Diffusible signals, not autonomous mechanisms, determine the main proximodistal limb subdivision 总被引:1,自引:0,他引:1
Vertebrate limbs develop three main proximodistal (PD) segments (upper arm, forearm, and hand) in a proximal-to-distal sequence. Despite extensive research into limb development, whether PD specification occurs through nonautonomous or autonomous mechanisms is not resolved. Heterotopic transplantation of intact and recombinant chicken limb buds identifies signals in the embryo trunk that proximalize distal limb cells to generate a complete PD axis. In these transplants, retinoic acid induces proximalization, which is counteracted by fibroblast growth factors from the distal limb bud; these related actions suggest that the first limb-bud PD regionalization results from the balance between proximal and distal signals. The plasticity of limb progenitor cell identity in response to diffusible signals provides a unifying view of PD patterning during vertebrate limb development and regeneration. 相似文献

Several methods are used to measure lipid peroxidation (LPO) in spermatozoa. The objective of this study was comparing the thiobarbituric acid reactive species (TBARS) method and the BODIPY 581/591 C11 (B581) and BODIPY 665/676 C11 (B665) fluorescent probes to measure induced peroxidative damage in thawed epididymal spermatozoa from Iberian red deer. Samples from three males were thawed, pooled, diluted in PBS, incubated at room temperature and assessed at 0, 3, 6 and 24 h under different experimental conditions: Control, hydrogen peroxide (H2O2) 0.1 mm or 1 mm , or tert‐butyl hydroperoxide (TBH) 0.1 mm or 1 mm . LPO was assessed by the TBARS assay [malondialdehyde (MDA) detection] and by the fluorescence probes B581 and B665 (microplate fluorimeter and flow cytometry). Increasing MDA levels were only detectable at 1 mm of TBH or H2O2. Both fluorescence probes, measured with fluorometer, detected significant increases of LPO with time in all treatments, except Control. Flow cytometry allowed for higher sensitivity, with both probes showing a significant linear relationship of increasing LPO with time for all oxidizing treatments (p < 0.001). All methods showed a good agreement, except TBARS, and flow cytometry showed the highest repeatability. Our results show that both B581 and B665 might be used for LPO analysis in Iberian red deer epididymal spermatozoa, together with fluorometry or flow cytometry. Yet, the TBARS method offered comparatively limited sensitivity, and further research must determine the source of that limitation. 相似文献

A method has been developed for simultaneously determining alpha-tocopherol and cholesterol in fresh pig meat by HPLC. It allows a reduction in the number of analyses and brings savings in time and materials. The unsaponifiable fraction is extracted following the modified method of Liu et al. (Liu, Q.; Scheller, K. K.; Schaefer, D. M. Technical note: A simplified procedure for vitamin E determination in beef muscle. J. Anim. Sci. 1996, 74, 2406-2410). The modifications introduced are the use of nitrogen atmosphere during the extraction, the addition of an antioxidant in the organic extraction phase, and the use of alpha-tocopherol itself as an internal standard. There is then a chromatographic analysis which allows the separation of the two compounds in question. To identify and quantify, two different detectors are used in series: the first is a fluorescence detector (alpha-tocopherol), and the second is a light-scattering detector (cholesterol). The technique shows sufficient sensitivity to determine the normal levels of alpha-tocopherol and cholesterol in meat, with recovery percentages of 78% and 97%, respectively. The average amount of alpha-tocopherol and cholesterol in samples from pig Longissimus dorsi muscle analyzed using this method is 1.8 and 620 mg/kg of fresh meat, respectively. 相似文献

An optimized multiplex RT-PCR assay was developed to discriminate three necrovirus (Olive latent virus 1 (OLV-1), Tobacco necrosis virus D (TNV-D) and Olive mild mosaic virus (OMMV)) that infect olive trees. An olive orchard consisting of 54 trees of cv. “Galega vulgar” in the south of Portugal
was surveyed. dsRNA fraction was used as template and revealed the 3 viruses, singly or in multiple infections, present in
17 out of 54 trees in the orchard. OMMV was the most frequent occurring in 15 trees, followed by OLV-1 in 12 and TNV-D in
4 plants. The results obtained showed that necrovirus- specific dsRNAs do exist in infected tissues in amounts below the resolution
permitted by gel electrophoresis analysis and that the developed multiplex PCR based assay is of much higher sensitivity.
The design of the specific primers described enabled, for the first time, to discriminate between OMMV and TNV-D by means

(-)-Epicatechin conjugates with sulfur-containing moieties are strong free radical scavengers with cell-protecting activities, which may be in part modulated by their capacity to bind to biological membranes. We present here a study of the interaction of these conjugates with membrane models such as multilamellar vesicles and a phospholipid-coated silica column (immobilized artificial membrane), monitored by differential scanning calorimetry and high-performance liquid chromatography, respectively. The nonpolyphenolic moiety significantly influenced the membrane behavior of the whole molecules. Bulky and hydrophobic conjugates clearly interacted with the phospholipids and may have a tendency to penetrate into the hydrophobic core of the vesicles. In contrast, the smaller cationic 4beta-(2-aminoethylthio)epicatechin may be located at the outer interface of the lipid membrane. The outcomes from both experimental set-ups were in good agreement. The differences detected in the biological activities of the conjugates may be explained in part by their tendency to penetrate the cell membrane. 相似文献

