Contact-free digital image analysis was performed of the radial shrinkage of fresh, fully saturated small spruce wood beams. An experimental test set-up was developed to ensure constant distance from the charge-coupled device camera to the sample surface as well as constant climate and light conditions during the whole experiment. Dimensional changes were observed immediately after the drying process began. An unexpected distinct effect could be observed which could not be explained by drying surface layers only. After a fast initial radial shrinkage a slowing down of the dimensional changes occurred at high mean moisture contents. A complete interruption of any dimensional changes followed. Finally, a recovery from shrinkage was even observed. It is assumed that strong negative pressure occurred in the fully saturated capillaries owing to dehydration which led to additional dimensional changes. As a consequence, the break of the water column and aeration in these capillaries finally resulted in a recovery period in the shrinkage rate due to the pressure release. After this effect, the dehydration was characterized by a phase of fast and almost linear shrinkage due to drying surface layers. Finally, the shrinkage slowed down to zero when reaching equilibrium moisture content.  相似文献

The cellular immune response to tissue damage and infection requires the recruitment of blood leukocytes. This process is mediated through a classical multistep mechanism, which involves transient rolling on the endothelium and recognition of inflammation followed by extravasation. We have shown, by direct examination of blood monocyte functions in vivo, that a subset of monocytes patrols healthy tissues through long-range crawling on the resting endothelium. This patrolling behavior depended on the integrin LFA-1 and the chemokine receptor CX(3)CR1 and was required for rapid tissue invasion at the site of an infection by this "resident" monocyte population, which initiated an early immune response and differentiated into macrophages.  相似文献

The production of nitric oxide and other reactive nitrogen intermediates (RNI) by macrophages helps to control infection by Mycobacterium tuberculosis (Mtb). However, the protection is imperfect and infection persists. To identify genes that Mtb requires to resist RNI, we screened 10,100 Mtb transposon mutants for hypersusceptibility to acidified nitrite. We found 12 mutants with insertions in seven genes representing six pathways, including the repair of DNA (uvrB) and the synthesis of a flavin cofactor (fbiC). Five mutants had insertions in proteasome-associated genes. An Mtb mutant deficient in a presumptive proteasomal adenosine triphosphatase was attenuated in mice, and exposure to proteasomal protease inhibitors markedly sensitized wild-type Mtb to RNI. Thus, the mycobacterial proteasome serves as a defense against oxidative or nitrosative stress.  相似文献

Rising atmospheric carbon dioxide (CO2) concentrations over the past two centuries have led to greater CO2 uptake by the oceans. This acidification process has changed the saturation state of the oceans with respect to calcium carbonate (CaCO3) particles. Here we estimate the in situ CaCO3 dissolution rates for the global oceans from total alkalinity and chlorofluorocarbon data, and we also discuss the future impacts of anthropogenic CO2 on CaCO3 shell-forming species. CaCO3 dissolution rates, ranging from 0.003 to 1.2 micromoles per kilogram per year, are observed beginning near the aragonite saturation horizon. The total water column CaCO3 dissolution rate for the global oceans is approximately 0.5 +/- 0.2 petagrams of CaCO3-C per year, which is approximately 45 to 65% of the export production of CaCO3.  相似文献

In a prospective cross-over study, the duration and magnitude of effect on the electrolyte and plasma volume changes of intravenous (IV) hypertonic hydroxyethyl starch (hyperHES) (7.5%/6%) and mannitol (20%) were compared. Eight Beagle dogs received an IV infusion of 4mL/kg hyperHES (group H) and 4mL/kg mannitol 20% (group M) on separate occasions. Urine and blood samples were taken in the first (T(60)) and second (T(120)) hour after infusion. Significant increases in plasma volume at each time point in group H and M were noted when compared to baseline (start of infusion=T(0)) level. There was no significant difference between groups. Both fluids resulted in diuresis, although no significant difference between groups was noted. A significant increase in plasma sodium (Na) was demonstrated in group H between T(0) and T(60) with a significant increase in the Na and chloride (Cl) fractional excretion (FE) between T(0), T(60) and T(120). In group M no changes in plasma electrolyte concentrations were detected, although FE of Na, Cl and K was increased significantly between T(0) and T(60). In conclusion, hyperHES and mannitol appear to have a volume expanding effect lasting at least 120 min. The hypernatraemia induced by hyperHES was minimal compared to previous reports of hypertonic saline use, and no clinical side effects were noted. HyperHES showed comparable effects to mannitol in increasing plasma volume and diuresis and could be considered for these applications.  相似文献

