It has been reported that phosphatidylinositol 3-kinase (PI3K)-protein kinase B (PKB) pathway plays a crucial role in the meiotic resumption and progression to the metaphase II (MII) stage of oocytes. However, the role of this pathway in meiotic arrest at the MII stage (cytostatic activity) is not well understood. In this study the effect of a PI3K inhibitor, LY294002, on the MAPK and p34cdc2 kinase activities of matured porcine oocytes was examined. After maturation culture, both the MAPK and p34cdc2 kinase activities in the oocytes were gradually decreased in a time-dependent manner. Although 25 µmol/L LY294002 did not affect either the MAPK or p34cdc2 kinase activities, 50 µmol/L LY294002 suppressed the PKB phosphorylation and slightly decreased MAPK activity, but not the p34cdc2 kinase activity. Therefore the effect of 10 µmol/L Ca2+ ionophore which was reported as inducing a transient decrease of p34cdc2 kinase but not MAPK activities, was also examined in LY294002-treated oocytes. By additional treatment with LY294002 after Ca2+ ionophore, both the MAPK and p34cdc2 kinase activities were decreased in a time-dependent manner, concomitantly with improvement of pronuclear formation. Therefore, we concluded that PI3K is involved in the maintenance of MAPK activity in matured porcine oocytes.  相似文献

To evaluate the effects of forest fire and post-fire stand recovery on the organic layer chemistry and solute release within mound and trough microrelief elements (termed earth hummock microtopography) that mainly distribute permafrost affected area, we chose five fire plots (larch forests burned in 1951, 1981, 1990, 1994 and 2005) paired with adjacent control plots in mature larch forests in Central Siberian permafrost terrain. We determined total carbon, nitrogen and ash content in solid organic soils, and analyzed total carbon, nitrogen, bases and major anions in water extracts. There was a significant correlation between water-extracted organic carbon (WEOC) and total carbon (kg m−2) in area basis, implying that the quantity of total carbon was a major factor in WEOC production. WEOC correlated negatively with pH, indicating strong control by organic horizons (organic solute leaching) on soil acidity and base cation dynamics. The sum of water extractable base cations was also correlated significantly to total carbon, indicating that cations can be released through organic matter decomposition. Organic horizons in troughs in burned plots released greater amounts of Ca, Mg and K than those in mounds, probably due to greater content of organic matter as a cation source. Anions including nitrate and phosphate and WEOC also accumulated in trough depressions, due probably to organic matter degradation. The contrasting distribution of solutes between mounds and troughs in burned plots seems to be controlled by organic horizon development via changes in microtopography after forest fires.  相似文献

A previous serosurvey of Japanese encephalitis virus (JEV) among dogs suggested that dogs are well suited for use as sentinels for assessing the risk of JEV transmission to humans. To examine the clinical symptoms and duration of anti-JEV antibodies in dogs, three dogs were experimentally challenged with JEV. All JEV-infected dogs did not show any clinical signs or abnormal blood tests, except for C-reactive protein. Virus-neutralization titers rapidly increased and were maintained until 70 days postinfection, and neither the virus nor the viral genome was detected in blood. Thus, since dogs live in close proximity to humans as companion animals, they are well suited for use as sentinels for surveying the human risk of JEV infection.  相似文献

Data of 20 balance measurements from Holstein dairy cows and urine samples from 24 Japanese Black beef cows were collected to evaluate the relationships between urine pH and electrolyte status in cows fed forages. The ratio of forages in the diet was 70–100% in dairy cows and beef cows were fed Italian ryegrass silage and wheat bran. Mean urine pH in dairy cows was 8.10, ranging from 7.27 to 8.71, and that in beef cows was 7.73, ranging from 7.42 to 8.12. There were positive correlations between urine pH and urinary K contents (P = 0.0012) or K intake (P = 0.019) in dairy cows, although plasma Na, Cl and K had no effect on urine pH. There was a weak negative correlation (P = 0.039) between urine pH and urinary Na content in dairy cows. However, there were no significant correlations between urine pH and urinary Na, Cl and K contents in beef cows. These results indicate that the concentrated urinary K due to the increased K intake may directly enhance urine pH in dairy cows fed mainly forages.  相似文献

The ability to incorporate individual atoms in a surface following predetermined arrangements may bring future atom-based technological enterprises closer to reality. Here, we report the assembling of complex atomic patterns at room temperature by the vertical interchange of atoms between the tip apex of an atomic force microscope and a semiconductor surface. At variance with previous methods, these manipulations were produced by exploring the repulsive part of the short-range chemical interaction between the closest tip-surface atoms. By using first-principles calculations, we clarified the basic mechanisms behind the vertical interchange of atoms, characterizing the key atomistic processes involved and estimating the magnitude of the energy barriers between the relevant atomic configurations that leads to these manipulations.  相似文献

Meiotic cohesin complexes at centromeres behave differently from those along chromosome arms, but the basis for these differences has remained elusive. The fission yeast cohesin molecule Rec8 largely replaces its mitotic counterpart, Rad21/Scc1, along the entire chromosome during meiosis. Here we show that Rec8 complexes along chromosome arms contain Rec11, whereas those in the vicinity of centromeres have a different partner subunit, Psc3. The arm associated Rec8-Rec11 complexes are critical for meiotic recombination. The Rec8-Psc3 complexes comprise two different types of assemblies. First, pericentromeric Rec8-Psc3 complexes depend on histone methylation-directed heterochromatin for their localization and are required for cohesion during meiosis II. Second, central core Rec8-Psc3 complexes form independently of heterochromatin and are presumably required for establishing monopolar attachment at meiosis I. These findings define distinct modes of assembly and functions for cohesin complexes at different regions along chromosomes.  相似文献

Regolith particles on the asteroid Itokawa were recovered by the Hayabusa mission. Their three-dimensional (3D) structure and other properties, revealed by x-ray microtomography, provide information on regolith formation. Modal abundances of minerals, bulk density (3.4 grams per cubic centimeter), and the 3D textures indicate that the particles represent a mixture of equilibrated and less-equilibrated LL chondrite materials. Evidence for melting was not seen on any of the particles. Some particles have rounded edges. Overall, the particles' size and shape are different from those seen in particles from the lunar regolith. These features suggest that meteoroid impacts on the asteroid surface primarily form much of the regolith particle, and that seismic-induced grain motion in the smooth terrain abrades them over time.  相似文献
