The successful isolation and identification of rotavirus from newborn calves with diarrhoea is reported for the first time in Egypt. From 25 faecal samples taken from diarrhoeic calves, ten virus isolates were found to give cytopathogenic effects on bovine embryonic kidney cells. Three of the isolates were identified as rotavirus using fluorescent antibody staining, serum-neutralization, complement fixation and agar gel precipitation. The complement fixation test revealed the presence of rotavirus antibodies in 18 of 105 serum samples obtained from other calves slaughtered at Cairo abattoir.  相似文献

Background:Tuberculosis infection still represents a global health issue affecting patients worldwide. Strategies for its control may be not as effective as it should be, specifically in case of resistant strains of M.tb. In this regard, the role of mycobacterial MTases in TB infection can be fundamental, though it has not been broadly deciphered. Methods:Five resistant isolates of M.tb were obtained. M.tb H37Rv (ATCC 27249) was used as a reference strain. Seven putative mycobacterial MTase genes (Rv0645c, Rv2966c, Rv1988, Rv1694, Rv3919c, Rv2756c, and Rv3263) and Rv1392 as SAM synthase were selected for analysis. PCR-sequencing and qRT-PCR were performed to compare mutations and expression levels of MTases in different strains. The 2-ΔΔCt method was employed to calculate the relative expression levels of these genes.Results:Only two mutations were found in INHR strain for Rv3919c (T to G in codon 341) and Rv1392 (G to A in codon 97) genes. Overexpression of Rv0645c, Rv2756c, Rv3263, and Rv2966c was detected in all sensitive and resistant isolates. However, Rv1988 and Rv3919c decreased and Rv1694 increased in the sensitive strains. The Rv1392 expression level also decreased in INHR isolate.Conclusion:We found a correlation between mycobacterial MTases expression and resistance to antibiotics in M.tb strains. Some MTases undeniably are virulence factors that specifically hijack the host defense mechanism. Further evaluations are needed to explore the complete impact of mycobacterial MTases within specific strains of M.tb to introduce novel diagnosis and treatment strategies.Key Words: Drug resistance, Methyltransferases, Mycobacterium tuberculosis, S-adenosylmethionine  相似文献

A long‐term experiment comparing no‐till with conventional tillage systems across five rotations was evaluated 11 years after initiation. The objectives of the present paper are (1) to report differences in soil chemical properties (namely soil organic matter, total nitrogen, phosphorus, potassium and pH) that have resulted by converting from conventional to no‐till under contrasting cropping systems and (2) to draw tentative conclusions and recommendations on fertility status and fertilizer use and management. Soil in the no‐till system had increased surface soil organic C levels relative to conventional tillage regardless of rotation. In addition, depending on the rotation, the N and P content of the soil improved with no‐till compared with conventional tillage. In other words, no‐till has helped to retain soil organic matter (SOM), conserved more N, and resulted in increased extractable P and exchangeable K concentrations in the upper root‐zone. Hence, wheat produced in a no‐till system may receive more nutrients from decomposition of SOM and acidification of the seed zone. It is possible that lesser amounts of fertilizer nutrients will be needed because of the greater efficiency of nutrient cycling in no‐till systems relative to conventional systems. Three factors were examined to determine their effect on the reproduction and sex-ratio in the predacious miteAmblyseius zaheri Yousef & El-Borolossy in the laboratory. The factors studied included multiple matings, disturbed mating and age of mating females. Fecundity of females mated more than once was 3 times higher than in females mated only once. Artificial curtailment of copulation to 120, 60 and SO min resulted in reduction of fecundity and oviposition period. The Age of mated females has an influence also on fecundity, old females produce fewer eggs compared with young females under constant condition of abundant prey. All 3 factors studied have no influence on the sex-ratio of the progeny.  相似文献

In this research, phosphorus (P) desorption was investigated using DMT-HFO as an “infinite” sink for 10 calcareous soils over a period of 1–35 days and the kinetic data was related to soil properties and P uptake by maize (Zea mays L.). First- and second-order, and simple Elovich equations adequately described P desorption kinetics, however parabolic, exponential and zero order equations provided the best fits to the data. A systematic relationship exists between the second order rate parameter (k2) and the clay content of the soils. There are positive relationships between P desorption rate constants (for zero order, Elovich and parabolic equations), and both dry matter and P content of maize shoots and also for roots. These relationships indicate that P desorption rate may be a limiting factor for maize growth in these soils. The DMT-HFO technique may be used to assess the long-term availability of P to plants.  相似文献

Localization of Na+, K+-ATPase-rich cells in the gill and urinary system of Acipenser persicus fry was performed through immunofluorescence light microscopy using a mouse monoclonal antibody IgGα5 raised against the α-subunit of chicken Na+, K+-ATPase. Different types of epithelia were clearly identified in the gill epithelium: epithelia of branchial arch, interbranchial septum, filament and lamellar epithelium. The Na+, K+-ATPase-rich cells were found in the epithelia of branchial arch, interbranchial septum, filament, interlamellar region and also in the lamellae. Histologically, the urinary system is divided into head kidney, trunk kidney and short caudal kidney. The head kidney is composed of the pronephric tubules and the haemopoietic tissues, while the trunk kidney is composed of a large number of glomeruli and convoluted nephrons. Each nephron consisted of a large glomerulus and tubules (neck, proximal, distal and collecting tubules) which connected to ureters. Posteriorly, ureters extended and joined together to form a small urinary bladder. In the urinary system, no specific fluorescence staining was observed in the glomerulus, neck segment and proximal tubules. The distal tubule cells and collecting tubule cells showed a strong immunostaining of Na+, K+-ATPase. Epithelia of ureters and urinary bladder also showed several isolated immunofluorescent cells. Immunofluorescent cells were rich in Na+, K+-ATPase enzyme which is very important for osmoregulation.  相似文献

Wheat production in Morocco is constrained by both scarce climate and degraded soil quality. There is an urgent need to revert production decline while restoring country’s soils. Among conservation tillage systems known for their improvement in yield, no-till technology was found to influence soil quality as well. Soil quality indices are also affected by wheat rotations at medium and long-terms. This paper discusses changes in selected properties of a Calcixeroll soil, including total and particulate soil organic matter (SOM), pH, total N and aggregation, subjected, for 11 consecutive years, to various conservation and conventional agricultural systems. Tillage systems included no-tillage (NT) and conventional tillage (CT). Crop rotations were continuous wheat, fallow–wheat, fallow–wheat–corn, fallow–wheat–forage and fallow–wheat–lentils. Higher aggregation, carbon sequestration, pH decline and particulate organic matter (POM) buildup are major changes associated with shift from conventional- to NT system. Better stability of aggregates was demonstrated by a significantly greater mean weight diameter under NT (3.8 mm) than CT system (3.2 mm) at the soil surface. There was 13.6% SOC increase in (0–200 mm) over the 11-year period under NT, while CT did not affect much this soil quality indicator. Another valuable funding is the stratification of SOC and total nitrogen in NT surface horizon (0–25 mm) without their depletion at deeper horizon compared to tillage treatments. Fallow–wheat system resulted in reduction of SOC compared to WW, but 3-year wheat rotation tended to improve overall soil quality. Benefits from crop rotation in terms of organic carbon varied between 2.6 and 11.7%, with fallow–wheat–forage exhibiting the maximum. Combined use of NT and 3-year fallow rotation helped to improve soil quality in this experiment.  相似文献
