Using open-top chambers (OTC) on the shortgrass steppe in northern Colorado, changes of microbial community composition were followed over the latter 3 years of a 5-year study of elevated atmospheric CO2 as well as during 12 months after CO2 amendment ended. The experiment was composed of nine experimental plots: three chambered plots maintained at ambient CO2 levels of 360±20 μmol mol?1 (ambient treatment), three chambered plots maintained at 720±20 μmol mol?1 CO2 (elevated treatment) and three unchambered plots. The abundance of fungal phospholipid fatty acids (PLFAs) shifted in the shortgrass steppe under the influence of elevation of CO2 over the period of 3 years. Whereas the content of the fungal signature molecule (18:2ω6) was similar in soils of the ambient and elevated treatments in the third year of the experiment, CO2 treatment increased the content of 18:2ω6 by around 60% during the two subsequent years. The shift of microbial community composition towards a more fungal dominated community was likely due to slowly changing substrate quality; plant community forage quality declined under elevated CO2 because of a decline of N in all tested species as well as shift in species composition towards greater abundance of the low forage quality species (Stipa comata). In the year after which CO2 enrichment had ceased, abundances of fungal and bacterial PLFAs in the post-CO2 treatment plots shifted slowly back towards the control plots. Therefore, quantity and quality of available substrates had not changed sufficiently to shift the microbial community permanently to a fungal dominated community. We conclude from PLFA composition of soil microorganisms during the CO2 elevation experiment and during the subsequent year after cessation of CO2 treatment that a shift towards a fungal dominated system under higher CO2 concentrations may slow down C cycling in soils and therefore enhance C sequestration in the shortgrass steppe in future CO2-enriched atmospheres. 相似文献

To overcome soil nutrient limitation, many plants have developed complex nutrient acquisition strategies including altering root morphology, root hair formation or colonization by arbuscular mycorrhizal fungi (AMF). The interactions of these strategies and their plasticity are, however, affected by soil nutrient status throughout plant growth. Such plasticity is decisive for plant phosphorus (P) acquisition in P‐limited soils. We investigated the P acquisition strategies and their plasticity of two maize genotypes characterized by the presence or absence of root hairs. We hypothesized that in the absence of root hairs plant growth is facilitated by traits with complementary functions, e.g., by higher root mycorrhizal colonization. This dependence on complementary traits will decrease in P fertilized soils. At early growth stages, root hairs are of little benefit for nutrient uptake. Regardless of the presence or absence of root hairs, plants produced average root biomass of 0.14 g per plant and exhibited 23% root mycorrhizal colonization. At later growth stages of maize, contrasting mechanisms with functional complementarity explained similar plant biomass production under P limitation: the presence of root hairs versus higher root mycorrhizal colonization (67%) favored by increased fine root diameter in absence of root hairs. P fertilization decreased the dependence of plant on specific root traits for nutrient acquisition. Through root trait plasticity, plants can minimize trade‐offs for developing and maintaining functional traits, while increasing the benefit in terms of nutrient acquisition and plant growth. The present study highlights the plasticity of functional root traits for efficient nutrient acquisition strategies in agricultural systems with low nutrient availability. 相似文献

