In order to apply state-of-the-art molecular breeding techniques in fibre crop it is necessary to have a good knowledge of major polymer biosynthesis gene sequences and their expression pattern. Polymerase chain reaction was employed to isolate sequences of the major genes for lignin and cellulose biosynthesis in a kenaf cultivar. CeSA, 4cl, c4h, cad, and ccr gene primers were designed according to their conservative regions; partial sequences of lignin and cellulose biosynthesis genes were obtained. One actin II gene sequence was also isolated from the kenaf genome as a housekeeping gene to be employed in qPCR analysis. Expression levels of genes c4h, cad and CeSA in bark and core from plants harvested at three different growth stages were evaluated. Using qPCR analyses it was found that the expression levels of the two biosynthesis lignin genes in bark tissues increased during plant growth, while a negative trend was recorded in core tissues. In both bark and core, the quantity of lignin was positively correlated to plant growth while cellulose content decreased. 相似文献

Effect of K+ and Mg2+ in water on the performance of Litopenaeus vannamei using bioflocs was evaluated. Control had water with K:Mg (1:4.57) and other treatments (K:Mg of 1:5.4, 1:4.65, and 1:3.77), both with salinity of 4 g/L. Initial mean weight of shrimp was 0.08 ± 0.007 g. No difference in shrimp growth performance and survival was observed (P > 0.05) except in the control. Our results suggest that it is possible to culture postlarvae of Litopenaeus vannamei in low-salinity water with bioflocs when the water has an initial potassium concentration of 30.90 ± 8.5 mg/L and magnesium of 167.0 ± 3.9 mg/L. 相似文献

Paratrichodorus allius, a vector of the tobacco rattle virus which causes the corky ringspot disease (CRS) in potato, was identified in soil samples from Oregon and Washington. Two populations from potato fields in Pasco, WA and Umatilla, OR reproduced successfully on alfalfa, wheat, and corn which are commonly rotated with potato, and most weeds associated with this crop in the Pacific Northwest. Greenhouse studies showed thatP. allius was sensitive to low soil moisture, and increased 10 and 100 fold on Samsun NN tobacco in soil above field capacity compared to 1/2 and 1/3 field capacity, respectively. Population dynamic studies for 1996 and 1997 on a potato-wheat cropping sequence showed thatP. allius declined after the winter wheat cover crop was disked in the spring of 1996, and remained low on potato at the 0–90 cm soil profile, with no evidence of downward migration. In 1997,P. allius declined early in the season on wheat, and remained low until June–July when it peaked before declining again. In soil columns,P. allius placed 30 cm below a confined tobacco root system was able to reach and transmit tobacco rattle virus. ObjectiveTo evaluate the anti-nociceptive and sedative effects of slow intravenous (IV) injection of tramadol, romifidine, or a combination of both drugs in ponies.Study designWithin-subject blinded.AnimalsTwenty ponies (seven male, 13 female, weighing mean ± SD 268.0 ± 128 kg).MethodsOn separate occasions, each pony received one of the following three treatments IV; romifidine 50 μg kg− (R) tramadol 3 mg kg−1 given over 15 minutes (T) or tramadol 3 mg kg−1followed by romifidine 50 μg kg−1 (RT). Physiologic parameters and caecal borborygmi (CB) were measured and sedation and response to electrical stimulation of the coronary band assessed before and up to 120 minutes following drugs administration. Results were analyzed using the Friedman’s test and 2 way anova as relevant.ResultsWhen compared to baseline, heart (HR, beats minute−1) and respiratory rates (fR, breaths minute−1) increased with treatment T (highest mean ± SD, HR 43 ± 1; fR 33 ± 2) and decreased with R (lowest HR 29 ± 1 and fR 10 ± 4) and RT (lowest HR 32 ± 1 and fR 9 ± 3). There were no changes in other measured physiological variables. The height of head from the ground was lower following treatments R and TR than T. There was slight ataxia with all three treatments. No excitatory behavioural effects were observed. The response to electrical stimulation was reduced for a prolonged period relative to baseline following all three treatments, the effect being significantly greatest with treatment RT.ConclusionTramadol combined with romifidine at the stated doses proved an effective sedative and anti-nociceptive combination in ponies, with no unacceptable behavioural or physiologic side effects.Clinical relevanceSlow controlled administration of tramadol should reduce the occurrence of adverse behavioural side effects. 相似文献

