ABSTRACT: Over the last 10 years or so, infections caused by bacteria belonging to a particular branch of the genus Francisella have become increasingly recognised in farmed fish and molluscs worldwide. While the increasing incidence of diagnoses may in part be due to the development and widespread availability of molecular detection techniques, the domestication of new organisms has undoubtedly instigated emergence of clinical disease in some species. Francisellosis in fish develops in a similar fashion independent of host species and is commonly characterised by the presence of multi-organ granuloma and high morbidity, with varying associated mortality levels. A number of fish species are affected including Atlantic cod, Gadus morhua; tilapia, Oreochromis sp.; Atlantic salmon, Salmo salar; hybrid striped bass, Morone chrysops × M. saxatilis and three-lined grunt, Parapristipoma trilinineatum. The disease is highly infectious and often prevalent in affected stocks. Most, if not all strains isolated from teleost fish belong to either F. noatunensis subsp. orientalis in warm water fish species or Francisella noatunensis subsp. noatunensis in coldwater fish species. The disease is quite readily diagnosed following histological examination and identification of the aetiological bacterium by culture on cysteine rich media or PCR. The available evidence may indicate a degree of host specificity for the various Francisella strains, although this area requires further study. No effective vaccine is currently available. Investigation of the virulence mechanisms and host response shows similarity to those known from Francisella tularensis infection in mammals. A standard method for assaying protein in red wine is currently lacking. The method described here is based on protein precipitation followed by dye binding quantification. Improvements over existing approaches include minimal sample processing prior to protein precipitation with cold trichloroacetic acid/acetone and quantification based on absorbance relative to a commercially available standard representative of proteins likely to be found in wine, the yeast mannoprotein invertase. The precipitation method shortened preparation time relative to currently published methods and the mannoprotein standard yielded values comparable to those obtained by micro-Kjeldahl analysis. The assay was used to measure protein in 48 Pinot noir wines from 6 to 32 years old. The protein content of these wines was found to range from 50 to 102 mg/L with a mean value of 70 mg/L. The availability of a simple and relatively rapid procedure for assaying protein provides a practical tool to quantify a wine component that has been overlooked in routine analyses of red wines. 相似文献

Through pyric herbivory (i.e., fire-induced grazing patterns), native grasslands were historically a spatially heterogeneous environment. It is hypothesized that the mosaic of habitats created by pyric herbivory supports a more diverse invertebrate community compared to modern range management. Patch-burn management, a pyric herbivory technique, is an application of prescribed fire and grazing whereby the timing and location of the burned and grazed patches is varied, creating a diversity of habitat conditions. Although disturbance in sandsage (Artemisia filifolium Torr.) prairie historically included fire and grazing, fire disturbance has been nearly eradicated from this ecosystem in western Oklahoma. We compared patch-burn management to traditional management (i.e., moderate grazing with no fire) in sandsage prairie to evaluate the impact of these two management regimes on aboveground invertebrates. We sampled invertebrates at 44 points in each of 3 mo (May, June, and July) with the use of Dietrick vacuum sampling. Diversity, total abundance, and abundance of seven invertebrate orders (Araneae, Diptera, Hemiptera, Homoptera, Hymenoptera, Lepidoptera, and Orthoptera) were similar between patch-burn and traditional pastures. When examined on a patch level, five invertebrate orders were also similar between their respective time since burn patch and the traditional patches. Araneae and Coleoptera abundance were higher in traditional patches, and Hemiptera abundance was higher in current-year burn patches. Our results suggest a heterogeneity-based management scheme based on pyric herbivory does not negatively impact the overall invertebrate community and may benefit a wider variety of invertebrates by providing areas of varying levels of disturbance. In comparison, homogeneous landscapes such as those created by traditional management may only benefit segments of the invertebrate community that have habitat associations with moderately disturbed or undisturbed areas. Therefore, a disturbance regime involving the interaction of fire and grazing may be valuable for maintaining biodiversity and productivity within sandsage prairie ecosystems. 相似文献

Achieving economically optimum livestock production on rangelands can conflict with conservation strategies that require lower stocking rate to maintain wildlife habitat. Combining the spatial and temporal interaction of fire and grazing (pyric–herbivory) is a conservation-based approach to management that increases rangeland biodiversity by creating heterogeneous vegetation structure and composition. However, livestock production under pyric–herbivory has not been reported. In both mixed-grass prairie and tallgrass prairie, we compared livestock production in pastures with traditional fire and grazing management (continuous grazing, with periodic fire on tallgrass prairie and without fire on mixed-grass prairie) and conservation-based management (pyric–herbivory applied through patch burning) at a moderate stocking rate. Stocker cattle weight gain, calf weight gain, and cow body condition score did not differ (P > 0.05) between traditional and conservation-based management at the tallgrass prairie site for the duration of the 8-yr study. At the mixed-grass prairie site, stocker cattle gain did not differ in the first 4 yr, but stocker cattle gained more (P ≤ 0.05) on conservation-based management and remained 27% greater for the duration of the 11-yr study. Moreover, variation among years in cattle performance was less on pastures under conservation management. Traditional management in mixed-grass prairie did not include fire, the process that likely was associated with increased stocker cattle performance under conservation management. We conclude that pyric–herbivory is a conservation-based rangeland management strategy that returns fire to the landscape without reduced stocking rate, deferment, or rest. 相似文献

