological traits of the earthworm species present, and can be modified by species interactions. Knowledge of these effects can be made useful in the prevention of nutrient losses and increased soil fertility in agricultural systems, that typically have a low earthworm diversity. 相似文献

Intervertebral disc disease, as well as the associated alteration of the radiographic intervertebral disc space width, has been reported in horses. Disc height index (DHI) has proven to be an accurate and objective parameter in other species but data related to this parameter are lacking in horses. Therefore, the aims of this retrospective longitudinal diagnostic accuracy study were (a) to evaluate the reliability of measurements within and between observers of the equine Disc Width Index (EDWI) as a parameter for radiographic equine cervical intervertebral disc space width, and (b) to evaluate the sequential development of the EDWI over time. For this, EDWI from all intervertebral disc spaces between second cervical (C) to first thoracic (Th) vertebrae were obtained in a group of 39 Dutch Warmblood horses at 1, 5, and 18 months of age, by one European College of Veterinary Diagnostic Imaging (ECVDI) board‐certified veterinary radiologist (S.V.) and two veterinary students. Bland‐Altmann plots and intraclass Correlation Coefficient revealed a good intra‐ and interobserver agreement. A linear mixed‐effect model did reveal that mean EDWI increases significantly toward the caudal cervical spine, but did not differ significantly for a certain location over time or between sexes. Spearman's rank test did show a significant correlation between the vertebral alignment angle induced by different head‐neck positions and a normalized EDWI (ρ = 0.33, P < .0001). Student's t‐test revealed that the presence of C6‐C7 transposition of the transverse processes did not influence EDWI significantly. It was concluded that EDWI represents a reliable parameter for equine cervical radiographic intervertebral disc space width. Practical implementation of EDWI warrants monitoring in a group of adult horses while maintaining a standardized head‐neck position. 相似文献
