Interleukin‐10 (IL‐10) is a multifunctional cytokine and plays an important role in diseases. In this study, IL‐10 gene was cloned and characterized from catla (Catla catla), which is a highly commercially important fish species in the Indian subcontinent. The result indicated that the full‐length catla IL‐10 (cIL‐10) gene had five exons and four introns with an open reading frame of 540 nucleotides encoding a polypeptide of 179 amino acids. A phylogenetic analysis of cIL‐10 gene sequence showed that cIL‐10 clustered with freshwater carps group as expected. Quantitative real‐time polymerase chain reaction analysis showed that cIL‐10 was expressed in gill, liver, kidney, intestine, skin and heart and its expression profile was up‐regulated in bacterial infection and LPS treatment. A close relationship of high cIL‐10 expression and low pro‐inflammatory cytokine IL‐1β expression was observed in the treated group of fish, which might reveal the role of cIL‐10 as an anti‐inflammatory cytokine. Mechanism of cIL‐10 induction was investigated by blocking nuclear factor (NF)‐κB ‐signalling with BAY 11‐7082 in catla kidney cell culture. Blocking NF‐κB suppressed IL‐10 induction by LPS, and thus it revealed that cIL‐10 was induced through NF‐κB signalling. These data could be helpful to understand the function of IL‐10 in fish in response to vaccinations, probiotics and various diseases.  相似文献

Potato leaves infected with Phytophthora infestans produced a serine protease inhibitor (PLPKI) with specificity for microbial proteases. Sequencing of the first twenty residues at the NH2‐terminus of the mature PLPKI polypeptide demonstrated that PLPKI is a novel member of the potato protease inhibitor I family. PLPKI inhibited the activity of extracellular proteases produced by two pathogens of potato, P. infestans and Rhizoctonia solani, but was inactive against proteases secreted into the culture media by the binucleate Rhizoctonia N2, a non‐pathogenic fungus for potato. Western blot analysis showed a positive correlation between the levels of PLPKI and the degree of horizontal resistance, showing its highest accumulation in clone OKA 5632.11, which has been described as highly resistant. This correlation, together with the ability of PLPKI to completely abolish the secreted serine protease activity of P. infestans, suggests that PLPKI may have an active role in protecting potato plants from this pathogenic oomycete and that it could be used as a suitable biochemical marker to help breeders in the selection of cultivars with high degree of horizontal resistance.  相似文献

From early times, jute fibre has been generally conditioned for easy spinning by adding oil and water in the form of an emulsion. The commonly used oil consists of C12–C31 fractions of mineral oil that sometimes impart different intensities of oily (kerosene) or fishy smell to the end product. In the present work, efforts have been made to find a suitable sustainable substitute of mineral oil based conditioning agent for spinning of jute yarn and for this, three types of vegetable oil (rice bran oil, palmolein oil and castor oil), a silicone emulsion, a mixed enzyme system and glycerine have been used separately or in combinations as conditioning agents for jute fibre before its mechanical processing for making yarn in jute spinning machines. Considering comparable mechanical process performance for spinning of jute fibre (viz., fibre loss as droppings during processing, moisture retention prior to spinning stage and spinning end breakage rate), tensile properties of yarn, and lower yarn hairiness, it may be suggested to use 2.5% castor oil alone, or 2% castor oil in combination with 0.1–0.5% glycerine in the form of oil-in-water emulsion as the most suitable alternatives to conventional mineral oil-based jute conditioning agent to spin ordinary jute yarn.  相似文献

Guggal (Commiphora wightii (Arnott) Bhandari comb. nov.) is a small tree which is tapped for medicinally important oleo?Cgum?Cresin. Naturally infected plant oozes oleo?Cgum?Cresin from its trunk and primary branches. However, in either case, the plant dies slowly after oozing. A bacterium was established to be responsible for these phenomena. Four isolates of this bacterium were characterised by biochemical tests, Biolog GN2 microplate reaction, rDNA sequencing, which suggested that the pathogen belonged to the genus Xanthomonas. However, phylogenetic analysis based on chaperone protein (dnaK) gene, TonB?Cdependent receptor (fyuA) gene, DNA gyrase B (gyrB) gene and RNA polymerase sigma factor (rpoD) gene sequences placed it as a member of X. axonopodis 9.2 rep?CPCR/DNA?CDNA homology cluster close to X. perforans, X. alfalfae and X. euvesicatoria. Further elucidation of phylogenetic position of the test strains was achieved from a gyrB based tree considering sequences from 71 representative strains. Test strains were confirmed to be members of X. axonopodis. These had very narrow infectivity limited to Commiphora spp. Hence, we propose a novel pathovar, X. axonopodis pv. commiphoreae pv. nov. as the cause of gum oozing in guggal. Pathotype is DXA 01 = CFBP 7580 = LMG 26789.  相似文献

