Sapoviruses (SaVs) within the Caliciviridae family are an important cause of gastroenteritis in both humans and animals. Although the widespread occurrence of divergent human SaV strains has been reported, there have only been a few studies of porcine SaVs examining their genetic diversity. The aim of this study was to assess the genetic diversity of porcine SaVs in piglets with diarrhea in South Korea. Two hundred and thirty-seven fecal specimens from piglets with diarrhea were examined from 78 farms over a 2-year period from six provinces in South Korea. Overall, 69 (29.1%) of the samples from five provinces tested positive for porcine SaVs by either RT-PCR or nested PCR with the primer pairs specific to porcine SaVs. An analysis of the partial capsid gene (757bp) of 12 porcine SaVs detected from fecal samples showed genetic divergence, not only among the Korean porcine SaVs (67.7-99.1%), but also between Korean and American porcine SaVs (69.1-90.2%). Interestingly, one strain (Po/SaV/JN-MA113/05/Korea), formed a second porcine SaV/GIII genotype, and is tentatively called GIII/2. This strain had a 0.236-0.405 inter-cluster distance with the other strains in the same genogroup, which is comparable to the distances between the established GI and GII SaVs. Furthermore, two potential recombinant porcine SaVs were identified. In conclusion, porcine SaV infections are common in diarrheic piglets in South Korea. The infecting strains are genetically diverse, and include a newly recognized genotype and recombinant viruses within genogroup III.  相似文献

Parentage analysis is of vital importance for hatchery production and breeding programmes. Two multiplex PCR protocols including seven tropical spiny lobster (Panulirus homarus) microsatellites (Pho‐G06, Pho‐G25, Pho‐G53, Pho‐G62, Pho‐G74, Pho‐G89 and Pho‐G100) were introduced for parentage assignment. All loci were polymorphic with allele sizes from 113 to 337 base pairs (bp), observed alleles (k) from two to seven and polymorphic information content (PIC) of 0.25 to 0.73. Twenty‐four stage‐3 phyllosoma larvae (39 days posthatching) were collected for paternity exclusion study using selected microsatellites. Exclusion‐based parentage analysis unambiguously assigned 83% of fry (20 of 24) to a single female parent. Of ten putative female parents, five have contributed to the 20 allocated offspring, with one being true parent of 11. Four others were assigned to two or more potential female parents, probably due to genotyping error or the presence of null allele. The exclusion power (EP) for the seven loci varied between 13% and 54% with known genotypes of one parent (P1) and 19% and 71% for given the genotypes of both parents (P2). The theoretical combined parentage exclusion (cEP) power for all seven microsatellites was P1 = 95% and P2 = 99%. The power of discrimination for each locus varied between 0.18 and 0.86. This report presents the first study to utilize microsatellite markers for successful parentage assignment of P. homarus. The selected microsatellites provide a practical tool for parentage analysis in hatchery production of juveniles as well as in future commercial breeding programme of tropical spiny lobsters.  相似文献

The aim of this research was to study the effect of sulphur (S) fertilization on oil biosynthesis and its related variables at various stages of seed development, and to find possible explanations for increased oil content in the seeds of mustard (Brassica juncea L. Czern and Coss) due to S fertilization. Acetyl-CoA carboxylase activity and contents of oil, acetyl-CoA, soluble protein, total RNA, total sugar and sulphur were determined in the developing seeds of mustard grown in the field with sulphur (+S) and without sulphur (-S). The period between 10 to 30 days after flowering was observed as the active period of oil accumulation in the developing seeds of mustard. The accumulation of the oil was preceded by a marked rise in acetyl-CoA carboxylase activity and acetyl-CoA concentration, which declined rapidly when oil accumulation reached a plateau. Total sugar content decreased, while protein content increased during the active period of oil accumulation in the developing seeds (i.e. between 10–30 days after flowering). Sulphur fertilization significantly (P < 0.05) enhanced the oil accumulation in the developing seeds at all the growth stages. The increase in the oil content was 5–63% with S fertilization over the control treatment. Acetyl-CoA carboxylase activity and contents of acetyl-CoA, soluble protein, total RNA and sugar were significantly (P < 0.05) higher in the developing seeds of +S-treated plants compared to — S-treated plants. It is suggested that the increase in the oil content with sulphur fertilization may be associated with the increases in acetyl-CoA carboxylase activity through the enhancement of acetyl-CoA concentration. Further, the increased sugar content due to S fertilization provided enough carbon source and energy for oil biosynthesis.  相似文献

A battery trial was conducted to evaluate the comparative pathogenicity of five field strains of Eimeria tenella from Behera, Khafr El-Sheikh, Alexandria, Gharbia and Matrouh provinces in Egypt. Two-week-old broiler chickens were infected with 25×10(3) sporulated oocysts of each strain of E. tenella. The comparative pathogenicity of the strains was assessed by calculating body weight gain, feed conversion ratio, lesion scores, dropping nature scores, cecal scrapings, mortality percentage and oocysts count. Hematological parameters including hemoglobin (Hb) content, packed cell volume (PCV%) and total erythrocytic count, were also evaluated. Enteric viruses in diarrheic turkey poults   总被引:4,自引:0,他引:4  
Thirty-three intestinal samples from 10-to-21-day-old diarrheic turkey poults were examined for the presence of enteric viruses by electron microscopy. Samples originated from 32 flocks in six commercial operations located in six states. Mortality in these flocks ranged from 3 to 15%, and birds from recovered flocks varied greatly in size. Rotavirus-like agents (RVLA) were the most common viruses associated with diarrhea outbreaks in the flocks examined, occurring in five out of six operations. Other viruses detected either singly or in combination, in order of prevalence, were astroviruses, reoviruses, rotaviruses, enteroviruses, and adenoviruses. With the exception of RVLA and rotaviruses, the other viruses were identified solely on the basis of morphology. Salmonellae were isolated from only one of the intestinal samples. By electron microscopy, RVLA were morphologically indistinguishable from rotaviruses, occurring as both 55-nm single-shelled and 70-nm double-shelled particles. However, immune electron microscopy was useful for antigenic differentiation of these two viruses. Turkey rotaviruses reacted with antisera to porcine and bovine rotaviruses, whereas turkey RVLA did not. Neither turkey rotaviruses nor RVLA reacted with antisera to porcine para-rotavirus or an antigenically distinct bovine rotavirus (bovine rotavirus-like agent). Similarly, convalescent anti-turkey RVLA serum (from recovered specific-pathogen-free poults) reacted with homologous virus but did not react with mammalian or avian rotaviruses or reoviruses. Further, RVLA were found to possess RNA electrophoretic migration patterns unlike those of conventional rotaviruses or reoviruses. Six hundred fifty-five male and female turkeys representing four genetic lines were challenged in 10 experiments over a 3-year period with a field isolate of Pasteurella multocida. Poults were challenged at 45 days of age with 1 ml of an inoculum containing 1.2 x 10(7) bacteria per ml. The lines of turkeys included two randombred control lines (RBC1 and RBC2), a subline (E) of RBC1 selected for increased egg production, and a subline (F) of RBC2 selected for increased 16-week body weight. The number of days from exposure to severe clinical signs or death for Line F (5.8 days) differed significantly from that of Line E (8.2 days), Line RBC1 (8.0 days), and Line RBC2 (8.2 days). There were no significant differences due to sex of poult for number of days from exposure to severe clinical signs or death. Overall mortality observed was 51.2%. Mortality was highest for Line F (72.1%) and differed significantly from that of the other lines. Mortality among male poults did not differ significantly from mortality among female poults.  相似文献
