Calcium ion (Ca2+) influx through voltage-gated Ca2+ channels is important for the regulation of vascular tone. Activation of L-type Ca2+ channels initiates muscle contraction; however, the role of T-type Ca2+ channels (T-channels) is not clear. We show that mice deficient in the alpha1H T-type Ca2+ channel (alpha(1)3.2-null) have constitutively constricted coronary arterioles and focal myocardial fibrosis. Coronary arteries isolated from alpha(1)3.2-null arteries showed normal contractile responses, but reduced relaxation in response to acetylcholine and nitroprusside. Furthermore, acute blockade of T-channels with Ni2+ prevented relaxation of wild-type coronary arteries. Thus, Ca2+ influx through alpha1H T-type Ca2+ channels is essential for normal relaxation of coronary arteries.  相似文献

Producers often identify compaction as an important problem, so bulk density is usually included in minimum data sets used to evaluate tillage and crop management effects on soil quality. The hypothesis for this study was that bulk density and associated water content would be useful soil quality indicators for evaluating the transitional effects associated with changing tillage and crop management practices on deep-loess soils. The study was conducted on three deep-loess, field-scale watersheds located in western Iowa, USA. The soils are classified as Haplic Phaeozems, Cumulic-Haplic Phaeozems, and Calcaric Regosols. Watersheds 1 and 2 were converted in 1996 from conventional tillage to no-tillage, while watershed 3 was maintained using ridge-tillage and continuous corn (Zea mays L.), a practice implemented in 1972. Watershed 1 was converted to a corn—soybean (Glycine max (L.) Merr.) rotation while watershed 2 was converted to a 6-year rotation that included corn, soybean, corn plus 3 years of alfalfa (Medicago sativa L.). Bulk density and water content were measured at three landscape positions (summit, side-slope, and toe-slope), in 20 mm increments to a depth of 300 mm, five times between September 1996 and May 2000. Organic C and total N were also measured to a depth of 160 mm during the initial sampling. Neither bulk density nor water content showed any significant differences between the two watersheds being converted to no-tillage or between them and the ridge-till watershed. There also were no significant differences among landscape positions. Bulk densities and water contents showed some differences when adjacent sampling dates were compared, but there was no overall or consistent trend. Our results show that bulk density is not a useful soil quality indicator for these soils within the bulk density range encountered (0.8–1.6 Mg m3). Our results also confirm that producers do not necessarily have to worry about increased compaction when using ridge-tillage or changing from conventional to no-tillage practices on these or similar deep-loess soils.  相似文献

Trials in southern Tasmania examined the thinning effects of ethephon on ‘Gala’ and ‘Jonagold’ apples. Untreated controls were compared with ethephon thinning sprays applied at full bloom at 50,100,200,400,800 and 1 600 mg I“1 to both cultivars. Thinning of both cultivars was related to the concentration of the spray and in most cases logistic models were fitted. Thinning effectiveness was largely reflected in increased fruit weight and size. The ‘Gala’ fruit was still not large enough for Australian markets. Ethephon at 200 mg I“1 effectively thinned ‘Jonagold’ and produced the required increases in fruit size and weight. ‘Jonagold’ was overthinned by the high concentrations of ethephon, but this was not reflected in increased fruit weight and size. Ethephon also reduced vegetative growth at the higher concentrations which was considered an advantage. More work is required to establish specific recommendations for either cultivar.  相似文献

A trial, at the Grove Research Station in southern Tasmania, studied the thinning effect of benzyladenine (BA) on five year old red ‘Fuji’ apple trees. Untreated control trees were compared with trees treated at four timings, from full bloom (FB) to 30 d after full bloom (AFB), with four concentrations of BA (50, 100, 200 or 400 mg I-1). The most effective time of thinning was 20 d AFB, when increasing concentration also increased thinning. Further advantages of this successful thinning were reflected in better fruit size and weight. At 30 d AFB high concentrations of BA depressed fruit weight. Return bloom was improved significantly by both 20 and 30 d. AFB sprays with a concentration effect at 20 d AFB. Timing of sprays had no effect on the incidence of russet which was increased only by applications of 400 mg l-1. Both the higher concentrations and later timings resulted in an increase in shoot numbers and total growth. Reduction in mean shoot length was related more to the concentration of BA used than to differences in the timing of sprays. BA has shown potential as a thinner for red ‘Fuji’ apple at 20 d AFB.  相似文献

Fire and grazing significantly impact small-scale patch structure and dynamics in savanna grasslands. We assessed small-scale grass-forb associations in long-term fire and grazing experiments in North America (NA) and Southern Africa (SA). Transects of 128 0.25 m2 contiguous quadrats were sampled in areas with different combinations of grazing (ungrazed, single grazer, or multiple grazers) and fire frequency (unburned or annually burned). We predicted that (1) the patch structure of each of the dominant grasses in NA and SA would respond similarly to fire and grazing, and (2) that forb richness would be correlated to grass patch structure. Semi-variance analysis was used to determine patch structure of dominant grasses and forb cover. Community structure responded similarly in NA and SA to fire, grazing, and fire-grazing interactions. Species richness, diversity, and community heterogeneity were significantly higher in unburned-grazed sites. Grazing significantly increased forb cover and decreased cover of the dominant grasses, and the effects of fire on community structure depended on the grazing regime. Contrary to our prediction, we found that small-scale patch structure of the dominant grass species in NA and SA responded differently to grazing and fire. We found strong grass patch structure in unburned-ungrazed grasslands in both sites; however, grazing and fire reduced patch structure in NA but not SA, and in no instance did grass patch structure influence forb community structure. We conclude that fire and grazing have larger impacts on small-scale patch structure in NA than they do in SA even though overall community structure responded similarly on both continents.  相似文献

