A total of 1,734 cattle faecal samples from 296 dairy-keeping households were collected from urban settings in Nairobi, Kenya. Modified Ziehl–Neelsen staining method and an immunofluorescence assay were used to identify those samples with Cryptosporidium oocyst infection. Oocysts from positive faecal samples were isolated by Sheather's sucrose flotation method and picked from the concentrate using cover slips. Genomic DNA was extracted from 124 of the faecal samples that were positive for Cryptosporidium and was used as template for nested PCR of the 18S rRNA gene. Twenty-five samples (20 %) were PCR-positive for Cryptosporidium, and 24 of the PCR products were successfully cloned and sequenced. Sequence and phylogenetic analysis identified 17 samples (68 %) as Cryptosporidium parvum-like, four samples (16 %) as Cryptosporidium ryanae, three samples (12 %) as Cryptosporidium andersoni and one sample (4 %) as Cryptosporidium hominis. To the best of our knowledge, this is the first genotyping study to report C. parvum-like, C. andersoni and C. hominis in cattle from Kenya. The results of this study show Cryptosporidium infections in calves and cattle may be potential zoonotic reservoirs of the parasite that infects humans. 相似文献

The objective of this experiment was to determine if titanium dioxide (TiO2) dosed through an automated head chamber system (GreenFeed; C-Lock Inc., Rapid City, SD, USA) is an acceptable method to measure fecal output. The GreenFeed used on this experiment had a 2-hopper bait dispensing system, where hopper 1 contained alfalfa pellets marked with 1% titanium dioxide (TiO2) and hopper 2 contained unmarked alfalfa pellets. Eleven heifers (BW = 394 ± 18.7 kg) grazing a common pasture were stratified by BW and then randomized to either 1) dosed with TiO2-marked pellets by hand feeding (HFD; n = 6) or 2) dosed with TiO2-marked pellets by the GreenFeed (GFFD; n = 5) for 19 d. During the morning (0800), all heifers were offered a pelleted, high-CP supplement at 0.25% of BW in individual feeding stanchions. The HFD heifers also received 32 g of TiO2-marked pellets at morning feeding, whereas the GFFD heifers received 32 g of unmarked pellets. The GFFD heifers received a single aliquot (32 ± 1.6 g; mean ± SD) of marked pellets at their first visit to the GreenFeed each day with all subsequent 32-g aliquots providing unmarked pellets; HFD heifers received only unmarked pellets. Starting on d 15, fecal samples were collected via rectal grab at feeding and every 12 h for 5 d. A two-one sided t-test method was used to determine agreement and it was determined that the fecal output estimates by HFD and GFFD methods were similar (P = 0.04). There was a difference (P < 0.01; Bartlett’s test for homogenous variances) in variability between the dosing methods for HFD and GFFD (SD = 0.1 and 0.7, respectively). This difference in fecal output variability may have been due to variability of dosing times-of-day for the GFFD heifers (0615 ± 6.2 h) relative to the constant dosing time-of-day for HFD and constant 0800 and 2000 sampling times-of-day for all animals. This research has highlighted the potential for dosing cattle with an external marker through a GreenFeed configured with two (or more) feed hoppers because estimated fecal output means were similar; however, consideration of the increased variability of the fecal output estimates is needed for future experimental designs. 相似文献

Fast-growing woody species grown in dense, short-rotation plantations on land previously in agriculture offer potential economic benefits in products such as engineered construction material, boiler fuel, non-food-based biofuel feed stocks and other carbon (C)-based products and credits. However, information on the effects on major C pools of short-rotation culture is relatively sparse. In this study, Populus deltoides and P. deltoides × P. nigra hybrid clones were grown for 5 years at 1 m × 1 m spacing in plantations on a former pasture of high native fertility in the Missouri River floodplain in the lower Midwest U.S.A. Fire is a widespread natural disturbance agent in most conifer-dominated forests. In light of climate change and the effects of fire exclusion, single and repeated high-severity (stand-replacement) fires have become prominent land management issues. We studied bird communities using point counting in the Klamath-Siskiyou ecoregion of Oregon, USA at various points in time after one or two high-severity fires. Time points included 2 and 3 years after a single fire, 17 and 18 years after a single fire, 2 and 3 years after a repeat fire (15 year interval between fires), and >100 years since stand-replacement fire (mature/old-growth forest). Avian species richness did not differ significantly among habitats. Bird density was highest 17 and 18 years after fire, lowest 2 years after fire, and intermediate in repeat burns and unburned forest. Bird community composition varied significantly with habitat type (A = 0.24, P < 0.0001) with two distinct gradients in species composition relating to tree structure (live to dead) and shrub stature. Using indicator species analysis, repeat burns were characterized by shrub-nesting and ground-foraging bird species while unburned mature forests were characterized by conifer-nesting and foliage-gleaning species. Bird density was not related to snag basal area but was positively related to shrub height. Contrary to expectations, repeated high-severity fire did not reduce species richness, and bird densities were greater in repeat burns than in once-burned habitats. Broad-leaved hardwoods and shrubs appear to play a major role in structuring avian communities in the Klamath-Siskiyou region. In light of these results, extended periods of early seral broadleaf dominance and short-interval high-severity fires may be important to the conservation of avian biodiversity. 相似文献

