A 13-year-old Bornean orangutan diagnosed with life threatening Streptococcus pyogenes broncho-pneumonia was kept in a state of deep sedation for 20 days via continuous intra-venous (IV) infusion of zolazepam -tiletamine and IV haloperidol to allow consistent IV administration of ceftazidime and gatifloxacine. The use of long-term deep sedation allowed carrying out a particularly demanding treatment not generally associated with zoological patients. The treatment was ultimately successful.  相似文献

Herbage production is regarded as having environment-friendly credentials. However, as the ruminant production it supports is facing major challenges on sustainability, environmental footprint and human health concerns, EU herbage cultivar testing must contribute to the solutions. Before new cultivars can be sold in a member state (MS) and gain EU-wide marketing, they must pass official tests to prove they are both novel (distinct, uniform and stable, DUS) with improved value for cultivation and use (VCU). Herbage species present specific challenges, as their allogamy imposes a wide within-cultivar variation that adds complexity to DUS tests and their “value” is only realized in ruminant produce. Current VCU systems measure production, chemical composition and disease/stress tolerances, often on large numbers of candidate cultivars, but prohibitive labour costs and logistics mean that animal intake, ruminant output or environmental benefits cannot be measured directly. Furthermore, some candidate cultivars with proven superior VCU fail DUS even though the non-distinct comparison is with a significantly lower performing registered cultivar. To resolve these problem cases, a “vmDUS” distinctness tool is proposed, which uses molecular markers but conforms to UPOV-declared principles. A short overview of current grassland research shows smart proxy measures of animal value can easily and quickly be adopted into an integrated pan-European (EU-VCU) test network. The proposed EU-VCU scheme will reallocate test resources to conduct these additional tests by placing MS in data sharing collaborations, while retaining their national listing authority. The benefits to all stakeholders from adopting these new testing procedures are considered.  相似文献

Hydrolysates obtained by autohydrolysis-posthydrolysis of corncobs were detoxified with charcoal, concentrated, supplemented with nutrients, and fermented with Debaryomyces hansenii. After biomass removal, the fermented media contained 0.1137 kg of nonvolatile components (NVC)/kg of liquor, which corresponded mainly to xylitol (0.6249 kg/kg of NVC) but also to minor amounts of inorganic components (measured as ashes), proteins, nonfermented sugars (xylose and arabinose), uronic acids, arabitol, and other nonvolatile components (ONVC). The media were subjected to further processing (sequential stages of adsorption, concentration, ethanol precipitation, concentration, and crystallization) to obtain food-grade xylitol. Adsorption experiments were carried out at various solid-to-liquor ratios. Under selected conditions (1 kg of charcoal/15 kg of liquors), the xylitol content increased to 0.6873 kg/kg of NVC, and almost total decoloration was achieved. The resulting liquor was concentrated by evaporation to increase its NVC content to 0.4032 kg/kg of liquor (corresponding to a xylitol concentration of 0.280 kg/kg of liquor), and ethanol was added to precipitate a part of the NVC (mainly proteins, but also uronic acids, ashes, and other nonvolatile compounds). Refined liquors (containing 0.7303 kg of xylitol/kg of NVC) were concentrated again, and ethanol was added (to reach 40-60% volume of the stream) to allow crystallization at -10 or -5 degrees C. Under selected conditions, 43.7% of xylitol contained in the initial fermentation broth was recovered in well-formed, homogeneous crystals, in which xylitol accounted for 98.9% of the total oven-dry weight. Material balances are presented for the whole processing scheme considered in this work.  相似文献

It is likely that in the near future sequence information from sequencing programmes and EST libraries will generate an abundance of genic microsatellite markers. This study is focused on the assessment of their likely impact and performance vis-à-vis their genomic counterparts. Microsatellites from two sources were used to assess the genetic diversity in 56 old and new varieties of bread wheat on the UK Recommended List. A set of 12 microsatellite markers generated from genomic libraries and 20 expressed sequence tag (EST)-derived microsatellites were used in the study, and the performance of both marker sets assessed. The EST-derived or genic microsatellites delivered fingerprints of superior quality, amplifying clear products with few stutter bands. Diversity levels as revealed bygenic microsatellites are similar to the few published results. The PIC values for the genic markers were generally lower than those calculated for the genomic microsatellites, though advantages of both marker classes for variety identification applications are discussed. The causation may be multifactorial, and the signs are often nonspecific. Purpose

Sediment transport and riverbed sedimentation were investigated in an alpine stream below a small hydropower reservoir desilted by a controlled sediment flushing (CSF) operation. The term “controlled” refers to the operational tasks implemented to mitigate the downstream environmental impact of the operation. The experimental dataset acquired before, during, and after the CSF was also used to carry out and calibrate a one-dimensional sediment transport model of the monitored event.

Materials and methods

The investigated reservoir is located in the central Italian Alps, and its original storage was 160,000 m3, about 30% filled by a mixture of sand and silt/clay before the CSF. Downstream sediment concentration was controlled by releasing clear water from upstream reservoirs and regulating the work of earth-moving equipment in the emptied reservoir. A 3.6-km-long reach with average slope of 0.015 was monitored: concentration and grain size of suspended sediment were measured during the CSF and the riverbed alteration was evaluated by volumetric sampling and measurements of the deposits’ thickness. Sedimentation and River Hydraulics—One Dimensional (SRH-1D) was used to simulate sediment transport during the monitored CSF. Model parameters were calibrated by comparing the computed and the observed amount of sediment deposited along the study reach.

Results and discussion

Sediment flushing was carried out in October 2010 for 3 days. Ca. 16,000 m3 of sediment were evacuated, representing approximately 30% silt/clay and 70% sand. 2.4 Mm3 of clear water was released to reduce sediment concentration and increase transport capacity downstream. About 3000 m3 of sand was deposited in the study reach after the CSF, with maximum height up to 0.2 m. Although the riverbed before the CSF was simply set as mono-granular, after calibrating the parameters, good agreement was achieved between the depositional pattern computed by SRH-1D and the one observed, both in terms of deposit thickness and grain size of deposited sediment. The sensitivity analysis revealed a major role of the parameters controlling bed mixing processes in affecting the simulated deposition after the CSF.

Conclusions

Sediment below 0.1 mm in diameter was not detected in river deposits after the flushing: the effects on river biota associated with substrate clogging by very fine sediment were therefore minimized. After proper calibration, 1-D sediment transport modeling can effectively support the planning of CSF operations: to minimize the downstream environmental effects, concurrently achieving acceptable flushing efficiency, the analyzed scenarios as well as the model outputs need to be carefully evaluated from a multidisciplinary perspective.
