Artificial insemination (AI) can help to avoid inbreeding and genetic degeneration
for sustaining genetically healthy populations of endangered species in captivity.
Collection of a sufficient quantity of viable sperm is an essential first step in the
AI process. In the present study, we examined the effects of frequent
electroejaculation on semen characteristics in a Siberian tiger. We collected semen
in all 17 trials during 6 breeding seasons (6 years). The mean number of sperm and
the percentage of motile sperm were 294.3 ± 250.2×106/ejaculate and 82.4 ±
11.4%, respectively. The number of motile sperm tended to increase during frequent
electroejaculation in the same breeding season. Semen collection by
electroejaculation can be performed effectively up to the fourth sequential
ejaculate, which contained the most sperm in the study. In conclusion, frequent
collection of sperm by electroejaculation from tigers may be effective for collection

We cloned a cDNA fragment encoding a feline homologue of L-selectin (CD62L). The extracellular region of the feline CD62L fragment contained a calcium-dependent (C-type) lectin domain, an epidermal growth factor-like domain, and two Sushi/CCP/SCR domains. To evaluate the effects of ketamine continuous rate infusions (CRI) at two dose rates on cardiovascular function and serum creatine kinase MB isoenzyme (CK-MB) and troponin I in healthy conscious dogs.Study design
Experimental, prospective, crossover, randomized, blinded study.Animals
Eight adult mixed-breed dogs, aged 6 ± 1 years and weighing 19 ± 8.6 kg (mean ± standard deviation).Methods
Dogs were administered an intravenous bolus of ketamine (0.5 mg kg?1) followed by a ketamine CRI for 12 hours (20 μg kg?1 minute?1; treatment TC20 or 40 μg kg?1 minute?1; treatment TC40). Sedation, heart rate (HR), mean arterial pressure (MAP), electrocardiographic and echocardiographic parameters were evaluated at baseline (T0) and 1 (T1), 2 (T2), 4 (T4), 8 (T8), 12 (T12) and 24 (T24) hours after ketamine infusion started. Serum concentrations of CK-MB and troponin I were measured at baseline and 12, 24 and 48 hours after infusion started.Results
HR increased over the first 4 hours, significantly at T1 in TC20 and at T4 in TC40 when compared with T0 (p < 0.05). MAP was significantly increased at T2 in TC40 when compared with TC20. Behavioral changes, such as stereotypical head movements and twitches, occurred within 4 hours in TC40. There were no significant changes in echocardiographic examinations in any dog when compared with baseline. There were no temporal changes in serum CK-MB activity either within or between treatments (p > 0.05). No troponin I was detected in any sample.Conclusions and clinical relevance
No indication of myocardial injury resulting from ketamine infusion was detected in this study in healthy dogs. Further studies are needed to assess the ketamine infusion effects on antinociception and other organ function not evaluated in the present study. 相似文献45.

RAPD analysis was applied to reveal the genetic diversities of 4 speciesof subg. Lithocerasus within the genus Prunus using 40accessions representing the subgenera Prunophora, Amygdalus,Lithocerasus and Cerasus. The accession of subg. Lithocerasus are phenotypically similar to members of subgenus Cerasus but different with them in interspecific crossing tests and isozymeprofiles. In neurons, individual dendritic spines isolate N-methyl-d-aspartate (NMDA) receptor-mediated calcium ion (Ca2+) accumulations from the dendrite and other spines. However, the extent to which spines compartmentalize signaling events downstream of Ca2+ influx is not known. We combined two-photon fluorescence lifetime imaging with two-photon glutamate uncaging to image the activity of the small guanosine triphosphatase Ras after NMDA receptor activation at individual spines. Induction of long-term potentiation (LTP) triggered robust Ca2+-dependent Ras activation in single spines that decayed in approximately 5 minutes. Ras activity spread over approximately 10 micrometers of dendrite and invaded neighboring spines by diffusion. The spread of Ras-dependent signaling was necessary for the local regulation of the threshold for LTP induction. Thus, Ca2+-dependent synaptic signals can spread to couple multiple synapses on short stretches of dendrite. 相似文献

Most agriculturally important traits are regulated by genes known as quantitative trait loci (QTLs) derived from natural allelic variations. We here show that a QTL that increases grain productivity in rice, Gn1a, is a gene for cytokinin oxidase/dehydrogenase (OsCKX2), an enzyme that degrades the phytohormone cytokinin. Reduced expression of OsCKX2 causes cytokinin accumulation in inflorescence meristems and increases the number of reproductive organs, resulting in enhanced grain yield. QTL pyramiding to combine loci for grain number and plant height in the same genetic background generated lines exhibiting both beneficial traits. Hydrolysates obtained from porcine myofibrillar proteins by protease treatment (papain or actinase E) exhibited high antioxidant activity in a linolenic acid peroxidation system induced by Fe(2+). Hydrolysates produced by both papain and actinase E showed higher activities at pH 7.1 than at pH 5.4. The antioxidant activity of the papain hydrolysate was almost the same as that of vitamin E at pH 7.0. These hydrolysates possessed 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity and chelating activity toward metal ions. Antioxidant peptides were separated from the papain hydrolysate by ion exchange chromatography. The acidic fraction obtained by this method exhibited higher activity than the neutral or basic fractions. Antioxidant peptides in the acidic fraction were isolated by high-performance liquid chromatography on an ODS column and shown to possess the structures DSGVT, IEAEGE, DAQEKLE, EELDNALN, and VPSIDDQEELM. The DAQEKLE peptide showed the highest activity among these peptides. 相似文献
