The multiplication kinetics of the Flavescence dorée phytoplasma in broad bean after inoculation by the experimental vector Euscelidius variegatus was determined. The number of phytoplasma cells, measured by quantitative real-time PCR in the aerial parts of the plants, increased exponentially over the time. After 22 to 30 days post inoculation, when symptoms appeared, bacterial growth reached a stationary phase. Whatever the time following inoculation there were no statistical differences between numbers of phytoplasma cells in plants infected by Map-FD1 (FD-CAM05) and Map-FD2 (FD92 and FD-PEY05) genotype strains. On the contrary, temperature had an influence on Flavescence dorée phytoplasma multiplication which was nearly twice as fast in broad beans incubated at 25 °C than in broad beans incubated at 20 °C. At 25 °C, plants expressed symptoms 1 week earlier. In a context of climate change, the consequences of a global warming on the Flavescence dorée epidemics are discussed.  相似文献

A survey for phytoplasma diseases in tomato and pepper fields in Lebanon was conducted during 2003 and 2004. Tomato plants with stunting, yellowing or purplish leaves, proliferation of laterals buds, hypertrophic calyxes and virescent flowers were found in 25% of the tomato fields surveyed, where they represented 2–8% of the plants. Pepper plants displaying stunting and yellowing of leaves, were found in 27% of the fields and 1–4% of the plants were affected. Phytoplasmas infecting tomato and pepper had identical 16S-rDNA RFLP profiles and sequences. A phytoplasma isolate named PTL was transmitted by dodder from a diseased tomato plant to a periwinkle (Catharanthus roseus) plant in which it induced leaf yellowing, virescence and phyllody. 16S-rDNA phylogenetic analysis classified PTL as a strain of ‘Candidatus Phytoplasma trifolii’.  相似文献

The objective of this study was to synthesize and characterize two novel four-arm star-shaped copolymers of N-isopropylacrylamide (NIPAAm), methacrylic acid (MAA), N,N-(dimethylamino) ethyl methacrylate (DMAEMA) and 1-vinyl-2-pyrrolidone (VP) that was initiated from a new tetramethacrylate monomer (TMAM) as the core by the free radical polymerization approach. Novel multiacrylic monomer was synthesized via three key consecutive reactions. All the intermediates and the final product have been fully characterized. The polymerization reaction was conducted by the core first method and the star-shaped copolymers were analyzed by FT-IR, 1H NMR and Thermo-Gravimetric Analysis (TGA). MTT assay was used to evaluate in vitro cytotoxicity of the star copolymers on lung cancer cell lines. These star copolymers showed no in vitro cytotoxicity to A549 cells. The results showed that these copolymers can be promising drug carriers because they showed no in vitro cytotoxicity and they have various functional groups for association and interaction with drugs.  相似文献

Beta-cryptoxanthin (b-Cr) is a pro-vitamin A and one of the major carotenoids that can be commonly found in mammalian serum and tissues. Foods rich in certain fatty acids are known to be effective to gain a healthy immune system. In the present study, we evaluated the effect of b-Cr on rabbit humoral and cellular immune responses to have a better vision about the mechanism of effect of carotenoids on immune system. Twenty rabbits were randomly divided into five groups (4 per group): Groups consisted of: 1) control group (normal saline; 2) b-Cr (control); 3) vaccine control; 4) 5 mg/kg b-Cr o.p. + vaccine; 5) 10 mg/kg b-Cr o.p. + vaccine. Blood samples were obtained from the marginal ear artery at three time points: days 0, 14 and 21 of the study. Blood CD4+ and CD8+ lymphocytes and Serum Immunoglobulin and Cytokines content were evaluated. Results show that b-Cr administration increased the blood CD4+ lymphocytes count (P?>?0.01). Serum IgG, IgM and IgA levels increased (P?>?0.05) following b-Cr administration. b-Cr treatment increased serum IL-4 levels (P?>?0.05). According to presented results, b-Cr may increase the humoral immunity in mammals. So, it would possible has a potentially beneficial effect on health and on prevention of the immunity related diseases.  相似文献

ABSTRACT Marginal chlorosis has affected strawberry production in France for about 15 years. A phloem-restricted uncultured bacterium, "Candidatus Phlomobacter fragariae," is associated with the disease. A large-scale survey for marginal chlorosis in French strawberry production fields and nurseries by polymerase chain reaction amplification of "Ca. P. fragariae" 16S rDNA revealed that symptoms of marginal chlorosis were not always induced by "Ca. P. fragariae" and that the stolbur phytoplasma could induce identical symptoms. "Ca. P. fragariae" was found to be predominant in strawberry production fields, whereas the stolbur phytoplasma was predominantly detected in nurseries. Two transmission periods of the disease, one in spring and the other from late summer to early fall, were evident. Cixius wagneri planthoppers captured on infected strawberry plants were demonstrated to be efficient vectors of "Ca. P. fragariae." The involvement in natural disease spread of the whitefly Trialeurodes vaporariorum, previously shown to acquire and multiply "Ca. P. fragariae" under greenhouse conditions, remains uncertain.  相似文献

