The effect of low salinity on survival and growth of the Pacific white shrimp Litopenaeus vannamei was examined in the laboratory due to the interest of raising shrimp inland at low salinities. In three separate experiments, individual L. vannamei postlarvae (∼ 0.1 g) were cultured at salinities of either 0.5, 1, 1.5, 2, or 3 ppt ( N = 5 or 10/treatment) for 18 to 40 d at 30 C in individual 360-mL containers. In each experiment controls of 0 and 30 ppt were run. There was no postlarval survival at salinities < 2 ppt. Survival was significantly different ( P < 0.01) at 2 ppt (20%) compared to 30 ppt (80%). Growth was also significantly different ( P < 0.01) at 2 and 3 ppt compared to 30 ppt (416%, 475%, and 670%, respectively). A fourth experiment compared juveniles (∼ 8 g) and postlarvae (∼ 0.05 and 0.35 g). Shrimp were cultured at salinities of 0, 2, 4, and 30 ppt for 40 d at 25 C, in individual 360-mL and 6-L containers ( N = 7/treatment). There was no postlarval survival at < 2 ppt. Postlarval survival at 4 ppt (86%) was not significantly different (P > 0.05) from 30 ppt (100%). Juveniles exhibited better survival at lower salinities (100% at 2 ppt) than 0.05 and 0.35 g postlarvae (29% and 14% respectively, at 2 ppt). The effects of salinity on growth varied with sizdage. Final growth of 0.05 g postlarvae at 2 ppt (693%) was significantly less ( P < 0.01) than at 4 ppt (1085%) and 30 ppt (1064%). Growth of 0.35 g postlarvae was significantly less ( P < 0.01) for 4 ppt (175%) than for 30 ppt (264%). There was no growth data for juveniles (8 g). It appears from these experiments that the culture of L. vannamei poses risks when performed in salinities less than 2 ppt.  相似文献

Continuation of mammalian species requires the formation and development of the sexually dimorphic germ cells. Cultured embryonic stem cells are generally considered pluripotent rather than totipotent because of the failure to detect germline cells under differentiating conditions. Here we show that mouse embryonic stem cells in culture can develop into oogonia that enter meiosis, recruit adjacent cells to form follicle-like structures, and later develop into blastocysts. Oogenesis in culture should contribute to various areas, including nuclear transfer and manipulation of the germ line, and advance studies on fertility treatment and germ and somatic cell interaction and differentiation.  相似文献

This study assessed the apoptotic process occurring in the hemocytes of the Pacific oyster, Crassostrea gigas, exposed to Alexandrium catenella, a paralytic shellfish toxins (PSTs) producer. Oysters were experimentally exposed during 48 h to the toxic algae. PSTs accumulation, the expression of 12 key apoptotic-related genes, as well as the variation of the number of hemocytes in apoptosis was measured at time intervals during the experiment. Results show a significant increase of the number of hemocytes in apoptosis after 29 h of exposure. Two pro-apoptotic genes (Bax and Bax-like) implicated in the mitochondrial pathway were significantly upregulated at 21 h followed by the overexpression of two caspase executor genes (caspase-3 and caspase-7) at 29 h, suggesting that the intrinsic pathway was activated. No modulation of the expression of genes implicated in the cell signaling Fas-Associated protein with Death Domain (FADD) and initiation-phase (caspase-2) was observed, suggesting that only the extrinsic pathway was not activated. Moreover, the clear time-dependent upregulation of five (Bcl2, BI-1, IAP1, IAP7B and Hsp70) inhibitors of apoptosis-related genes associated with the return to the initial number of hemocytes in apoptosis at 48 h of exposure suggests the involvement of strong regulatory mechanisms of apoptosis occurring in the hemocytes of the Pacific oyster.  相似文献

The current study examined the effects of inclusion of flaxseed in a red clover silage diet on growth, carcass quality, adipose tissue and muscle fatty acid (FA) profiles of beef steers. Twelve British×Continental crossbred steers (363±26.5 kg) were randomly assigned to two pens and fed diets containing 70% red clover silage, 15% steam rolled barley/vitamin–mineral supplement and either additional 15% steam rolled barley or 15% triple rolled flaxseed (all DM basis) for 215 d. Diet had no effect (P>0.05) on average daily gain or carcass quality atributes. Overall, FA profiles in all tissues followed similar trends when flaxseed was added to the diet but intramuscular fat had the highest proportions of n-3 and n-6 polyunsaturated FA (PUFA) while subcutaneous fat had largest proportions of conjugated linoleic acids (CLA) and perirenal fat had the highest proportions of trans-18:1 isomers, particularly vaccenic acid (VA). Feeding flaxseed increased (P<0.05) percentages of α-linolenic acid (ALA; 1.1–1.6% of total FA), total n-3 PUFA, conjugated linolenic acids, non-conjugated linoleic acids (mostly t11,c15-18:2), and led to unprecedented increases in CLA (mainly rumenic acid, RA; 1.1–2.9%), and trans-18:1 isomers (chiefly VA; 5.9–9.5%) in muscle and adipose tissues. Flaxseed also reduced individual and total n-6 PUFA, cis-monounsaturated FA, branched-chain FA and saturated FA (mainly 16:0) in beef lipids. Overall, feeding flaxseed in a red clover silage diet enhanced the levels of FA with a positive health image (ALA, 2-fold; RA, 3 to 4-fold and VA, 4 to 5-fold) and reduced the levels of less desirable FA (n-6 PUFA; 14:0, 16:0) in muscle and adipose tissues without adverse effects on growth and carcass quality. Differences in tissue FA composition were also noted, and high levels of VA in perirenal fat in particular have potential for use in testing for health effects and development of value added beef and other food products.  相似文献

