We conducted 3 independent experiments to demonstrate functional G-coupled protein receptor 43 (GPR43) and GPR120 in bovine intramuscular (i.m.) and subcutaneous (s.c.) adipose tissues. We hypothesized that media volatile fatty acids and long-chain fatty acids would affect cAMP-activated protein kinase-alpha (AMPKα) protein expression and cAMP concentrations differently in i.m. and s.c. adipose tissue. Experiment 1: oleic acid (18:1n-9) decreased phosphorylated AMPKα protein (p-AMPKα) and the p-AMPKα/AMPKα protein ratio in i.m. preadipocytes, increased the p-AMPKα/AMPKα protein ratio in bovine satellite cells, and had no effect in s.c. preadipocytes. Experment 2: ex vivo explants from the 5th to 8th longissimus thoracic rib muscle section of Angus crossbred steers were cultured 48 hr in media containing 0.25 µM ciglitizone, 5 mM glucose, and 5 mM acetate, in the absence or the presence of 100 µM oleic acid. Oleic acid increased acetate incorporation into fatty acids and GPR43 gene expression in i.m. adipose tissue (P < 0.05), but oleic acid had no effect on fatty acid synthesis or GPR43 expression in s.c. adipose tissue. Experiment 3: fresh s.c. and i.m. adipose tissue from the 5th to 8th longissimus thoracic rib muscle section of Angus crossbred steers was transferred immediately to 6-well culture plates containing 3 mL of KHB/Hepes/5 mM glucose. Samples were preincubated with 0.5 mM theophylline plus 10 μM forskolin for 30 min, after which increasing concentrations of acetate or propionate (0, 10−3, 10−2.3, and 10−3 M) in the absence or the presence of 100 μM oleic acid or 100 µM palmitic acid (16:0) were added to the incubation media. Acetate had no effect on forskolin-stimulated cAMP production in s.c. adipose tissue but decreased cAMP in i.m. adipose tissue (P < 0.05); this indicates a functional GPR43 receptor in i.m. adipose tissue. The combination of 10−2 M acetate and oleic acid decrease cAMP production in s.c. adipose tissue, consistent with GPR120 receptor activity, but oleic acid and palmitic acid attenuated the depression of cAMP production caused by acetate in i.m. adipose tissue. Palmitic acid depressed cAMP production in s.c. adipose tissue, and increased cAMP production in i.m. adipose tissue (P < 0.05). Propionate had no effect on cAMP production in s.c. or i.m. adipose tissue. These results provide evidence for functional GPR43 receptors in i.m. adipose tissue and GPR120 receptors in s.c. adipose tissue, both of which would suppress lipolysis.  相似文献

Background

Data on the movement behavior of translocated wild pigs is needed to develop appropriate response strategies for containing and eliminating new source populations following translocation events. We conducted experimental trials to compare the home range establishment and space-use metrics, including the number of days and distance traveled before becoming range residents, for wild pigs translocated with their social group and individually.

Results

We found wild pigs translocated with their social group made less extensive movements away from the release location and established a stable home range ~5 days faster than those translocated individually. We also examined how habitat quality impacted the home range sizes of translocated wild pigs and found wild pigs maintained larger ranges in areas with higher proportion of low-quality habitat.

Conclusion

Collectively, our findings suggest translocations of invasive wild pigs have a greater probability of establishing a viable population near the release site when habitat quality is high and when released with members of their social unit compared to individuals moved independent of their social group or to low-quality habitat. However, all wild pigs translocated in our study made extensive movements from their release location, highlighting the potential for single translocation events of either individuals or groups to have far-reaching consequences within a much broader landscape beyond the location where they are released. These results highlight the challenges associated with containing populations in areas where illegal introduction of wild pigs occurs, and the need for rapid response once releases are identified. © 2023 The Authors. This paper describes the use of a geographical information system (GIS) to construct environmental models for land-based aquaculture development in the State of Sinaloa, Mexico. Based on the source data, submodels were created focusing on three different themes: general environmental issues, water resources and water quality. Models enabled multicriteria and multiobjective decision making concerning site selection and location. In assessing site considerations these general models identified wider resource management options and solved conflicts of land allocation and land use between aquaculture and agriculture. Smaller-scale, more specific models enabled more detailed studies on environmental issues.  相似文献

