Context

Quantifying gene flow in natural populations is a key topic in both evolutionary and conservation biology. Understanding the extent to which the landscape matrix facilitates or impedes gene flow is becoming a high priority in a context of worldwide habitat loss and fragmentation.

Objectives

Unexpectedly, a lower genetic diversity and a higher genetic structure have been previously observed in the less fragmented and the most forested habitat across four pine marten (Martes martes) populations in France. Our aim was to quantify the effect of landscape on the spatial distribution of genetic diversity in two populations in contrasting habitats.

Methods

We conducted an individual-based landscape genetics analysis in a highly fragmented rural plain (Bresse, n = 126) and in a highly forested (50 %) mountainous area (Ariège, n = 88) in France. We tested for isolation-by-resistance using least-cost distances and used a causal modeling approach on 16,384 landscape and 104 elevation resistance scenarios.

Results

Landscape structure influenced the genetic differentiation in Bresse, with vegetation providing more genetic connectivity over the study area than open areas, while roads and human buildings showed unexpected low resistance to gene flow. In Ariège, genetic differentiation was mainly associated with changes in elevation, with an optimal elevation for gene flow of around 1700 m, likely associated with changes in vegetation structure.

Conclusions

The pine marten seems to be able to cope with human-dominated landscapes and with fragmented forest landscapes, whereas elevation is the major driver of genetic differentiation in our mountainous landscape. Additionally, we highlight the importance of spatial replication in landscape genetics for deriving reliable conservation and management measures over the species distribution.

OBJECTIVE: To evaluate the effects of three anaesthetic combinations in adult European badgers (Meles meles). STUDY DESIGN: Prospective, randomized, blinded, experimental trial. ANIMALS: Sixteen captive adult badgers. METHODS: The badgers were each anaesthetized by intramuscular injection using the three techniques assigned in random order: romifidine 0.18 mg kg(-1), ketamine 10 mg kg(-1) and butorphanol 0.1 mg kg(-1) (RKB); medetomidine 0.1 mg kg(-1), ketamine 9 mg kg(-1) and butorphanol 0.1 mg kg(-1) (MKB); and medetomidine 0.1 mg kg(-1) and ketamine 10 mg kg(-1) (MK). Initial drug doses were calculated based on a body mass of 10 kg. Additional anaesthetic requirements, time to drug effect, duration of action and recovery from anaesthesia were recorded. Heart rate and rhythm, respiratory rate and rhythm, rectal and subcutaneous microchip temperature and oxygen saturation were recorded every 5 minutes. Depth of anaesthesia was assessed using: muscle tone; palpebral and pedal reflexes; and tongue relaxation at these time points. Blood samples and a tracheal aspirate were obtained under anaesthesia. Atipamezole was administered if the badger had not recovered within 60 minutes Parametric data were analysed using anova for repeated measures, and nonparametric data using Friedman's, and Cochran's Q tests: p < 0.05 was considered significant. RESULTS: All combinations produced good or excellent muscle relaxation throughout the anaesthetic period. RKB had the shortest duration of anaesthesia (16.8 minutes compared with MKB 25.9 minutes and MK 25.5 minutes) and antagonism was not required. RKB depressed respiratory rate less than MK and MKB. There was no significant difference between techniques for heart rate and rhythm. CONCLUSIONS AND CLINICAL RELEVANCE: All combinations provided anaesthetic conditions suitable for sampling and identification procedures in adult badgers. The RKB protocol provided a significantly shorter period of anaesthesia when compared with the combinations containing medetomidine.  相似文献

Diversity and structure of populations may differ substantially between morphological traits and molecular markers. Explanations of such discrepancies are crucial for further progress in breeding as well as for the maintenance of genetic resources. Our objective was to compare indices of among-cultivars differentiation for morphological traits (QST) and molecular markers (FST) in alfalfa (Medicago sativa), a legume forage species. Ten cultivars representing the Northern and Southern types commonly grown in Europe were investigated. For each cultivar, 40 genotypes were analysed with 16 SSR markers and four morphological traits measured in two locations and several cuts. QST values were in general high (0.02–0.39) compared to the differentiation observed with molecular markers (FST = 0.01), especially for growth habit, indicating that morphological traits were more efficient to structure the diversity than molecular markers. For morphological traits, a clear separation of Northern and Southern cultivars was observed, whereas for molecular markers, no clear structure was detected. Nevertheless, the grazing type cultivar Luzelle was separated from the rest of the cultivars for both morphological traits and molecular markers. Although pairwise differences between cultivars were significant for both morphological traits and molecular markers, the main part of the variation was found within cultivars. This large within-cultivar variation may be explained, besides the outcrossing reproductive mode and autotetraploid genetics, by the recent history of M. sativa domestication in Europe and the frequent seed exchanges. Selection for morphological traits (QST > FST) was achieved without modification of within-cultivar neutral diversity.  相似文献