Background, aims and scope One issue that remains particularly problematic for integrated and sustainable sediment management is that sediment plays
differing roles in various sectoral, regulatory and statutory objectives. This means that across Europe, the level of consideration
afforded to sediment management has thus far been primarily left to the discretion of individual countries and agencies. One
of the consequences of the complex way in which sediments are regulated in Europe, and a possible over-reliance on the precautionary
principle, is that there appears to be less regulatory acceptance of risk-based (rather than mass-based or chemical threshold-based)
sediment remedial decisions, and thus resistance to some of the risk-based and in situ remedial technologies currently favoured
in North America; “presumptive remedies” are being pushed by a number of agencies, and in-place and risk-based management
are meeting great resistance. This summary discusses some of the current and emerging European regulations and strategies
and their implications for risk-based sediment management.
Conclusions, recommendations and perspectives There are numerous examples of ex situ remedial strategies in Europe, where sediments are either treated or contained (and
increasingly, considered for beneficial use), but very few examples of in situ management. However, the risk-based evaluation
of ALL remedial options is entirely consistent with European environmental policy. In fact, in some cases, presumptive removal
of sediments can result in greater risks to human health and the environment than management in place, and thus a failure
to meet Europe’s ambitious environmental objectives. Decisions that only address a single, sectoral regulatory driver (such
as a desire to remove contaminants from a waterbody) may result in a net detriment to the environment. The European Commission
has stated that decisions and policies should be continuously evaluated in the light of emerging science and experience, and,
where possible, rigorous science-based risk evaluation should take the place of the application of conservative safety factors.
Whilst not specifically addressing sediment management, various European Directives and initiatives set out principles for
the selection of land-based remedial options that are consistent with a number of international consensus guidance documents
on the site-specific, risk-based selection of contaminated sediment remedial options. Although there are numerous technical
differences, the fundamental risk- and cost-based principles being applied to land management should be transferable to sediment
management decision frameworks. However, whilst the use of risk-based decision criteria is now well-established (either in
principle or in practice) in Europe for soil remedial decisions, these principles have not yet been translated to generally
applied decision frameworks or guidance for contaminated sediment management in Europe. There is no reason, however, why they

Bremia lactucae is one of the most devastating and widespread pathogens in lettuce production worldwide. Despite its economical importance,
uncertainty prevails about the species delimitation in the genus Bremia. Commonly, Bremia is considered to be monotypic, containing only Bremia lactucae, while taxonomists have described additional species, and molecular phylogenetic studies have shown significant sequence
divergence between accessions from different hosts. Here, we report that several previously described species are genetically
highly distinct from Bremia lactucae parasitic to Lactuca sativa. These include Bremia lapsanae, Bremia sonchicola, and Bremia taraxaci. In addition to these host-specific species, a plurivorous species is revealed, which infects hosts from three different
tribes in the Asteraceae subfamilies Asteroideae and Carduoideae. The broad host range of clade 1 is exceptional for downy
mildews and only paralleled by Pseudoperonospora cubensis, which infects a broad range of Cucurbitaceae. The taxonomic status of Bremia cirsii and of Bremia centaureae remains unresolved, as the accessions from Cirsium and Centaurea, respectively, did not form a monophylum but were partly contained in the plurivorous clade 1. Bremia lactucae was found to be restricted to Lactuca sativa and Lactuca serriola. Thus, it can be assumed that Bremia infections on weeds apart from Lactuca species do not pose a significant risk for lettuce production. However, it is unlikely that breeding resistance genes from
Lactuca serriola into Lactuca sativa will result in durable resistance of lettuce to downy mildew disease, because the current study provides additional evidence

The applicability, transferability, and scalability of visible/near-infrared (VNIR)-derived soil total carbon (TC) models are still poorly understood. The objectives of this study were to: i) compare models of three multivariate statistical methods, partial least squares regression (PLSR), support vector machine (SVM), and random forest methods, to predict soil logarithm-transformed TC (logTC) using five fields (local scale) and a pooled (regional-scale) VNIR spectral dataset (a total of 560 TC spectral datasets), ii) assess the model transferability among fields, and iii) evaluate their up- and downscaling behaviors in Florida, USA. The transferability and up- and downscaling of the models were limited by the following factors: i) the spectral data domain, ii) soil attribute domain, iii) methods that describe the internal model structure of VNIR-TC relationships, and iv) environmental domain space of attributes that control soil carbon dynamics. All soil logTC models showed excellent performance based on all three methods with R2 > 0.86, bias < 0.01%, root mean squared error (RMSE) = 0.09%, residual predication deviation (RPD) > 2.70%, and ratio of prediction error to interquartile range (RPIQ) > 4.54. The PLSR method performed substantially better than the SVM method to scale and transfer the TC models. This could be attributed to the tendency of SVM to overfit models, while the asset of the PLSR method was its robustness when the models were validated with independent datasets, transferred, and/or scaled. The upscaled soil TC models performed somewhat better in terms of model fit (R2), RPD, and RPIQ, whereas the downscaled models showed less bias and smaller RMSE based on PLSR. We found no universal trend indicating which of the four limiting factors mentioned above had the most impact that constrained the transferability and scalability of the models. Given that several factors can impinge on the empirically derived soil spectral prediction models, as demonstrated by this study, more focus on their applicability and scalability is needed. 相似文献