The Mexican silverside, Menidia estor, is a species with great regional importance and with very high prices in local markets. Unfortunately, due to high fisheries pressure, environmental degradation and pollution, the species has become endangered. Recently, there has been much progress in the biotechnology of this species, aimed at its culture, and the present paper describes the advances in feeding and nutrition of those important fish. M. estor is a stomachless, zooplantophagous fish, which also occasionally feeds on small fish and crustaceans in the adult stage. Studies on the digestive enzymatic activities show high proteolytic capacity and a late or different model of digestive maturation from that described for marine stomach fish. Nutritional studies on M. estor have shown that juveniles have dietary requirements of about 400 g kg?1 protein and 80 mg kg?1 vitamin C. The upper level of dietary carbohydrate for good growth and survival for juveniles is about 150 g kg?1. Based on the high docosahexaenoic acid (DHA) content in flesh when fed on diets low in DHA, it is believed that the species has the capacity to biosynthetize >20‐carbon fatty acids from 18‐carbon fatty acids. The high levels of DHA in flesh makes this fish a very significant potential component of human nutrition. Using these findings the first practical diets for commercial culture of the species have been developed.  相似文献

为研究鉴选不同质地土壤秸秆还田适宜的秸秆腐熟剂,于2017—2018年在西辽河平原灌区中壤土和砂壤土秸秆还田配施人元腐熟剂和中农绿康腐熟剂,以秸秆还田不施腐熟剂为对照,共设置6个处理,测定玉米秸秆腐解率、玉米产量、玉米根系特性和土壤化学性状,研究不同秸秆腐熟剂对玉米产量及土壤特性的影响。结果表明:不同质地土壤2种腐熟剂秸秆腐解率间无显著差异;中壤土2年各处理玉米产量及根系特性、土壤化学性状均表现为人元腐熟剂中农绿康腐熟剂秸秆还田不施腐熟剂,且中农绿康腐熟剂和人元腐熟剂显著高于秸秆还田不施腐熟剂,而2种腐熟剂处理之间无显著差异;砂壤土2年各处理玉米产量及根系特性、土壤化学性状均表现为中农绿康腐熟剂人元腐熟剂秸秆还田不施腐熟剂,且中农绿康腐熟剂显著高于人元腐熟剂和秸秆还田不施腐熟剂。2年不同质地土壤中配施腐熟剂在根系特性、土壤化学性状方面表现为砂壤土配施中农绿康腐熟剂效果优于人元腐熟剂,而中壤土配施2种腐熟剂效果无显著性差异。  相似文献

Polyols have been incorporated into fish oil emulsions as a means for the inhibition of lipid oxidation and suppression of fishy flavor. However, the role of sugars and polyhydric alcohols as antioxidants has not been clearly established. Selected polyols were evaluated for their performance as antioxidants and modifiers of oxidation pathways in a model system. Oil/water (O/W) emulsions were prepared with freshly steam-deodorized menhaden oil. A layer of emulsion in aluminum pans held at 5 degrees C was exposed to 2550 lx fluorescent lights for 24 h before peroxide values and volatile flavor compounds were analyzed by GC headspace entrainment procedure. Antioxidant activity was confirmed for fructose, sucrose, raffinose, sorbitol, or mannitol when incorporated at 16% of the aqueous phase into model fish oil-in-water emulsions. Peroxide values were suppressed 10-18% in treated samples compared to control samples. Viscosity data did not exclude possible contributions from a restricted oxygen diffusion mechanism in the antioxidant activity, but revealed that emulsion viscosity did not govern fish oil oxidation rates. Combining polyols with phenolic antioxidants (alpha-tocopherol, BHT, or TBHQ) frequently diminished the antioxidant activity compared to that for individual phenolic antioxidants, which was interpreted as indicating that the H-donating activity of phenolic antioxidants was hindered by the H-bonding activity of polyols. A viscosity-based inhibition of the retroaldol conversion of (E,Z)-2,6-nonadienal to (Z)-4-heptenal with a high fructose concentration (67%) was attributed to a restriction of molecular mobility of reactants, but the conversion was only slightly inhibited by the concentration of fructose (16%) used in experimental emulsions. The data supported a hypothesis that either or both free radical scavenging and transition state metal chelation activities were provided by polyols in fish oil emulsions. Also, polyols retarded the water-requiring retroaldol decomposition of (E,Z)-2,6-nonadienal to (Z)-4-heptenal in the model systems and the reaction may be involved in some suppression of fishy flavors in emulsions.  相似文献