The maintenance of biodiversity in agro-ecosystems is often viewed as an insurance against an uncertain future, yet, there is increasing evidence of biodiversity loss in agricultural landscapes. Here, we examined long term trends in the arable weed flora of North-East France by revisiting 158 arable fields initially surveyed in the 1970s. We assessed changes in species richness, density and frequency of occurrence in weed communities as well as for subgroups that are recognised for their conservation value. We also evaluated the importance of crop edges as potential refugia for the threatened arable weeds. Among the 121 species recorded in both surveys, 40% had significantly declined in frequency while 10% had significantly increased. At the field level, we recorded a 42% decline in species richness and a 67% decline in species density. Trends were comparable for weed species of particular conservation value. In the 2000s survey, crop edges harboured levels of weed diversity that were intermediate between those found in field cores in the 1970s and in the 2000s survey. Our results indicated that many species of conservation value had disappeared or seriously decreased in the field core but still persisted in the crop edge. The consequences of this general decline and possible conservation options are discussed in the light of the results.  相似文献

Increased competition between agri-food supply chains has strained relationships between farmers and processing factories while reducing individual profit margins. Decisions at different levels of the supply chain can no longer be considered independently, since they may influence profitability throughout the supply chain. This paper presents a decision support approach based on the MAGI® simulation tool, which aims to facilitate discussion and negotiation between stakeholders while collectively exploring satisfactory solutions.The simulation tool helps sugarcane growers and millers in designing and assessing new ways of organizing cane supply management within a mill area. It addresses key issues such as restructuring mill areas or changing cane delivery allocation rules in order to increase total sugar production and total net revenue at the mill area level. This approach has been implemented for two mills in Réunion and one mill in South Africa. Simulations showed that sugar gains may be obtained by rearranging supply scheduling according to quality-based zoning within a mill area. Discussions led to further studies regarding the practicality of the best scenarios. MAGI® is now available as freeware for testing in different settings.  相似文献

ABSTRACT The asexual phase of the life cycle of Plasmopara halstedii, the causal agent of downy mildew of sunflower, plays a key role in the propagation of the disease. We investigated the morphological and ultrastructural changes that occur during the asexual development of the pathogen. Direct examination of infected cotyledons confirmed the presence of sporangiophores. In contact with water, important ultrastructural changes occurred, affecting the surface of zoosporangia, which became smoother, and their cytoplasm, which differentiated into flagellate zoospores. The subsequent encystment of zoospores was characterized by the synthesis of a cell wall and the loss of the flagella. In addition, two monoclonal antibodies (MAbs) specific for P. halstedii were used to analyze the immunochemical changes associated with these modifications. MAb 16A6, which bound to a 48-kDa glycoprotein, mainly labeled the surface of mobile or encysted zoospores and of mother cells of germ tubes. Conversely, MAb 2F9, which recognized highly glycosylated antigens, labeled the surface of zoosporangia and of flagellate zoospores, but not the encysted zoospores. These results provide new insights into the morphological and ultrastructural changes associated with the release and the encystment of zoospores which may be interesting targets for the development of new antimicrobial products.  相似文献

Over the last 30 years, many studies have surveyed weed vegetation on arable land. The ‘Arable Weeds and Management in Europe’ (AWME) database is a collection of 36 of these surveys and the associated management data. Here, we review the challenges associated with combining disparate datasets and explore some of the opportunities for future research that present themselves thanks to the AWME database. We present three case studies repeating previously published national scale analyses with data from a larger spatial extent. The case studies, originally done in France, Germany and the UK, explore various aspects of weed ecology (community composition, management and environmental effects and within-field distributions) and use a range of statistical techniques (canonical correspondence analysis, redundancy analysis and generalised linear mixed models) to demonstrate the utility and versatility of the AWME database. We demonstrate that (i) the standardisation of abundance data to a common measure, before the analysis of the combined dataset, has little impact on the outcome of the analyses, (ii) the increased extent of environmental or management gradients allows for greater confidence in conclusions and (iii) the main conclusions of analyses done at different spatial scales remain consistent. These case studies demonstrate the utility of a Europe-wide weed survey database, for clarifying or extending results obtained from studies at smaller scales. This Europe-wide data collection offers many more opportunities for analysis that could not be addressed in smaller datasets; including questions about the effects of climate change, macro-ecological and biogeographical issues related to weed diversity as well as the dominance or rarity of specific weeds in Europe.  相似文献
