We previously reported that the majority of in vitro monocyte/macrophage activation exhibited by extracts of Echinacea and other botanicals depends upon bacterial lipopolysaccharides and Braun-type bacterial lipoproteins. We determined the contribution made by these bacterial components to the overall immune-enhancing activity detected in E. purpurea and E. angustifolia bulk root and aerial material obtained from six major growers/suppliers in North America. Substantial variation in activity (up to 200-fold) was observed in extracts of these materials when tested in two monocyte/macrophage cell lines. The majority of activity was negated by treatment with agents that target bacterial lipoproteins (lipoprotein lipase) and lipopolysaccharides (polymyxin B). Experiments comparing the activity of freeze-dried, freshly harvested Echinacea plants to those harvested and dried using various commercially relevant conditions suggest that postharvesting procedures do not substantially contribute to the variation observed in the commercial material.  相似文献

Equine proliferative enteropathy (EPE) is a disease of foals caused by the obligate intracellular organism Lawsonia intracellularis. This emerging disease affects mainly weanling foals and causes fever, lethargy, peripheral oedema, diarrhoea, colic and weight loss. The diagnosis of EPE may be challenging and relies on the presence of hypoproteinaemia, thickening of segments of the small intestinal wall observed upon abdominal ultrasonography, positive serology and molecular detection of L. intracellularis in faeces. Although the clinical entity, diagnostic approach and treatment of EPE are well established and described, the epidemiology for this disease has remained largely unaddressed. This article focuses on new developments in the field of EPE, including epidemiology, pathophysiology, clinical signs, diagnosis, treatment and prevention. Ten-week-old layer chickens obtained from a commercial source were eye-drop vaccinated with chicken-embryo-origin (CEO) or tissue-culture-origin (TCO) vaccines for infectious laryngotracheitis (ILT). Controls were not vaccinated. Approximately one-third of the layers were challenged with virulent ILT virus at 21, 40, or 60 weeks of age. Serum samples taken from the layers before challenge were used in a virus neutralization (VN) test to determine vaccination titers at those three ages. Both vaccines induced low VN titers (geometric mean titer [GMT] less than 6). At 21 weeks of age, the titers produced by the two vaccines were not significantly different, but at 40 and 60 weeks of age the VN GMT of the CEO-vaccinated group was significantly greater than that of the TCO-vaccinated group. The VN GMTs did not drop over time in either group and actually rose between 21 and 60 weeks of age in the CEO group. Both vaccines protected layers against severe challenge with virulent ILT virus, neither being significantly better than the other under these experimental conditions. Unvaccinated sentinel chickens were maintained in contact with the vaccinated layers during three intervals between 1 day and 6 weeks post-vaccination. Diagnostic tests performed on the sentinels to detect lateral spread of vaccine virus from vaccinated to unvaccinated chickens showed scattered positive results.  相似文献

Development of effective and safe treatments for companion animals with cancer requires the collaboration of numerous animal health professionals and the full engagement of animal owners. Establishing ‘Best Practice Recommendations’ for clinical trials in veterinary oncology represents an important step toward meeting the goal of rigorous clinical trial design and conduct that is required to establish valid evidence. Likewise, optimizing patient welfare and owner education and advocacy is crucial to meet the unique ethical obligations to both owners and animals enrolled in these clinical trials and to ensure trust in the team conducting the research. To date, ‘Best Practice Recommendations’ for clinical trial conduct have not been reported for veterinary oncology. This document summarizes the consensus of a workshop held in November, 2014 to identify relevant ethical principles and to ensure responsible conduct of clinical research in companion animals with cancer. It is intended as a working document that will be updated as advances in science and ethical considerations require. To the extent possible, existing guidelines for the conduct and oversight of clinical trials in humans have been adapted for veterinary trials to avoid duplicative effort and to facilitate integration of clinical trials such that translational research with benefits for both companion animals and humans are encouraged.  相似文献

The nitrifying activity of the filter bed of aquaria was monitored in hypersaline conditions. Salinity was raised in 5 ppt steps from normal sea water to 126 ppt in a course of 35 and 41 days. Twenty grey mullet fry (Chelon labrosus and Mugil cephalus) were used in two 53 l aquaria. Daily monitoring of total ammonia–nitrogen (TAN), nitrite, nitrate ions, and pH was performed. After an initial peak of TAN (0.65 mg/l), total ammonia stabilized at safe low levels (0–0.2 mg/l). Nitrite exhibited more peaks at levels of about 10 mg/l, lasted for 3–8 days but they also subsided to almost zero afterwards. Nitrate finally stabilized at levels around 100 mg/l. The water was alkaline, with pH fluctuating in the region of 8.2–8.8. These findings indicate that the filter bed may be effectively conditioned for hypersalinity and that fish may be safely maintained in the aquaria.  相似文献

