A computer algorithm was created to analyze and quantify scanned images from DNA microarray slides developed for detecting pathogenic Escherichia coli isolates recovered from agricultural food products. The algorithm computed centroid locations for signal and background pixel intensities in RGB space and defined a plane perpendicular to the line connecting the centroids as a decision boundary. The algorithm was tested on 1534 potential spot locations which were visually classified depending on the strength of the signal. Three other standard measures of SNR (SSR, SBR, and SSDR) were also performed for each potential spot location. The number of errors as compared to visual classifications was computed for each of the four measures. SSR and SSDR, which depend on pixel intensity standard deviations, performed poorly with high false positive results, while the current algorithm and SBR, which were independent of standard deviations, performed much better. Overall error rates were 1.4% for the reported algorithm, 2.0% for SBR, 14.2% for SSDR, and 16.8% for SSR.  相似文献

Pet and food animal (hogs, chicken, and fish) feeds were recently found to be contaminated with melamine (MEL). A quantitative and confirmatory method is presented to determine MEL residues in edible tissues from fish fed this contaminant. Edible tissues were extracted with acidic acetonitrile, defatted with dichloromethane, and cleaned up using mixed-mode cation exchange solid-phase extraction cartridges. Extracts were analyzed by liquid chromatography with tandem mass spectrometry with hydrophilic interaction chromatography and electrospray ionization in positive ion mode. Fish and shrimp tissues were fortified with 10-500 microg/kg (ppb) of MEL with an average recovery of 63.8% (21.5% relative standard deviation, n = 121). Incurred fish tissues were generated by feeding fish up to 400 mg/kg of MEL or a combination of MEL and the related triazine cyanuric acid (CYA). MEL and CYA are known to form an insoluble complex in the kidneys, which may lead to renal failure. Fifty-five treated catfish, trout, tilapia, and salmon were analyzed after withdrawal times of 1-14 days. MEL residues were found in edible tissues from all of the fish with concentrations ranging from 0.011 to 210 mg/kg (ppm). Incurred shrimp and a survey of market seafood products were also analyzed as part of this study.  相似文献

Three tasty (BR-139, FA-624, and FA-612) and two less tasty (R-144 and R-175) fresh greenhouse tomato cultivars, which significantly differ in their flavor profiles, were screened for potent odorants using aroma extract dilution analysis (AEDA). On the basis of AEDA results, 19 volatiles were selected for quantification in those 5 cultivars using gas chromatography-mass spectrometry (GC-MS). Compounds such as 1-penten-3-one, ( E, E)- and ( E, Z)-2,4-decadienal, and 4-hydroxy-2,5-dimethyl-3(2 H)-furanone (Furaneol) had higher odor units in the more preferred cultivars, whereas methional, phenylacetaldehyde, 2-phenylethanol, or 2-isobutylthiazole had higher odor units in the less preferred cultivars. Simulation of the odor of the selected tomato cultivars by preparation of aroma models and comparison with the corresponding real samples confirmed that all important fresh tomato odorants were identified, that their concentrations were determined correctly in all five cultivars, and that differences in concentration, especially of the compounds mentioned above, make it possible to distinguish between them and are responsible for the differential preference. To help elucidate formation pathways of key odorants, labeled precursors were added to tomatoes. Biogenesis of cis- and trans-4,5-epoxy-( E)-2-decenals from linoleic acid and methional from methionine was confirmed.  相似文献

Insecticides are the most commonly used tactic to control western flower thrips (WFT), Frankliniella occidentalis Pergande (Thysanoptera: Thripidae), on greenhouse cucumber. However, WFT has developed resistance to several of the insecticides presently in use. In addition, some of these insecticides adversely affect greenhouse biological control agents used to control WFT, resulting in subsequent pest resurgence. Therefore, there is a need to identify novel insecticides with unique modes of action for use in integrated pest management (IPM) programs to effectively control WFT with minimal impact on associated biological control agents. In laboratory bioassays conducted in 2001, immature and adult WFT and three associated greenhouse biological control agents: Amblyseius cucumeris Oudemans (Acarina: Phytoseiidae), Orius insidiosus Say (Hemiptera: Anthocoridae) and Encarsia formosa Gahan (Hymenoptera: Aphelinidae) were exposed to direct, direct/residual, and residual contact applications of the novel biopesticide, spinosad (Conserve 120 SC), and the industry standard for whitefly control, endosulfan (Thiodan 50 WP). In all three types of assay, spinosad was effective against immature and adult WFT life stages. It showed low toxicity to A. cucumeris, moderate toxicity to O. insidiosus and high toxicity to E formosa. Greenhouse studies involving exposure of immature and adult WFT and adult biological control agents to cucumber leaves sprayed previously with spinosad supported the laboratory data. Spinosad showed low toxicity to A. cucumeris exposed to leaves 1 day after treatment (DAT), moderate toxicity to O. insidiosus 1 and 8 DAT, and high toxicity to E. formosa up to 28 DAT. These data, along with spinosad's unique mode of action, suggest it would be a valuable reduced-risk control agent for greenhouse cucumber IPM programs.  相似文献

