Several species of fungi, which infect cereals and grains, can produce a class of compounds, known as trichothecene mycotoxins, which is characterized by a substituted epoxy-trichothecene ring structure. Cattle are susceptible to intoxication from feeds contaminated with T-2 toxin, one of the more potent trichothecene mycotoxins, while swine refuse to ingest feed contaminated with T-2 toxin. The bovine platelet has been used as a model cell system to evaluate the effects of T-2 toxin and its natural metabolites, HT-2 toxin and T-2 tetraol, on cell function in vitro. Due to the lipophilic nature of these mycotoxins, a biologically active phospholipid was used to stimulate the platelets in the presence and absence of the toxins. The mycotoxin T-2 toxin and its major metabolite HT-2 toxin inhibited platelet activating factor-stimulated bovine platelets, suspended in homologous plasma, in a concentration but not time dependent manner. Significant inhibition of platelet function (p less than 0.01) occurred with 135 ng T-2 toxin per 10(6) platelets and with 77 ng HT-2 toxin per 10(6) platelets. These mycotoxins exerted an additive inhibitory effect on the platelet aggregation response. In contrast, the minor metabolite T-2 tetraol had no inhibitory effect on platelet function and had no influence on the responses of T-2 toxin or HT-2 toxin when the mycotoxins were present together in the platelet suspensions.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

A prospective study was designed to document the frequencies of equine health problems in the state of Michigan, USA. A total of 2469 horses from a random sample of 138 equine operations were monitored in the study in two 12-month periods: 1992–1993 and 1993–1994. All the major breeds of horses in the state were proportionately represented in the sample. Using weighted annual incidence densities as measures of disease frequencies, the 10 most frequently observed groups of health problems were (from most to least frequent) leg lameness, dermatological problems, respiratory problems, hoof and foot problems, reproductive problems, systemic problems, colic, whole body lameness, neurological problems and gastrointestinal problems (other than colic). This ranking of the top 10 health problems was different from the ranking provided by equine owners/operators at the beginning of the study.

Overall, very low specific mortality rates were observed in the study. However, the conditions that were associated with mortality resulted in fairly high case fatalities. The five specific illnesses that most commonly resulted in fatalities were (from greatest to least case fatality risk) systemic problems, colic, gastrointestinal problems (other than colic), neurological problems, and foot and hoof problems.

Impact of disease was evaluated in terms of average duration of a case and days lost for performance. Neurological problems, lameness and dermatological problems had the longest duration per case. Neurological problems, lameness, respiratory problems and gastrointestinal problems (other than colic) had the longest average days lost per case.  相似文献

Factor XI (F XI) is a plasma protein that participates in the blood coagulation process. A study of the expression of F XI activity in Holstein cattle has confirmed that the inheritance of F XI deficiency is autosomal with severe deficiency in homozygotes (mean F XI level 2%, SD 1%), and partial deficiency in heterozygotes (mean F XI level 38%, SD 10%; normal mean F XI level 94%, SD 21%). In a total of 1469 males evaluated for F XI levels, 47 or 3.1% were identified as heterozygous and only one as homozygous for the disorder. In part because of the lack of a discrete distinction in the expression of F XI between heterozygous and normal animals, not all of the animals tested could be uniquely classified on the basis of the plasma F XI values. A mean F XI value of 53% (SD 7%) was found in a group of animals that were categorized as low normal/high heterozygous. If this group of cattle had been classified on the basis of the criterion used to classify human beings then these animals would have been categorized as heterozygous since the mean F XI value for proven bovine heterozygotes is approximately 20% lower than the values found in the human counterpart. Like the human form of the disease, however, there appears to be a low frequency of hemorrhagic episodes associated with F XI deficiency in cattle.  相似文献

An immunoblot procedure was used to evaluate porcine antibody response to inoculation with Mycoplasma hyopneumoniae. Mycoplasmas solubilized with sodium dodecyl sulfate were used as antigens. Antibodies to 5 antigens, estimated to be of molecular weight (mol wt) 110,000, 64,000, 50,000, 41,000, and 36,000, were detected in sera collected during the course of induced mycoplasmal pneumonia. Mycoplasma hyopneumoniae antigens, mol wt 110,000, 50,000, 41,000, and 36,000, cross-reacted with M flocculare when antigen prepared from M flocculare or hyperimmune serum against it were used in the immunoblot procedure. The 36,000-dalton (D) antigen reacted with M hyopneumoniae and M hyorhinis convalescent sera. The 64,000-D M hyopneumoniae antigen was the only antigen that did not cross-react with M flocculare or M hyorhinis. Exposure of immunoblot strips with antigens to trypsin before reacting them with the convalescent sera abolished binding ability of the 110,000-D and 36,000-D antigens, but had no effect on binding by 64,000-D, 50,000-D, or 41,000-D antigens. None of the 5 antigens bound to 11 lectins.  相似文献