Interspecific somatic hybrids were produced between tetraploidSolanum tuberosum and a nematode-resistant accession of the diploid speciesSolanum bulbocastanum by protoplast fusion. Hybrid cells were selected using dual fluorescent labeling of protoplast preparations prior to fusion. Hybridity of regenerated plants was confirmed with a combination of morphological assessment, chromosome counting and isozyme analysis. Deslorelin acetate is a GnRH agonist used for contraception in dogs. This study aimed to evaluate the treatment of pre‐pubertal female dogs with deslorelin acetate implants, to better investigate the primary stimulatory effect of the drug and the long‐term effects on the genital tract, throughout repeated treatments. Sicilian hound female dogs (24) were randomly assigned to treated group, control group 1 and control group 2. First group bitches were implanted at 4.5, 9.0 and 13.5 months and monitored clinically, ultrasonographically and endocrinologically, throughout the study period (13.5 months). Control group 1 bitches were not implanted and clinically monitored for the same period. At 18 months, the animals underwent ovariohysterectomy, thus allowing evaluation of the internal genitalia. Control group 2 bitches were ovariohysterectomized at the age of 4.5 months. The suppression of oestrus was obtained in the treated group despite the fact that the first implant caused a modest increase in plasmatic levels of 17‐beta estradiol and an evident cornification of the vaginal mucosa cells (50–80%). Estradiol and progesterone were at baseline levels for the remaining study period, in which no other oestrous manifestations were observed. The external genitalia maintained a juvenile appearance. The ovaries, ultrasonographically, showed no follicular structures and stayed the same size. At 18 months, the genital tract was still juvenile with inactive small ovaries and a thin filiform uterus. Deslorelin suppressed ovarian activity in pre‐pubertal bitches, and oestrous induction was not observed despite the presence of the primary stimulatory effect of the drug. The SCS-CN, Horton and USLE-family models are used worldwide, but few examples are available for olive groves on steep and clayey soils under mulching of pruning residues. We evaluate the accuracy in predicting runoff and soil loss of a steep (20%) and clayey olive grove subject to three soil conservation practices: mechanical tillage (MT), mulching with pruning residues (NTR) and standard protection (SP), measured at plot scale in Southern Italy during 30 months under natural rainfall. The models were calibrated by adjusting the Curve Numbers (for the SCS-CN model) and the C-factors (MUSLE and USLE-M); the Horton model was not calibrated. The model performance was assessed by qualitative and quantitative procedures. In all practices, the SCS-CN was more accurate for runoff predictions (mean difference of 7% compared with corresponding observations) compared with the Horton (mean difference of 30%). For soil erosion, the MUSLE showed better performance in soils subject to MT or total protection (differences lower than 10%), while the USLE-M was more reliable to simulate soil loss in mulched plots (differences of 8%). A set of Curve Numbers (95 for MT, 70 for SP and 85 for NTR) and C-factors (0.4 for MT, 0.2 for SP and 0.1 for NTR) are proposed for applications in steep slope and clayey soil olive orchards using the SCS-CN and USLE proposed models, respectively. Validation exercises in other environmental experimental conditions would enhance the export these models for runoff and erosion control in agricultural soils treated with mulching. 相似文献

In this study, flow-through respirometry was used to test the effect of acute temperature change on resting routine metabolic
rates of two benthic elasmobranchs, Atlantic stingrays, Dasyatis sabina (n = 7) and whitespotted bamboo sharks, Chiloscyllium plagiosum (n = 7) kept under fluctuating temperature regime of 24–27 and 23–25°C, respectively. Atlantic stingrays and whitespotted bamboo
sharks showed a temperature sensitivity (Q10) of 2.10 (21–31°C) and 2.08 (20–28°C), respectively. Not surprisingly, oxygen consumption (MO2) increased in both species as temperature was raised. Acute increases in oxygen uptake may be useful during activities such
as foraging, and some elasmobranchs may alter physiological processes by taking advantage of thermal variability in the environment.
However, further investigation of different physiological processes is needed to better understand how temperature variation