Purpose
The objective of this work is to better understand the role that biological mediation plays in the behaviour of fine sediments. This research is supported by developments in ecological theory recognising organisms as “ecosystem engineers” and associated discussion of “niche construction”, suggesting an evolutionary role for habitat modification by biological action. In addition, there is acknowledgement from engineering disciplines that something is missing from fine sediment transport predictions.Materials and methods
Advances in technology continue to improve our ability to examine the small-scale 2D processes with large-scale effects in natural environments. Advanced molecular tools can be combined with state-of-the-art field and laboratory techniques to allow the discrimination of microbial biodiversity and the examination of their metabolic contribution to ecosystem function. This in turn can be related to highly resolved measurements and visualisation of flow dynamics.Results and discussion
Recent laboratory and field work have led to a paradigm shift whereby hydraulic research has to embrace biology and biogeochemistry to unravel the highly complex issues around on fine sediment dynamics. Examples are provided illustrating traditional and more recent approaches including using multiple stressors in fully factorial designs in both the laboratory and the field to highlight the complexity of the interaction between biology and sediment dynamics in time and space. The next phase is likely to rely on advances in molecular analysis, metagenomics and metabolomics, to assess the functional role of microbial assemblages in sediment behaviour, including the nature and rate of polymer production by bacteria, the mechanism of their influence on sediment behaviour.Conclusions
To fully understand how aquatic habitats will adjust to environmental change and to support the provision of various ecosystem services, we require a holistic approach. We must consider all aspects that control the distribution of sediment and the erosion-transport-deposition-consolidation cycle including biological and chemical processes, not just the physical. In particular, the role of microbial assemblages is now recognised as a significant factor deserving greater attention across disciplines.498.

Grasses, which are currently at the basis of cattle feeding, will, in the near future, be a major source of cell wall carbohydrates for sustainable biofuel production. The association of lignins with other matrix components, together with linkages between cell wall carbohydrates, greatly influences cell wall properties, including the degradability of structural polysaccharides by micro-organisms in animal rumen or industrial fermenters. The improvement in biofuel production from plants is based on the understanding of the cell wall composition and assembly, and on the discovery of genetic and genomic mechanisms involved in each component biosynthesis and their depositions in each lignified tissue. While nearly 40 QTL have been shown for lignin content, only seven locations appeared of greater importance in investigated genetic resources. Expression studies highlighted that several genes in the lignin pathway are less expressed in lines with higher cell wall degradability. However, only a few lignin pathway genes mapped in QTL positions, and the fully relevant candidates might be genes involved in regulation of lignin pathway genes, or in regulation of lignified tissue assembly. 相似文献

Calibration of pesticide leaching models may be undertaken to evaluate the ability of models to simulate experimental data, to assist in their parameterisation where values for input parameters are difficult to determine experimentally, to determine values for specific model inputs (e.g. sorption and degradation parameters) and to allow extrapolations to be carried out. Although calibration of leaching models is a critical phase in the assessment of pesticide exposure, lack of guidance means that calibration procedures default to the modeller. This may result in different calibration and extrapolation results for different individuals depending on the procedures used, and thus may influence decisions regarding the placement of crop-protection products on the market. A number of issues are discussed in this paper including data requirements and assessment of data quality, the selection of a model and parameters for performing calibration, the use of automated calibration techniques as opposed to more traditional trial-and-error approaches, difficulties in the comparison of simulated and measured data, differences in calibration procedures, and the assessment of parameter values derived by calibration. Guidelines for the reporting of calibration activities within the scope of pesticide registration are proposed. 相似文献