OBJECTIVE: To identify scintigraphic abnormalities in the pelvic region of horses examined because of hind limb lameness or poor performance and determine the clinical relevance of areas of abnormal radiopharmaceutical uptake (ARU) in these horses. DESIGN: Retrospective study. ANIMALS: 128 horses. PROCEDURE: Medical records were reviewed, and information on signalment, history, admitting complaints, physical examination findings, and results of lameness examinations was recorded. Clinical relevance of areas of ARU was determined by comparison with results of other diagnostic tests. For horses with clinically relevant areas of ARU, follow-up information was obtained through telephone interviews with owners and trainers and analysis of race records. RESULTS: Areas of ARU were identified in the tuber coxae (25 horses), ischiatic tuber (9), hip joint (10), third trochanter (10), ilium (5), sacral tuber region (22), greater trochanter (1), cranial femoral cortex (1), skeletal muscle surrounding the pelvis (34), or multiple areas (11). In 44 horses, areas of ARU were associated with the primary cause of lameness; in 51, areas of ARU were not associated with the primary cause of lameness; and in 33, the primary cause of lameness was not determined. Thirty-six of the 44 horses with clinically relevant areas of ARU were available for follow-up; 15 (42%) had a good outcome. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that pelvic scintigraphy may be useful in identifying abnormalities in horses with hind limb lameness or poor performance.  相似文献

OBJECTIVE: To report the outcome of surgical treatment of comminuted fractures of the proximal phalanx in horses. DESIGN: Retrospective study. ANIMALS: 64 horses. PROCEDURE: Medical records and radiographs were reviewed to obtain information regarding signalment, fracture classification, and treatment. Follow-up information was obtained by telephone conversation or evaluation of production records. RESULTS: Thirty-eight horses had moderately comminuted fractures of the proximal phalanx. Two horses were euthanatized immediately. Fractures of the proximal phalanx in 36 horses were repaired with open reduction and internal fixation with a successful outcome in 33 (92%) horses. Reconstruction of the fracture was performed in most horses by use of a long curved incision, transection of the collateral ligament of the metacarpophalangeal or metatarsophalangeal joint, and open exposure of the proximal articular surface of the proximal phalanx. Twenty-six horses had severely comminuted fractures of the proximal phalanx. Six horses were euthanatized immediately. One horse was euthanatized after 9 days of treatment with a cast alone. Severely comminuted fractures of the proximal phalanx in 13 horses were treated with an external skeletal fixation device, and fractures healed in 8 of those horses. Six horses with severely comminuted fractures of the proximal phalanx were treated with transfixation pins incorporated into a fiberglass cast, and fractures healed in 4 horses. CONCLUSIONS AND CLINICAL RELEVANCE: Moderately comminuted fractures of the proximal phalanx can be successfully repaired; however, fractures that are too severe to permit accurate reconstruction of the fragments remain difficult to treat and horses have only a fair prognosis for survival.  相似文献

: Public concerns over the widespread prophylactic use of antibiotics have led to a search for alternatives to dry cow therapy for the prevention of intramammary infections. A popular alternative is to infuse a teat seal at drying-off. The teat seal is a viscous non-antibiotic formulation and when it is infused into the teat canal and the teat sinus it forms an internal seal that provides a physical barrier to invasion by mastitis-causing pathogens. Enhancement of teat seal formulations may be achieved using non-antibiotic additives such as bacteriocins, potent proteins produced by some bacteria that have the ability to kill other microorganisms. This paper traces the history of investigations at Moorepark Research Centre into the efficacy of teat seal plus lacticin 3147, a bacteriocin produced by Lactococcus lactis DPC3147, in the prevention of intramammary infections in dry cows. Indications from on-going investigations are that a dry cow formulation combining the two products has considerable potential as a non-antibiotic prophylactic product.  相似文献