Transitioning farmland to certified organic vegetable production can take many paths, each varying in their costs and benefits. Here, the effects of four organic transition strategies (i.e., tilled fallowing, mixed-species hay, low-intensity vegetables, and intensive vegetable production under high tunnels), each with and without annual compost applications for 3 years prior to assessment, were characterized. Although transition cropping strategies differed in soil chemistry (P < 0.05), the magnitude of the changes typically were marginal and pairwise comparisons were rarely significant. In contrast, the compost amendment had a much greater impact on soil chemistry regardless of cropping strategy. For example, percent C and total P increased by 2- to 5-fold and K increased from 6- to 12-fold. Under controlled conditions, damping-off of both edamame soybean (cv. Sayamusume) and tomato (cv. Tiny Tim) was reduced from 2 to 30% in soils from the mixed-hay transition. In the field, damping-off of both crops was also significantly lower in plots previously cropped to hay (P < 0.05). Although not always significant (P < 0.05), this pattern of suppression was observed in all four of the soybean experiments and three of the four tomato experiments independent of compost application. The compost amendments alone did not consistently suppress damping-off. However, plant height, fresh weight, and leaf area index of the surviving seedlings of both crops were greater in the compost-amended soils regardless of the transitional cropping treatment used (P < 0.05 for most comparisons). These data indicate that mixed-hay cropping during the transition periods can enhance soil suppressiveness to damping-off. In addition, although compost amendments applied during transition can improve crop vigor by significantly enhancing soil fertility, their effects on soilborne diseases are not yet predictable when transitioning to certified organic production.  相似文献

Pot experiments were conducted to investigate the effect of soil water regimes on the formation of iron (Fe) plaque on the root surface of rice seedlings (Oryza sativa L.) and on the microbial functional diversity in a paddy soil. The rice seedlings were subjected to three moisture regimes (submergence, 100%, and 60% water‐holding capacity [WHC]), and were grown for 5 and 11 weeks. Aerobic lithotrophic Fe(II)‐oxidizing (FeOB) and acetate‐utilizing Fe(III)‐reducing bacteria (FeRB) in the rhizosphere and non‐rhizosphere soil were determined at 5 weeks using the most probable number (MPN) method. The carbon substrate use patterns of the microbial communities in the rhizosphere and non‐rhizosphere soil samples were determined at 11 weeks using Biolog‐GN2 plates. The amount of Fe plaque (per unit dry root weight) was much higher under submerged conditions than at lower soil moisture contents and decreased with plant age. There was a positive correlation between the amount of Fe plaque and phosphorus accumulated in the Fe plaque at both sampling times (r = 0.98 and 0.92, respectively, n = 12). Numbers of FeOB and FeRB in the submerged soil were lower than in aerobic soil, but by two orders of magnitude higher in the rhizosphere than in the bulk soil. On the other hand, the functional diversity of the rhizosphere microbial communities was much higher than that of the non‐rhizosphere soil, irrespective of soil water regimes. We conclude that soil flooding results in a decreased number and diversity of Fe‐oxidizing/reducing bacteria, while increasing the Fe‐plaque formation.  相似文献

Relaxin, a 6-kDa polypeptide hormone, is excreted in the urine during pregnancy in several mammalian species. A recent study showed that detection of urinary relaxin using a bench-top serum assay (Witness relaxin kit, Synbiotics Corp., San Diego, California 92127, USA) can be diagnostic for pregnancy in domestic cats (Felis silvestris catus), but it is unknown whether the bench-top kit is applicable with urine across felid species. Our objectives were to 1) examine modifications in urine processing to improve kit reliability in pregnant cats, 2) evaluate the impact of concentrating urine via filtration on relaxin detection, 3) assess the effect of sample freezing on relaxin concentrations, and 4) begin quantifying urinary relaxin levels in nondomestic felids. Urine and serum were collected from domestic cats and nondomestic cat species (Pallas' cat, Otocolobus manul; sand cat, Felis margarita; cheetah, Acinonyx jubatus; and lion, Panthera leo) at several times after breeding. Urine samples, subjected to various processing methods, were tested using the bench-top kit, and relaxin levels were later quantified via radioimmunoassay. For domestic cat urine samples, filtration and addition of protein/phosphate buffer improved the consistency of the relaxin kit for early pregnancy diagnosis. Urine freezing caused a slight (approximately 13%) but significant decrease in relaxin concentrations, but frozen-thawed samples still tested positive with the bench-top kit. In nondomestic felids, urinary relaxin immunoreactivity during pregnancy was similar to or higher than that of pregnant domestic cats, suggesting that relaxin is a reliable cross-species marker of pregnancy. Urinary relaxin was detectable using the bench-top kit in pregnant Pallas' cats, but urine samples from other species tested negative, regardless of processing methods. Findings suggest that measurement of urinary relaxin is a promising approach for noninvasive pregnancy diagnosis in exotic felids, but further assessment of urinary relaxin profiles among cat species and modification of the bench-top relaxin kit are warranted to improve cross-species utility.  相似文献