Rust fungi like Puccinia graminis f. sp. tritici are known to change their cell wall properties upon entering the plant tissue. Immunohistochemistry revealed the cellular localization of two class III chitin synthase isoforms in rust mycelia developing on and in the host plant. Isoform IIIa is restricted to fungal infection structures growing on the surface of the plant, such as germ tubes and, predominantly, appressoria. Isoform IIIb is found exclusively in haustoria developed inside the plant. Thus, the rust fungus uses at least two chitin synthase isoforms with specialized functions in the differentiation of infection structures during the biotrophic plant-pathogen-interaction.  相似文献

Sequential application of solvent extraction, gel permeation chromatography, and RP-HPLC in combination with taste dilution analyses, followed by LC-MS and 1D/2D-NMR experiments and thiolytic degradation, revealed that, besides theobromine and caffeine, the flavan-3-ols epicatechin, catechin, procyanidin B-2, procyanidin B-5, procyanidin C-1, [epicatechin-(4beta-->8)](3)-epicatechin, and [epicatechin-(4beta-->8)](4)-epicatechin were among the key compounds contributing to the bitter taste as well as the astringent mouthfeel imparted upon consumption of roasted cocoa. In addition, a series of quercetin, naringenin, luteolin, and apigenin glycopyranosides as well as a family of not previously identified amino acid amides, namely, (+)-N-[4'-hydroxy-(E)-cinnamoyl]-L-aspartic acid, (+)-N-[3',4'-dihydroxy-(E)-cinnamoyl]-L-aspartic acid, (-)-N-[3',4'-dihydroxy-(E)-cinnamoyl]-L-glutamic acid, (-)-N-[4'-hydroxy-(E)-cinnamoyl]-L-glutamic acid, (-)-N-[4'-hydroxy-(E)-cinnamoyl]-3-hydroxy-L-tyrosine, (+)-N-[4'-hydroxy-3'-methoxy-(E)-cinnamoyl]-L-aspartic acid, and (+)-N-(E)-cinnamoyl-L-aspartic acid, have been identified as key astringent compounds of roasted cocoa. Furthermore, (-)-N-[3',4'-dihydroxy-(E)-cinnamoyl]-3-hydroxy-l-tyrosine (clovamide), (-)-N-[4'-hydroxy-(E)-cinnamoyl]-L-tyrosine (deoxyclovamide), and (-)-N-[3',4'-dihydroxy-(E)-cinnamoyl]-L-tyrosine, reported previously as antioxidants, have been found as contributors of cocoa's astringent taste. By means of the half-tongue test, the taste thresholds of flavan-3-ols and glycosides have been determined.  相似文献

We tested whether a ‘Lockerbraunerde’ from the heights of the Zittauer Gebirge in Eastern Saxony exhibited andic properties and classified it according to the rules of the World Reference Base for Soil Resources (WRB, 1998). To achieve this, we characterized a selected soil by means of routine soil analysis; selective dissolution procedures; X‐ray diffraction (XRD); X‐ray fluorescence (XRF), and Transmission Electron Microscopy (TEM). We used field criteria (Thixotropy; NaF‐field test) to obtain a map of the spatial distribution of soils with potential andic properties. We found that the soil fulfilled all requirements to be classified as an Andosol. The composition of the colloidal phases was exactly intermediate between sil‐andic and alu‐andic. At the same time, the soil had a spodic horizon [determined through the depth function of the Alo+½Feo criterion]. As there was no indication of vertical translocation of metal‐organic complexes, but sufficient evidence to suggest the downward movement of mobile Al/Si‐phases, we maintain to classify the soil as an Endoskeleti‐Umbric Andosol and propose the existence of a pedogenetic pathway intermediate between Podsolisation and Andosolization. We conclude that the spodic horizon in the WRB is not well defined because of the dominance of the Alo+½Feo criterion over morphological evidence. We further suggest the German soil taxonomy to be modified to better represent soils containing short range order minerals.  相似文献
