A study was conducted to evaluate the semen production and sperm motion characteristics of ram lambs by computer-aided semen analysis technique. Eight Malpura rams were raised under intensive management system and were trained for semen collection at a weekly interval from the age of 6 months. Rams were scheduled for semen collection at a weekly interval up to 1 year of age to assess their potential for semen production and objective evaluation of semen quality. The average age of ram lambs at the time of first ejaculation was 219 days ranging from 186 to 245 days. The age of ram lambs significantly (p < 0.05) influenced sperm concentration, sperm velocities, and beat frequency of spermatozoa, which were higher in 9–12-month-old compared to 6–9-month-old ram lambs. However, the effect of age was not significant on semen volume, percent motility, percent rapid, medium or slow motile spermatozoa, percent linearity, percent straightness, amplitude of lateral head displacement, percent elongation, and area of sperm head. The body weight of ram lambs was significantly (p < 0.01) and positively correlated (r = 0.46) with age. The results indicate that Malpura ram lambs of 9–12 months of age raised under the intensive management system in a semiarid tropical environment can produce good quality of semen.  相似文献

The present study deals with the effects of addition of sulfur along with other acidifying agents for their ability to lower and maintain the pH in a given range for a longer period of time. The chemicals were subjected to batch test individually and in combinations. Treatments were applied to three soils of different textures: sandy clay loam, clay loam, and silt loam. A 1:1 soil/water paste along with the added amendment was maintained at room temperature for 2 months. Most of the chemical treatments lowered the pH significantly. Combinations containing S/Al2(SO4)3/H2SO4, S/Al2(SO4)3/H2O2, and S/H2O2/H2SO4 were found to be very effective in lowering the pH. The soil pH remained acidic for 2 months, indicating the suitability of chemically amended soil for the plantations requiring acidic soil pH.  相似文献

Antioxidants are known to prevent the reactive oxygen species (ROS)‐mediated peroxidative damage to the membrane lipids during hypothermic storage of mammalian spermatozoa. We hypothesized here that ROS also affect the lipid–protein interactions, thereby diminishing the membrane's integrity and proteins' anchorage to the bilayer. Antioxidants prevent these damages by scavenging the ROS. Ejaculates from Patanwadi rams were pooled after subjective evaluation and centrifuged using Percoll®. Sperm pellet was resuspended in soya lecithin–Tris–fructose diluent (400 × 106 cells/ml) containing either antioxidants (100 IU/ml catalase + 10 mM reduced glutathione) or no antioxidant. Aliquots were chilled to 5°C in a cabinet and stored in a refrigerator at 3–5°C for 72 hr. Sperm motility, viability, lipid peroxidation (LPO) and hypo‐osmotic swelling test (HOST) were performed at 0, 24, 48 and 72 hr. Sperm proteins extracted with 0.5% Triton X‐100 were resolved by SDS‐PAGE and quantified using Quantity One software (Bio‐Rad, USA). The rapid motility, linearity and straight‐line velocity (VSL) were found significantly (p < .05) higher in the antioxidant‐treated group compared to the control at 48 hr of storage. Sperm viability was found comparable between the groups. Higher HOST response and lower LPO were found in the antioxidant‐treated sample compared to the control both at 48 and at 72 hr. Overall, the proteins P1 (106.09 kDa), P2 (87.00 kDa) and P4 (51.14 kDa) were lower (p < .05) in the sperm extract of antioxidant‐treated group compared to the control. The content of P4 (51.14 kDa) in sperm extract was found to increase (p < .05) earlier (48 vs. 72 hr) in the control group compared to the antioxidant‐treated group. Altogether, the results suggested that antioxidants reduced LPO in spermatozoa, resulting in higher sperm motility, plasma membrane integrity and protection of proteins' anchorage to the plasma membrane at 48 and 72 hr of storage.  相似文献

Nutritional scarcity during summer and winter is a major constraint for sheep rearing in the semi-arid tropical region. In this region, a new crossbred sheep has been developed by the crossing of a native sheep breed Malpura with a breed of hot-humid coastal region breed Garole. A study was conducted for 9 weeks to assess the physiological response of crossbred (Garole x Malpura x Malpura, GMM) rams under nutritional scarcity in the semi-arid tropical region. Eighteen adult crossbred rams were randomly allocated into three groups of six animals each. The groups are G1 (control, ad libitum feeding), G2 (20% less than ad libitum feeding), and G3 (30% less than ad libitum feeding). The animals were stall-fed with a diet consisting of 70% roughage (Cenchrus ciliaris hay) and 30% concentrate feed. Body weight gain, average daily gain, and feed intake were significantly (P?<?0.05) lower in G2 and G3 as compared to G1 rams. Water intake was significantly (P?<?0.05) higher in G2 and G3. Physiological responses, blood biochemical and endocrine profile did not differ between the groups. The results indicate that the crossbred rams evolved through the crossing of the hot semi-arid tropical region breed with hot humid coastal region breed are able to maintain their body homeostasis despite nutritional scarcity although their body weight decreased.