This paper reports preliminary studies designed to evaluate hybridization through natural mating and artificial insemination between female Penaeus japonicus Bate, 1888 and male Penaeus kerathurus Forskäl, 1775. Although both species were able to mature in captivity, natural copulation was not achieved. Different techniques, including manual squeezing in the base of the fifth pair of pereiopods, electrical stimulation and dissection, were used for the artificial insemination experiments. However, only dissection gave spermatophores. Stained sperm showed no malformations and its viability was very high. Seven out of 10 inseminations (70%) produced eggs, but the development of embryos was abnormal. It is suggested that the high genetic distance between these species is responsible for these results. 相似文献

An industrial baking procedure for yeast-leavened whole-grain rye crisp bread was adapted to local laboratory conditions to study the effect of time and temperature of baking and the addition of fructose, asparagine, and oat-bran concentrate on the acrylamide content and color of the bread. Baking time and temperature affected acrylamide content that increased from 10 to 30 mug/kg of bread at the combination of a long time and high temperature, with a significant interaction between the two factors (p < 0.008). Added asparagine had a significant effect (p < 0.001) on the formation of acrylamide, but fructose did not. There was a correlation between acrylamide content and color of the milled bread in the time-temperature experiment, but this correlation was not observed in the experiment with added precursors. Added oat-bran concentrate with high content of mixed-linkage beta-glucan did not influence the acrylamide content in the breads. 相似文献

Moringa oleifera (MO) is a multipurpose, medium- or small-sized tree, from regions of north-west India and indigenous to many parts of Asia, Africa, and South America. Its pods have been employed as an inexpensive and effective sorbent for the removal of organics, and coagulant for water treatment. It is a non-toxic natural organic polymer. The main objective of this work was to use the MO seeds as a natural adsorbent for the treatment of dairy industry wastewater (DIW). The effects of agitation time, pH, MO biomass dose, and DIW concentration were evaluated. Removal efficiencies of up to 98%, for both color and turbidity, were reached using 0.2 g MO and 0.2 L of 1.0 g/L sorbate solution (DIW). The obtained results showed that MO seed keeps its adsorption power under a pH range between 5 and 8. The adsorption data was fitted to Langmuir isotherm. There was a significant uptake capacity of MO biomass, q max, which suggested a good affinity between DIW components and sorbent. We conclude that the MO biomass has the potential to be used in the dairy industry wastewater treatment in an efficient way and with low cost. 相似文献

The most distinctive variation in the monomer composition of lignins in vascular land plants is that between the two main groups of seed plants. Thus, whereas gymnosperm (softwood) lignins are typically composed of guaiacyl (G) units, angiosperm (hardwood) lignins are largely composed of similar levels of G and syringyl (S) units. However, there are some studies that suggest that certain angiosperm peroxidases are unable to oxidize sinapyl alcohol, and a coniferyl alcohol shuttle has been proposed for oxidizing S units during the biosynthesis of lignins. With this in mind, a screening of the presence of S peroxidases in angiosperms (including woody species and forages) was performed. Contrarily to what might be expected, the intercellular washing fluids from lignifying tissues of 25 woody, herbaceous, and shrub species, belonging to both monocots and dicotyledons, all showed both S peroxidase activities and basic peroxidase isoenzymes analogous, with regard the isoelectric point, to the Zinnia elegans basic peroxidase isoenzyme, the only S peroxidase that has been fully characterized. These results led to the protein database in the search for homologies between angiosperm peroxidases and a true eudicot S peroxidase, the Z. elegans peroxidase. The findings showed that certain structural motifs of S peroxidases are conserved within the first 15 million years of angiosperm history, because they are found in peroxidases from the two major lineages of flowering plants, eumagnoliids and eudicotyledons, of note being the presence of these peroxidases in Amborella and Nymphaeales, which represent the first stages of angiosperm evolution. These phylogenetic studies also suggest that guaiacyl peroxidases apparently constitute the most "evolved state" of the plant peroxidase family evolution. 相似文献

The aim of this work is to assess the influence of light intensity (plants with direct sun exposure and in a controlled light intensity) and age of leaves (6-24 months) on the characteristics of the extracts of mate tea leaves obtained from carbon dioxide at high pressures. Samples of mate were collected in an experiment conducted under agronomic control at Industria e Comercio de Erva-Mate Bar?o LTDA, Brazil. The content of selected organic compounds of the extracts was evaluated by gas chromatography together with mass spectrometry. Quantitative analysis of caffeine, theobromine, phytol, vitamin E, squalene, and stigmasterol was performed, and the results showed that field variables exert a strong influence on the liquid yield and on the chemical distribution of the extracts. 相似文献