The efficacy of ivermectin (0.08 per cent w/v oral solution) at different dose levels was evaluated against induced infections of adult Haemonchus contortus (21 days old) and Trichostrongylus colubriformis (21 days old) and fourth stage larvae of Oesophagostomum columbianum (17 days old), Ostertagia circumcincta (five days old) and Strongyloides papillosus (five days old). Twenty-five Boergoats (mutton goats) were randomly allocated by bodyweight within each sex to an untreated control group and four ivermectin treatment groups; ivermectin was administered at either 25, 50, 100 or 200 micrograms/kg orally, once. The goats were killed and processed for worm recovery 25 to 27 days after treatment. At 25 micrograms/kg the efficacy of ivermectin varied from 43 per cent for adult T colubriformis to more than 99 per cent for fourth larval stage O columbianum. Ivermectin at 50 micrograms/kg or higher was 99 per cent or more effective against all induced parasite infections with the exception of ivermectin at 50 micrograms/kg against S papillosus (97 per cent). For all parasites there was a statistically significant (P less than 0.05) difference between the control group and the pooled treated groups. No adverse reactions to ivermectin treatment were observed in the goats.  相似文献

The rust fungus Austropuccinia psidii has spread globally and naturalized in areas with naïve species of Myrtaceae. Previous studies have revealed multiple strains of A. psidii within South America and two strains outside of its native range. The rust spreads by windborne mitotic urediniospores, which are the dominant spore stage. Teliospores and basidiospores of A. psidii are also formed; however, the biological role of these stages in the life cycle is unknown. Experiments presented here tested whether basidiospores of A. psidii could infect Syzygium jambos. The sori produced by infection with basidiospores were screened with five microsatellite markers to confirm whether they were a product of recombination. The findings showed that basidiospores of A. psidii could cause infection on species of Myrtaceae and the resulting sori were a product of recombination. This has important implications for programmes that breed for resistance to this aggressive pathogen in commercial eucalypt forestry.  相似文献

Arthrotomies of middle carpal joints were done on 13 horses, and a 1-cm partial thickness, round defect was made on the radial facet of both third carpal bones. In one joint, 1-mm diameter 1-cm deep holes were drilled within the defect, and one joint was used as a control. horses were assigned to 2 groups--group 1 (n = 6 horses), 5 drill holes; group 2 (n = 7 horses), 11 drill holes. At 1 and 3 weeks after surgery, differences between joints in synovial fluid total protein values, WBC counts, or results of mucin precipitate tests were not significant (P = 0.005). Physically and radiographically, horses were the same during the 12 initial weeks they were housed in stalls and the 9 weeks they were kept in paddocks. Twenty-one weeks after surgery, horses were euthanatized. Joints with drill holes had a significantly greater area (P less than 0.05) of healthy fibrocartilage new tissue: group 1-33 to 68% new tissue, compared with 0 to 23% new tissue in controls; and group 2-22 to 64% new tissue, compared with 0 to 37% new tissue in controls. Differences between healing of defects with drill holes in groups 1 and 2 were not significant. Thickness of new tissue over drill holes was 33 to 61% of thickness of cartilage adjacent to the defect, and thickness of tissue between drill holes was 11 to 43% (group 1) and 8 to 79% (group 2) of the thickness of cartilage adjacent to the defect.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Epileptic seizures can lead to various reactions in the brain, ranging from neuronal necrosis and glial cell activation to focal structural disorganization. Furthermore, increased hippocampal neurogenesis has been documented in rodent models of acute convulsions. This is a report of hippocampal neurogenesis in a dog with spontaneous epileptic seizures. A 16-week-old epileptic German Shepherd Dog had marked neuronal cell proliferation (up to 5 mitotic figures per high-power field and increased immunohistochemical expression of proliferative cell nuclear antigen) in the dentate gyrus accompanied by microglial and astroglial activation. Some granule cells expressed doublecortin, a marker of immature neurons; mitotically active cells expressed neuronal nuclear antigen. No mitotic figures were found in the brain of age-matched control dogs. Whether increased neurogenesis represents a general reaction pattern of young epileptic dogs should be investigated.  相似文献

There is a need to develop farming systems that enable both a satisfactory level of crop production and suitable environmental conditions for natural species. Wildlife‐friendly cropping techniques, such as a reduced amount of applied herbicide or a lower crop density, might be adopted in order to maintain populations of weed species of biological interest. An alternative might be to adopt an intensive cropping system in a part of the field and spare the other part as set‐aside or field margins, available for the natural development of plant species. The objective of this study was to present a method to compare two strategies for maintaining a desirable level of abundance of a given species of interest in agricultural areas, specifically (i) a strategy based on a wildlife‐friendly cropping system in a large cultivated area and (ii) a strategy based on a more intensive cropping system in a reduced area of cultivation, i.e. with land‐sparing. The principle is to calculate the ratio of crop production obtained with strategy (i) to the production obtained with strategy (ii) for a given target density of natural species. We show that the value of this ratio, and thus the relative performance of the two strategies, depends on the density of the weed species that can be maintained in an uncultivated ecological area. The method is applied in case studies of two plant species with contrasting ecology and conservation goals. The numerical results show that the strategy based on a wildlife‐friendly cropping system is more profitable in most situations.  相似文献
