We hypothesized that replacing fish oil with 18:3n-3-rich linseed oil may enable salmon to maintain the levels of tissue n-3HUFA levels through a combination of increased desaturation activity and increased substrate fatty acid provision. To this end we investigated desaturation/elongation of [1-14C18:3n-3 in hepatocytes and intestinal enterocytes, and determined the extent to which 18:3n-3 was oxidized and desaturated by measuring both simultaneously in a combined assay. Salmon smolts were stocked randomly into five seawater pens and fed for 40 weeks on diets in which the fish oil was replaced in a graded manner by linseed oil. At the end of the trial, fatty acyl desaturation/elongation and oxidation activities were determined in isolated hepatocytes and intestinal enterocytes using [1-14C]18:3n-3 as substrate, and samples of liver and intestinal tissue were collected for analysis of lipid and fatty acid composition. The results showed that, despite increased desaturation of [1-14C]18:3n-3 in hepatocytes, provision of dietary 18:3n-3 did not prevent the decrease in tissue n-3HUFA in fish fed linseed oil. Intestinal enterocytes were a site of significant fatty acid desaturation but, in contrast to hepatocytes, the activity was not increased by feeding linseed oil and was generally lower in fish fed linseed oil compared to fish fed only fish oil. In contrast, oxidation of [1-14C]18:3n-3 in enterocytes was generally increased in fish fed linseed oil compared to fish fed the diet containing only fish oil. However, oxidation of [1-14C]18:3n-3 in hepatocytes was 4- to 8-fold lower than in enterocytes and was not affected by diet. Furthermore, oxidation of [1-14C]18:3n-3 in enterocytes exceeded desaturation irrespective of dietary treatment, whereas similar amounts of [1-14C]18:3n-3 were desaturated and oxidized in hepatocytes from fish fed only fish oil and desaturation exceeded oxidation by 3-fold in fish fed the diet containing 100% linseed oil. ABSTRACT: A challenge experiment was performed to investigate whether administration of Mycobacterium avium subsp. paratuberculosis (MAP) via the respiratory route leads to MAP infection in calves. Eighteen calves from test negative dams were randomly allocated to four groups. Six calves were challenged with MAP nasally and six calves were challenged by transtracheal injection; three orally challenged calves served as positive controls, and three non challenged calves as negative controls. The challenge was performed as a nine-fold trickle dose, 107 CFU in total. Blood and faecal samples were collected frequently. Calves were euthanized three months post-challenge and extensively sampled. Blood samples were tested for the presence of antibodies and interferon gamma producing cells by ELISA. Faecal and tissue samples were cultured in a liquid culture system and the presence of MAP was confirmed by IS900 realtime PCR. Fourteen out of fifteen calves had no MAP antibody response. The negative controls remained negative; all positive controls became infected. Two nasally challenged calves showed a Purified Protein Derivative Avian (PPDA) specific interferon gamma response. In all nasally challenged calves, MAP positive intestinal samples were detected. In three calves of the nasal group MAP positive retropharyngeal lymph nodes or tonsils were detected. In all calves of the transtracheal group MAP positive intestinal tissues were detected as well and three had a MAP positive tracheobronchial lymph node. These findings indicate that inhalation of MAP aerosols can result in infection. These experimental results may be relevant for transmission under field conditions since viable MAP has been detected in dust on commercial dairy farms.  相似文献

Two tospovirus isolates collected from tomato and bell pepper in Thailand were studied. The isolates induced severe necrotic mottling and/or necrotic spots and rings on the leaves and fruits of the respective plants as confirmed by back-inoculation. A polyclonal antiserum raised against its nucleocapsid (N) protein reacted only with an extract from plants infected with the homologous virus. Analysis of the nucleocapsid (N) gene sequence and its deduced amino acid sequence (Mw ∼31 kDa) showed 99% amino acid sequence homology with that of Tomato necrotic ring virus (TNRV). The nucleotide sequence of the 5΄ untranslated region and intergenic region flanking the N gene revealed typical features of the S RNA segment of tospoviruses. Mechanical inoculation of the virus on some plant species showed that most of the tested solanaceous species were susceptible to this virus. The biological, serological and molecular data presented here indicate that both isolates are identical to TNRV, a recently described tospovirus species in Thailand.  相似文献

Kelly et al. (Brevia, 20 July 2007, p. 337) questioned xenotransplant experiments supporting the cancer stem cell (CSC) hypothesis because they found a high frequency of leukemia-initiating cells (L-IC) in some transgenic mouse models. However, the CSC hypothesis depends on prospective purification of cells with tumor-initiating capacity, irrespective of frequency. Moreover, we found similar L-IC frequencies in genetically comparable leukemias using syngeneic or xenogeneic models.  相似文献

Our understanding of leukemia development and progression has been hampered by the lack of in vivo models in which disease is initiated from primary human hematopoietic cells. We showed that upon transplantation into immunodeficient mice, primitive human hematopoietic cells expressing a mixed-lineage leukemia (MLL) fusion gene generated myeloid or lymphoid acute leukemias, with features that recapitulated human diseases. Analysis of serially transplanted mice revealed that the disease is sustained by leukemia-initiating cells (L-ICs) that have evolved over time from a primitive cell type with a germline immunoglobulin heavy chain (IgH) gene configuration to a cell type containing rearranged IgH genes. The L-ICs retained both myeloid and lymphoid lineage potential and remained responsive to microenvironmental cues. The properties of these cells provide a biological basis for several clinical hallmarks of MLL leukemias.  相似文献

Cardiac arrhythmia was induced in mice by the injection of ouabain, lidocaine, and diphenylhydantoin. Temporal spreading of ventricular electrocardiographic complexes is shown consistently to follow injection of a cardioactive drug and to precede rhythm disturbances. A computerized monitoring technique based on temporal spreading and waveform distortion should find wide clinical and experimental application.  相似文献

Apparent motion was used to explore humans' ability to perceive the direction of motion in the visual field. A marked qualitative difference in this ability was found between short- and long-range motion. For short-range motion, the detection of the direction of motion is characterized by parallel operation over a wide visual field (that is, detection performance is independent of the number of objects in an array). When the positional displacement is large relative to an object's size, the direction of motion is detected in a serial manner. The process of detection is limited in this case by the ability to detect other events, such as appearance and disappearance of an object, and the ability to compute their spatio-temporal relations. The results are consistent with a previously suggested division of the motion detection system into short- and long-range processes. The direction of short-range motion can be perceived in parallel (preattentively), whereas long-range motion is attentive and requires more complicated computations. It seems that the detection of long-range motion is a conjunction task, combining the detection of disappearance and appearance.  相似文献