Pedigree information was traditionally used to assess inbreeding. The availability of high-density marker panels provides an alternative to assess inbreeding, particularly in the presence of incomplete and error-prone pedigrees. Assessment of autozygosity across chromosomal segments using runs of homozygosity (ROH) has emerged as a valuable tool to estimate inbreeding due to its general flexibility and ability to quantify the chromosomal contribution to genome-wide inbreeding. Unfortunately, the identification of ROH segments is sensitive to the parameters used during the search process. These parameters are heuristically set, leading to significant variation in the results. The minimum length required to identify an ROH segment has major effects on the estimation of inbreeding and inbreeding depression, yet it is arbitrarily set. To overcome this limitation, a search algorithm to approximate mutation enrichment was developed to determine the minimum length of ROH segments. It consists of finding genome segments with significant effect differences in trait means between animals with high and low burdens of autozygous intervals with a specific length. The minimum length could be determined heuristically as the smallest interval at which a significant signal is detected. The proposed method was tested in an inbred Hereford cattle population genotyped for 30,220 SNPs. Phenotypes recorded for six traits were used for the approximation of mutation loads. The estimated minimum length was around 1 Mb for yearling weight (YW) and average daily gain (ADG) and 4 Mb for birth weight and weaning weight. These trait-specific thresholds estimated using the proposed method could be attributed to a trait-dependent effect of homozygosity. The detection of significant inbreeding effects was well aligned with the estimated thresholds, especially for YW and ADG. Although highly deleterious alleles are expected to be more frequent in recent inbreeding (long ROH), short ROH segments (<5 Mb) could contain a large number of less deleterious mutations with substantial joint effects on some traits (YW and ADG). Our results highlight the importance of accurate estimation of the ROH-based inbreeding and the necessity to consider a trait-specific minimum length threshold for the identification of ROH segments in inbreeding depression analyses. The objectives of this study were to compare different models for analysing body weight (BW) and average daily feed intake (ADFI) data collected during a 70-day feedlot test period and to explore whether genetic parameters change over time to evaluate the implications of selection response. (Co)variance components were estimated using repeatability and random regression models in 2,071 Angus steers. Models included fixed effects of contemporary group, defined as herd–year–observation_date–age, with additive genetic and permanent environmental components as random effects. Models were assessed based on the log likelihood, Akaike's information criterion and the Bayesian information criterion. For both traits, random regression models (RRMs) presented a better fit, indicating that genetic parameters change over the test period. Using a two-trait RRM, the heritability from day 1 up to day 70 for BW increased from 0.40 to 0.50, while for ADFI, it decreased from 0.44 to 0.33. The genetic correlation increased from 0.53 at day 1 up to 0.79 at day 70. Selection based on an index assuming no change in genetic parameters would yield a 2.78%–3.13% lower selection response compared to an index using parameters estimated with RRMs and assuming these genetic parameters are correct. Results imply that it may be beneficial to implement RRMs to account for the change of parameters across the feedlot period in feed efficiency traits. 相似文献

Canine intracranial meningiomas can be graded based on histological classification as benign (grade I), atypical (grade II), and anaplastic or malignant (grade III). In people, grade II/III meningiomas behave more aggressively, have a higher potential for recurrence after surgical resection, and have lower apparent diffusion coefficient (ADC) values with diffusion weighted imaging (DWI). In this retrospective analytical cross‐sectional study, 42 dogs had ADC values quantified in an attempt to differentiate tumor histologic grade. Our hypothesis was that ADC values would be significantly lower in grade II and III versus grade I meningiomas in dogs. On each ADC image, a polygonal region of interest (ROI) was hand‐drawn along the lesion's periphery, excluding fluid‐filled and hemorrhagic regions. Mean ADC value (ADCmean) and minimum ADC value (ADCmin) were calculated. Additionally, two smaller, ovoid ROI were drawn within the lesion with mean ADC calculated (ADCmean sR and ADCmin sR). Normalized ADC values using white matter were also calculated (ADCn and ADCn sR). Grades of each tumor were assigned based on histopathology review. Association between ADC parameters and histological grade was tested by means of two‐sample t‐tests. There were 14 grade I (33.3%), 25 grade II (59.5%), and three grade III (7.2%) meningiomas. ADCmean sR and ADCmin sR were significantly lower when comparing grade II/III to grade I (P < .05). Grade II tumors had significantly lower ADCmean, ADCmean sR, ADCmin sR, ADCn, and ADCn sR than grade I meningiomas. Erwinia psidii is a gram-negative bacterium that threatens both guava and eucalypt plantations in several countries. Despite the economic importance of both crops, nothing is currently known about the molecular mechanisms underlying E. psidii pathogenicity and, consequently, how it evolved to infect Eucalyptus species besides its presumed native host Psidium guajava. In this study, we predicted putative type III secretion system effectors that may play important roles during plant–E. psidii interactions and conducted effector structure and phylogenetic analyses to gain important insights into their function and evolution. For that, the whole genomes of four E. psidii strains that exhibit differential aggressiveness towards eucalypt clones were sequenced and their effector repertoires predicted based on sequence identity with known effectors of the model phytopathogen Erwinia amylovora. Only proteins sharing significant sequence identity with the DspE and Eop1 effectors were found. Here, it is shown that these two E. psidii effectors retain all structural characteristics of their corresponding protein superfamilies, but exhibit allelic variations that are consistent with the observed aggressiveness differences between strains. Phylogenetic analyses revealed that whereas E. psidii housekeeping gene sequences are more closely related to those from Erwinia tracheiphila, the effector (either nucleotide or amino acid) sequences are more closely related to their Pantoea agglomerans counterparts, suggesting that dspE and eop1 were both acquired through horizontal gene transfer from the latter bacterial species. The results of this study provide important insights on E. psidii pathogenicity and set the stage for future effector functional studies. 相似文献