In this study, the effect of gonadotropin‐releasing hormone agonist (GnRHa) injection on milt production in spent rainbow trout was investigated. On day 0, 25 newly matured male rainbow trout (Oncorhynchus mykiss) were stripped manually, and sperm quantity (vol: mL fish?1) and quality, spermatocrit (%), sperm count (cell mL?1), motile sperm percentage and motility duration (s) were evaluated. After stripping, fish were randomly divided into five groups: intact; sham (injected with propylene glycol as a hormone vehicle); and groups receiving 4, 8 or 16 μg kg?1 BW of [d ‐Ala6 Des‐Gly10] mGnRHa. On day 7, the fish were stripped again and the same sperm characteristics as on day 0 were measured. At the beginning of the experiment, there were no significant differences in any of the sperm quantity characteristics between groups. On day 7, expressible milt volume was significantly reduced compared with day 0 (P<0.05, t‐test) in the intact and sham groups but milt quality remained the same (P>0.05, t‐test). The present study shows that GnRHa injection with a concentration as low as 4 μg kg?1 BW after first stripping could prevent a significant reduction in milt quantity collected 7 days later without any adverse effects on sperm quality.  相似文献

Kutum Rutilus frisii kutum (Kamenskii, 1901), Cyprinidae is an endemic fish of the Caspian Sea. Iranian Fisheries Organization (Shilat) produce up to 200 million fry (1–2 g body weight (b.w.)) to restock the Caspian Sea population annually. Some of these fry are produced by spawning induction in broodfish by carp pituitary extract (CPE). The objective of this study was to assay the effectiveness of the gonadotropin releasing hormone analogue (d ‐Ala6, Pro9‐Net GnRH) alone or in combination with metoclopramide (MET), a dopamine antagonist, on the percentage of ovulated females, latency period, ovulation index and fertilization success. The following hormone treatments were tested: single injection of 2 mg kg?1 b.w. of CPE as a positive control, GnRHa alone 20 and 40 μg kg?1 b.w. and combination of GnRHa and MET as follows: 5 μg+2.5 mg, 10 μg+ 5 mg and 20 μg+10 mg kg?1 b.w. Negative control group was injected with 0.7% saline. The percentage of ovulated females, ovulation index and fertilization success were 90%, 71.3±1.24%, 68.4±2.3%, respectively, in the group treated with GnRHa+MET at a dose of 20 μg+10 mg kg?1 b.w. and were significantly higher than those in the positive control (60%, 64.5±0.23%, 69.1±4.5%) (P<0.05). However, the latency period in this group was longer than that in the positive control (P<0.05). Only 20% and 40% fish ovulated in groups that received 20 or 40 μg kg?1 b.w. GnRHa. No fish ovulated in the negative control.  相似文献

Ralstonia solanacearum, the devastating causal agent of potato bacterial wilt, is a soil-borne bacterium that can survive in the soil for a long time. The development of sensitive on-field detection methods for this pathogen is highly desirable due to its widespread host range and distribution. A novel nanobiosensor was thus developed to detect unamplified genomic DNA of R. solanacearum in farm soil. Gold nanoparticles functionalized with single-stranded oligonucleotides served as a probe to detect R. solanacearum genomic DNA. The advantages of this strategy include rapidity, facile usage and being a visual colorimetric method.  相似文献

ABSTRACT

Climate change has led to a focus on forest management techniques to increase carbon (C) sequestration as a mitigation measure. Fertilisation and increased removal of biomass have been proposed. But these and other forest practices may have undesirable effects on surface water quality. In naturally acid-sensitive areas such as much of Fennoscandia a concern is acidification due to acid deposition in combination with forest practices that increase the removal of base cations and leaching of nitrate (NO3). Here we apply the biogeochemical model MAGIC to the coniferous-forested catchment at Birkenes, southernmost Norway, to simulate the effects of forest fertilisation and harvest on soil and streamwater. The model was calibrated to the 40-year data for water quality, soil and vegetation and then used to simulate fertilisation and clearcutting of the mature forest by either conventional stem-only harvest (SOH) or whole-tree harvest (WTH). The 5 – 10-year pulse of NO3 following clearcut was larger with SOH than WTH. WTH causes larger acidification of surface water relative to SOH, due to greater depletion of base cations, N and C from the soil. The use of forestry as a climate mitigation measure should take into consideration the potential effects on soil and surface water quality.  相似文献