Qualitative/quantitative phytochemical variations were observed in dried flowering tops of cultivated Hypericum perforatum L. cv. Zorzi infected by phytoplasmas of the "ash yellows" class, identified by direct and nested polymerase chain reaction (PCR); this is the first report of ribosomial group 16SrVII phytoplasmas in St. John's Wort. Methanolic extracts of healthy and infected plants were separated by reversed phase high-performance liquid chromatography to quantify naphthodianthrones and flavonoids, while essential oils were analyzed by means of gas chromatography (GC)-GC/MS. The affected plants exhibited decreased amounts of rutin (1.96 +/- 0.23 vs 4.96 +/- 0.02 mg/g), hyperoside (2.38 +/- 0.21 vs 3.04 +/- 0.05 mg/g), isoquercitrin (1.47 +/- 0.04 vs 3.50 +/- 0.08 mg/g), amentoflavone (0.12 +/- 0.01 vs 0.39 +/- 0.02 mg/g), and pseudohypericin (1.41 +/- 0.23 vs 2.29 +/- 0.07 mg/g), whereas the chlorogenic acid content was doubled (1.56 +/- 0.11 vs 0.77 +/- 0.02 mg/g). Hypericin, quercitrin, and quercetin contents were not severely affected. The essential oil yield was drastically reduced in infected material (0.11 vs 0.75% in healthy material) and revealed an increased abundance of sesquiterpenes (beta-caryophyllene, delta-elemene, and germacrene D, in particular) and a matching decrease in monoterpene hydrocarbons and aliphatics. The consequences that the phytopathological condition of cultivated H. perforatum plants has on the commercial quality, market value, and therapeutic efficacy are outlined.  相似文献

The aim of the study was to attempt the detection of West Nile virus (WNV) in wild birds in Poland. Forty-eight species of 1912 wild birds were used for the investigations. The birds were derived from various locations in Poland from early spring till late autumn of the years 2009-2011. The brain samples were homogenised and cellular RNA was isolated. Two methods (RT-PCR and nested RT-PCR) were used. The presence of WNV RNA was not detected in the samples examined. Additionally, a short analysis of the epizootiological situation regarding the presence of WNV in Poland is presented.  相似文献

A total of 107 faecal samples were collected from diarrhoeic lambs of high altitude terrains (2,000 to 5,000 m above the mean sea level) of Tawang and West Kameng districts of Arunachal Pradesh, India. Total 234 Escherichia coli were isolated and further subjected to PCR for the study of virulence repertoire characteristics of Shiga toxin-producing E. coli (STEC) and enterotoxigenic E. coli (ETEC). Out of the 234 isolated E. coli, 32% were found positive for STEC, and 9% were carrying virulence gene for ETEC. The isolated STEC serogroups were O159, O127, O120, O113, O60, O30, O25, O8 and O2. Of all the 74 STEC strains, PCR showed that 18% isolates carried stx 1 , 26% possessed stx 2 and 47% produced positive amplicon for both. Other virulent attributes like intimin (eaeA), enterohaemolysin (ehxA) and STEC auto-agglutinating adhesin (saa) were present in 18%, 43% and 44% of the isolates, respectively. The isolated ETEC serogroups were O172, O170, O159, O146, O127, O120, O113, O86, O75, O60, O30, O25, O8, O2, OR and OUT. Of the 22 ETEC-positive isolates, 23%, 18% and 4.5% possessed the gene only for LT, STa and STb, respectively, whereas 54% carried genes for both LT and STb. Some serogroups of E. coli like O159, O127, O120, O113, O60, O30, O25, O8 and O2 possessed genes for both Shiga toxin and enterotoxin. This study is the first report of ETEC isolation from diarrhoeic lambs in India. The moderately high proportion of STEC and ETEC in the diarrhoeic lambs implicated that these animals are important reservoir of STEC and ETEC. This is really a grave concern for the ‘brokpas’ and nomads (shepherds) who share a close relationship with this animals for their livelihood. This study also indicates that ETEC may be a major cause for frequent diarrhoeal episodes in lambs of this region.  相似文献

Deep brain stimulation (DBS) of the subthalamic nucleus markedly improves the motor symptoms of Parkinson's disease, but causes cognitive side effects such as impulsivity. We showed that DBS selectively interferes with the normal ability to slow down when faced with decision conflict. While on DBS, patients actually sped up their decisions under high-conflict conditions. This form of impulsivity was not affected by dopaminergic medication status. Instead, medication impaired patients' ability to learn from negative decision outcomes. These findings implicate independent mechanisms leading to impulsivity in treated Parkinson's patients and were predicted by a single neurocomputational model of the basal ganglia.  相似文献