Ecological invasions are a major driver of global environmental change. When invasions are frequent and prolonged, exotic species can become dominant and ultimately create novel ecosystem types. These ecosystems are now widespread globally. Recent evidence from Puerto Rico suggests that exotic-dominated forests can provide suitable regeneration sites for native species and promote native species abundance, but this pattern has been little explored elsewhere. We surveyed 46 sites in Hawai’i to determine whether native species occurred in the understories of exotic-dominated forests. Native trees smaller than 10 cm in diameter were absent in 28 of the 46 sites and rare in the others. Natives were never the dominant understory species; in fact, they accounted for less than 10% of understory basal area at all but six sites, and less than 4% on average. Sites with native species in the understory tended to be on young lava substrate lacking human disturbance, and were mostly located close to intact, native-dominated forest stands. Even where we found some native species, however, most were survivors of past exotic encroachment into native forest, rather than products of active recolonization by native species. In contrast with successional trajectories in Puerto Rico, Hawaii's exotic-dominated forests can emerge, via invasion, without human disturbance and native Hawaiian plants are largely unable to colonize them once they appear. We suggest that a wide diversity of growth strategies among the exotic species on Hawai’i may limit the opportunities for native plants to colonize exotic-dominated forests. 相似文献

In the context of forest management, a fixed harvesting policy consists in trying to convert stands of trees to a chosen state at fixed intervals, regardless of the stand state and of the state of the market. In an adaptive policy, instead, the post-harvest state and the timing of the harvest depend on the stand and market states at the time of the decision. The objective of this study was to determine the practical gain from the theoretically superior adaptive policies. To this end, we compared optimal fixed and adaptive policies obtained with identical models and assumptions, and with data from the Douglas-fir/western-hemlock forests in the Pacific Northwest of the United States. In maximizing economic returns from harvests over an infinite time horizon, the net present value was 17 percent higher with an adaptive than with a fixed policy. It was 22 percent higher when the objective was to maximize annual harvest. The adaptive policy was even more superior with undiscounted, non-economic objectives, such as the area of spotted owl habitat (+37 percent gain), or the area of late-seral forest (+51 percent), but less so in maximizing the stock of high quality logs (+6 percent). The two-grooves method used for estimating the surface locked-in strains on standing stems was studied using a finite-element
model simulating the gauge measurement resulting from the groove cutting, having in mind the particular of small diameter
trees. The assumed growth stress distribution was described by simple polynomial expressions of the relative radius with the
possible existence of a tension wood sector characterized by higher residual stresses in the longitudinal direction. The end
effect of the crosscutting vanished for a height/diameter ratio higher than 3 and the simulated gauge measurement reasonably
approached the local growth strain average. For very small trees with a diameter of about 2 cm, the distance between gauge
end and groove should not exceed 3 mm, a value of up to 5 mm is allowed in case of larger stems, 5–20 cm. For any combination
of stem diameter, gauge length, groove distance and depth, and assumptions on the internal stress distributions, the underestimation

Basmati rice is highly susceptible to bacterial blight (BB) caused by Xanthomonas oryzae pv. oryzae. Transfer of BB resistance genes from non‐Basmati sources to Basmati through cross‐hybridization requires strict monitoring for recovery of the desirable Basmati quality traits in the recombinants, which show complex inheritance pattern. We integrated background analysis using mapped microsatellite markers with foreground selection to identify superior lines that combine useful genes from a non‐Basmati BB resistance donor line IRBB55 with grain and cooking quality characteristics of the popular Basmati rice variety ‘Pusa Basmati 1’ (PB 1) employing backcross pedigree strategy. Foreground selection using linked markers ensured presence of two genes, xa13 and Xa21 for BB resistance from IRBB55, and the recurrent parent PB 1 allele for the waxy locus giving intermediate amylose content and maintainer allele at fertility restorer locus in the BC1F5 recombinants. Background analysis enabled selection of recombinants with recurrent parent genome to the extent of 86.3% along with the quality traits. The extent of introgression of non‐Basmati donor chromosome segments in the superior selections was estimated to be < 7.8 Mb and < 6.7 Mb in the xa13 and Xa21 linked genomic regions, respectively. Association mapping identified three quantitative trait loci, one each for 1000‐grain weight, fertile grains/panicle and cooked kernel length. The backcross‐pedigree breeding strategy facilitated recovery of additional desirable characteristics from the donor in some of the selections. The elite selection Pusa 1460‐01‐32‐6‐7‐67 with maximum genomic background and quality characteristics of the recurrent Basmati parent gave resistance reaction against BB, similar to that of the non‐Basmati resistant check variety and recorded an yield advantage of 11.9% over the best check in the multiplication agronomic trial in the Basmati growing region of India. This line, which has been released as a new variety in the name of ‘Improved Pusa Basmati 1’ for commercial cultivation in India, is an example of successful application of marker assisted selection to variety development. 相似文献