Seminal plasma (SP) is known to induce motility and capacitation in spermatozoa curtailing their lifespan when preserved. Hence, this study was conducted to examine the effects of removal of SP from sperm surface prior to liquid preservation either by high dilution (1/15) or by washing and the poststorage treatment with SP (15% and 25%, v/v) on the quality attributes of liquid‐preserved ram semen. Over the period of storage, the rapid motility (66.0% and 71.1% vs. 58.3%), straightness (87.1% and 82.1% vs. 79.4%), average path velocity (152.3 and 152.0 µm/s vs. 133.3 µm/s) and the straight‐line velocity (131.3 and 127.8 µm/s vs. 108.5 µm/s) were significantly (p < 0.05) higher in both the high‐dilution and wash groups as compared to the control (1/3 dilution). The functional membrane integrity (82.3% vs. 77.2%) and noncapacitated sperm count (65.0% vs. 58.7%) were also significantly (p < 0.05) higher in the high‐dilution and wash groups, respectively, as compared to the control. The poststorage treatment of sperm with SP significantly (p < 0.05) increased the functional membrane integrity (70.1% vs. 53.8%) and most of the motility attributes as compared to the control (without SP). In conclusion, both the removal of SP prior to liquid preservation and poststorage treatment with SP significantly improved the quality attributes of ram spermatozoa.  相似文献

A study was conducted to assess the effect of multiple stresses (thermal, nutritional, and walking stress) on the adaptive capability of Malpura ewes in terms of changes in physiological mechanisms and blood biochemical changes. The study was conducted for a period of 35 days covering two estrous cycles during summer season (April–May). The ewes were randomly allocated into two groups of 12 animals each, viz., GI (n?=?12; control) and GII (n?=?12; multiple stresses). GI ewes were maintained in the shed while GII ewes were subjected to multiple stresses. GI ewes were maintained in shed while GII ewes were subjected to heat stress by exposing them to 32–44 °C (average 42 °C) and RH of 12–26 % for 6 h from 10:00 to 16:00 h in natural environment. GI ewes were provided with ad libitum feeding while GII ewes were provided with restricted feed (30 % of intake of GI ewes) to induce nutritional stress. Further, GII ewes were subjected to walking stress by forcing them to walk 14 km in two spans between 9:00 and 10:30 h and 15:00 and 16:30 h. Blood collection was done at weekly intervals. Multiple stresses significantly (P?<?0.01) affected body weight, respiration rate, pulse rate, rectal temperature, sweating rate, tri-iodo-thyronine, thyroxine, cortisol, hemoglobin, packed cell volume, glucose, and total protein. The findings from this experiment provide useful information to understand the ideal nutrient requirement for these animals to adapt to such stresses in the semi-arid tropical environment without compromising production.  相似文献

Feed scarcity during hot summer months is one of the major predisposing factors for low reproductive efficiency of livestock reared in hot semiarid environment. A study was conducted to assess the effect of concentrate supplementation during summer months on growth, reproductive performance, and blood metabolites in Malpura ewes. Twenty adult Malpura ewes were used in the present study. The ewes were divided into two groups viz, group 1 (n?=?10; control) and group 2 (n?=?10; concentrate supplementation). The study was conducted for a period of 35 days covering two estrus cycles. In the first cycle, only PGF was given to all ewes, while in second cycle, all ewes were synchronized for estrus using progesterone-impregnated intravaginal sponges and pregnant mare serum gonadotropin. The animals were allowed for grazing for 8–10 h per day. Apart from grazing, group 2 ewes were supplemented with concentrate mixture at 1.5 % of body weight. Concentrate supplementation had significant influence on body weight, ADG, estrus percentage, estrus duration, onset of estrus, ovulation response, plasma glucose, total protein, and urea. The present study reveals that ewes supplemented with concentrate mixture at 1.5 % of body weights during summer season significantly influenced the growth and reproductive performance of Malpura ewes. Further, the study signifies the importance of providing additional feed supplementation to ewes kept grazing under the conditions of a hot, semiarid environment to improve their reproductive efficiency.  相似文献