This report describes the use of computed tomography (CT) to evaluate the tympanic bullae and angular processes of the mandible in a West Highland White Terrier with craniomandibular osteopathy. The patient presented initially for swelling of the right forelimb. Pea ( Pisum sativum L.) is an important world grain legume and vegetable in the south of Europe where it is grown in small farms and gardens during the cool season. Most of the pea production of this area occurs in high nitrogen (N) soils. Inoculation is required to increase yields through N fixation and reduce the external inputs. Three pea elite cultivars representing human food market classes (green, sugar and dry peas) were studied in order to characterize the cultivars' variability in symbiotic characters using two Rhizobium leguminosarum biovar viceae commercial strains and the indigenous soil strain, and three N-fertilization rates. Significant differences were observed among pea elite cultivars for the weight and length of plant parts, and the traits associated with N fixation. Pea cultivars showed a significant cultivar–strain interaction for shoot fresh weight and significant differences among strains were found for nodule fresh weight. Green and sugar pea cultivars had good N fixation with a particular commercial strain, while the dry pea cultivar did not show an effective symbiotic combination. The differences between the pea cultivars could be exploited in breeding programs for enhanced nodulation and N fixation in each pea market class.  相似文献

Ecosystem models typically use input temperature and precipitation data generated stochastically from weather station means and variances. Although the weather station data are based on measurements taken over a few decades, model simulations are usually on the order of centuries. Consequently, observed periodicities in temperature and precipitation at the continental scale that have been correlated with largscale forcings, such as ocean-atmosphere dynamics and lunar and sunspot cycles, are ignored. We investigated how these natural climatic fluctuations affect aboveground biomass in ecosystem models by incorporating some of the more pronounced continental-scale cycles in temperature (4, 11, 80, 180 year periods) and precipitation (11 and 19 year periods) into models of three North American forests (using LINKAGES) and one North American grassland (using STEPPE). Even without inclusion of periodicities in climate, long-term dynamics of these models were characterized by internal frequencies resulting from vegetation birth, growth and death processes. Our results indicate that long-term temperature cycles result in significantly lower predictions of forest biomass than observed in the control case for a forest on a biome transition (northern hardwoods/boreal forest). Lower-frequency, higher-amplitude temperature oscillation caused amplification of forest biomass response in forests containing hardwood species. Shortgrass prairie and boreal ecosystems, dominated by species with broad stress tolerance ranges, were relatively insensitive to climatic oscillations. Our results suggest periodicities in climate should be incorporated within long-term simulations of ecosystems with strong internal frequencies, particularly for systems on biome transitions.  相似文献

Although stifle disease is seen less frequently in cats than in dogs, accurate diagnosis and proper treatment are important to restoring joint function. OBJECTIVES: To compare the expression of canine relaxin, relaxin-like factor (RLF), and relaxin receptors within the muscles of the pelvic diaphragm of dogs with perineal hernia (PH) and clinically normal dogs. STUDY DESIGN: In vivo comparative study. ANIMALS: Fifteen client-owned intact male dogs with PH were studied. Four mature intact male dogs with no evidence of perineal pathology served as controls. METHODS: Biopsy samples from the levator ani, coccygeus, and internal obturator muscles were obtained. RNA samples were reverse transcribed and analyzed by real-time PCR for the expression of canine relaxin receptor LRG7, relaxin, and RLF. RESULTS: Significantly higher expression levels of canine relaxin receptors occurred in the musculature of the pelvic diaphragm and internal obturator muscle in dogs with PH compared with normal dogs. Expression of canine RLF revealed no significant difference between dogs with PH and controls. The difference in the expression of canine relaxin between groups was not statistically significant. CONCLUSIONS: Relaxin receptor up-regulation occurs in the coccygeus, levator ani, and internal obturator muscles of dogs with PH. CLINICAL RELEVANCE: The higher expression of relaxin receptors within the muscles of the pelvic diaphragm in dogs with PH suggests that relaxin might play a role in the pathogenesis of PH. Atrophy of these muscles, which predisposes to PH, may be attributable to increased relaxin activity.  相似文献

The purpose of the study was to identify factors influencing the outcome and prognosis of rupture of the tendon of the peroneus tertius muscle in 27 horses. Information on history, physical examination findings, diagnosis, treatment, and final outcome was summarized from medical records. Long-term follow-up information on horses was obtained by telephone survey. A stepwise logistic regression model was used to analyze factors influencing the outcome. Rupture occurred in the midbody of the tendon in 11 horses, at the insertion site in 11 horses, and at the origin in 2 horses. Overall, 18/23 (78.3%) horses returned to their previous level of exercise, 5/23 (21.7%) horses were euthanized due to persistent lameness. If the horse was racing at the time of injury or had an additional structure injured besides the peroneus tertius tendon, it was less likely to return to its intended use.  相似文献
