Strains of the pathogen causing bacterial blight of kiwifruit in New Zealand, previously identified as Pseudomonas viridiflava, were examined using phenotypic and genotypic methods. Percentage DNA–DNA reassociation values for strains of the pathogen, with the type strains representing P. viridiflava and P. savastanoi, and representative strains within P. syringae , were obtained using the S1 endonuclease method. Strains of the pathogen were most similar to the type strain of P. savastanoi. This similarity was supported by examination of the Δ T m between representative strains. It is concluded that the pathogen can be considered as a member of the P. savastanoi genomic species. The pathogen from kiwifruit in New Zealand was also differentiated in genomic terms from P. syringae pv. actinidiae . Strains of the kiwifruit pathogen compared using the Biomerieux API Biotype 100 system exhibited consistent determinative tests which distinguished the pathogen from P. viridiflava and P. syringae pv. actinidiae . Thirty-six isolates of the fasciation-inducing bacterium Rhodococcus fascians were evaluated for the presence and location of the fas-1 gene, which codes for an isopentenyl transferase, the committed step in cytokinin biosynthesis. The presence of fas-1 was determined by PCR using a set of primers to the most conserved regions of the gene and by Southern hybridization to genomic digests using the PCR fragment as a probe. Both methods revealed the presence of the gene in 18 virulent isolates and the absence of the gene in 18 avirulent isolates. Thus, there is a strong relationship between the presence of the gene and virulence of the organism. The location of fas-1 was determined by probing blots of linear and circular DNA. For most of the virulent isolates, the gene was localized to a 200 ± 10 kb linear plasmid. Three virulent isolates lacked a plasmid of this size, but contained fas-1 either on a linear plasmid of 130 kb or on a large circular plasmid. 相似文献

A family of mesoporous molecular sieves (denoted MSU-G) with vesiclelike hierarchical structures and unprecedented thermal (1000 degreesC) and hydrothermal stabilities (more than 150 hours at 100 degreesC) associated with high SiO4 cross-linking was prepared through a supramolecular assembly pathway that relies on hydrogen bonding between electrically neutral gemini surfactants of the type CnH2n+1NH(CH2)2NH2 and silica precursors derived from tetraethylorthosilicate. The vesicle shells are constructed of one or more undulated silica sheets that are about 3 nanometers thick with mesopores (average diameters from 2.7 to 4.0 nanometers) running both parallel and orthogonal to the silica sheets, which makes the framework structure bicontinuous and highly accessible. Elvers and glass eels of Anguilla anguilla were fed diets containing two types of feeding stimulants (FS) that were based on processed marine (MBFS) and yeast proteins (YBFS). Elvers (1.5 ± 0.3 g) were fed seven diets (MBFS and YBFS diets at 20 g kg?1, 40 g kg?1 and 60 g kg?1 plus control) for 60 days. Glass eels (250 ± 100 mg) were weaned to 60 g kg?1 MBFS, 60 g kg?1 YBFS and control diets for 30 days. Diets containing 60 g kg?1 FS had a beneficial effect in terms of growth, homogenous size distribution and feed intake in elvers. Elvers fed 60 g kg?1 MBFS and YBFS diets grew 11.9% and 5.6% faster than the control group. No differences in growth and size distribution were detected in glass eels fed 60 g kg?1 MBFS and YBFS diets. However, FS affected the digestive system maturation; fish fed the 60 g kg?1 MBFS and YBFS diets showed higher and intermediate values in the specific enzyme activities in comparison with the control group. This study revealed that the incorporation of FS into a pelleted diet was beneficial on the overall performance of European glass eels and elvers. Stability constants describing Al3+ combination with soil polycarboxylates were measured using the methods of Gregor et al. (1955) and Tanford (1961). It is suggested that the high stability of such complexes is derived largely from the electrostatic energy associated with the high surface charge of soluble humus acids. Chemically, the affinity of fulvic acids for Al3+ appeared to be no greater than that of a weak complexant such as adipic or acetic acid. A species distribution model was constructed showing the proportion of soluble Al that is bound by organic complexes. The model uses certain approximations regarding the degree of dissociation of the humus acids, but from measurements of soluble humus and Al in soil solutions it was suggested that these approximations were reasonably valid. The range of the model falls within conditions likely to occur in agricultural topsoils and demonstrates the importance of organic complexants in limiting Al3+ concentrations in natural aqueous systems. 相似文献

The limit to the precision with which moisture-release data may be related to soil structure is examined. Thin sections in two orthogonal planes were prepared from cores for which moisture release data and saturated conductivity measurements were also collected. Structural parameters are inferred indirectly through estimation of the mass fractal dimension from the moisture-release curve, and directly through measurement of the scaling properties of the pore space and solid matrix from the thin sections using image analysis. Theoretical results are presented which show that, in the absence of additional information, the interpretation of the moisture-release curve is ambiguous for several reasons. A power-law exponent is a consequence of either a fractal pore volume; a fractal solid volume; a fractal pore wall; or a non-fractal, self-similar pore wall, and one cannot infer from the measurement which is the case. Experimental results are presented which confirm that direct measurement of the fractal dimension of the solid matrix is a good predictor of the Brooks–Corey exponent for the soils studied here. Therefore, although a specific structural parameter characterizes the moisture-release curve, the latter cannot be used as a detailed measure of soil structure. 相似文献

A search was made for 26Mg (26Mg*) from the decay of 26Al (half-life = 0.73 million years) in Al-rich objects from unequilibrated ordinary chondrites. Two Ca-Al-rich inclusions (CAIs) and two Al-rich chondrules (not CAIs) were found that contained 26Al when they formed. Internal isochrons for the CAIs yielded an initial 26Al/27Al ratio [(26Al/27Al)0] of 5 x 10(-5), indistinguishable from most CAIs in carbonaceous chondrites. This result shows that CAIs with this level of 26Al are present throughout the classes of chondrites and strengthens the notion that 26Al was widespread in the early solar system. The two Al-rich chondrules have lower 26Mg*, corresponding to a (26Al/27Al)0 ratio of approximately 9 x 10(-6). Five other Al-rich chondrules contain no resolvable 26Mg*. If chondrules and CAIs formed from an isotopically homogeneous reservoir, then the chondrules with 26Al must have formed or been last altered approximately2 million years after CAIs formed; the 26Mg*-free chondrules formed >1 to 3 million years later still. Because 26Mg*-containing and 26Mg*-free chondrules are both found in Chainpur, which was not heated to more than approximately400°C, it follows that parent body metamorphism cannot explain the absence of 26Mg* in some of these chondrules. Rather, its absence indicates that the lifetime of the solar nebula over which CAIs and chondrules formed extended over approximately5 million years. 相似文献