An increasing interest in sustainable forms of agriculture exists worldwide and the demand for varieties specifically adapted to organic and low-input agriculture is rising. As a consequence, breeding methods need to be refined accordingly. In order to get better insight into needs and possibilities with this regard, a comprehensive ring test was performed from 2006 to 2008 with 14 winter wheat varieties in 36 environments in major cropping regions of Austria, France, Romania and Switzerland. Environments were grouped into 9 different subsets according to input systems, years, and countries. Input system N0 consisted of 13 organic and 6 no-input trials; 17 trials in input system N received various levels of synthetic nitrogen. For grain yield (YLD) and protein yield (PYLD), significant G × E was detected. Countries had a stronger effect on both traits than systems. Overall, it was more efficient to select YLD and PYLD in N, for targeting both systems N and N0. For PYLD, direct testing within a given country was always more efficient than indirect selection. Many traits could be scored equally well in both systems, N and N0, but for some traits particularly important for organic agriculture, such as soil coverage, better differentiation was observed under organic conditions. Therefore, we agree with other authors that a commercially sustainable breeding program for organic and low-input agriculture should combine information from high and low-input levels and from diverse regions. Reducing Emissions from Deforestation and Forest Degradation (REDD+) is an ambitious global programme oriented towards improving forest carbon management. It aims to attract new sources of ‘green’ capital to fund emissions reductions from avoided deforestation and sustainable forest management. REDD+ is transforming forest conservation, as a diverse array of new stakeholders become involved. Not surprisingly, REDD+ has proved divisive, as critics concern themselves with issues of power, justice, and commodification, while practice‐oriented researchers tackle similar issues from different perspectives, focusing on benefit sharing, safeguards, additionality, measuring and verification. In this paper we explore the different roles of critical and practical research, and argue that there is a need for greater sharing of knowledge across current divides. We draw on our own experiences of conducting a research project on REDD+ in Indonesia that involved critical and practice‐oriented researchers. We argue that critical research disconnected from practical matters can have perverse outcomes for practitioners who are ultimately working towards similar goals; while uncritical practice‐oriented research has the potential to lead to a dilution of core values of environmental justice and conservation. In contrast, forms of practical critique provide ways of researching REDD+ that have practical value while maintaining critical insights.  相似文献

Progesterone production is essential for growth and development of the conceptus during pregnancy. Abnormal development of the corpus luteum (CL) after conception can result in early embryonic loss or fetal abortion. Routine monitoring of bottlenose dolphin (Tursiops truncatus) pregnancy after artificial insemination or natural conception with ultrasonography and serum progesterone determination has allowed for the establishment of expected fetal growth rates and hormone concentrations. Using these monitoring techniques, we revealed four pregnant dolphins (12-24 yr old) with abnormally low progesterone production indicative of luteal insufficiency. Once diagnosed, animals were placed on altrenogest (0.044-0.088 mg/kg once daily) alone or with oral progesterone (50-200 mg twice daily). Doses of hormone were increased or decreased in each animal based on how fetal skull biparietal and thoracic growth rates compared with published normal values. Hormones were withdrawn starting from day 358 of gestation in animals 1 and 2, with labor occurring 6 and 7 days after withdrawal and at 376 and 373 days of gestation, respectively. Both deliveries were dystocic, with each calf requiring manual extraction and fetotomy for calf 1. The fetuses in animals 3 and 4 died at 348 and 390 days of gestation, respectively. Induction of labor was attempted in both animals, after fetal death, by using a combination of rapid progesterone withdrawal and steroid and prostaglandin F2alpha administration. The calf of animal 4 had to be removed with manual cervical dilation and fetotomy All adult females survived the procedures. These data provide the first in vivo evidence that the CL is the primary source of progesterone throughout pregnancy in the bottlenose dolphin. Until further characterization of hormones required during pregnancy and at parturition has been accomplished, the exogenous progestagen supplementation protocol described here cannot be recommended for treatment of progesterone insufficiency in bottlenose dolphins.  相似文献

Bioautography of extracts of the fruiting bodies of the basidiomycete Gomphus floccosus (Schw.) Singer indicated the presence of fungitoxic compounds in the ethyl acetate fraction against the plant pathogens Colletotrichum fragariae, Colletotrichum gloeosporioides, and Colletotrichum acutatum. Bioassay-guided fractionation of this fraction resulted in the isolation of the bioactive fatty acids (9 S,10 E,12 Z)-9-hydroxy-10,12-octadecadienoic acid (1), (9 E,11 Z)-13-oxo-9,11-octadecadienoic acid (2), and (10 E,12 E)-9-oxo-10,12-octadecadienoic acid (3). These three oxylipins were further evaluated for activity against a greater range of fungal plant pathogens (C. fragariae, C. gloeosporioides, C. acutatum, Botrytis cinerea, Fusarium oxysporum, Phomopsis obscurans, and Phomopsis viticola) in in vitro dose-response studies. Phomopis species were the most sensitive fungi to these compounds. At 120 h of treatment, the IC50 values for compounds 1, 2, and 3 for P. obscurans were 1.0, 4.5, and 23 microM, respectively, as compared to 1.1 microM for the captan positive control.  相似文献

The study of multiple pathosystems has played a central role in the development of botanical epidemiology, leading to a number of approaches and concepts. Multiple pathosystems are facts, which are experienced by many non-cultivated, or cultivated, plant communities. The shapes and composition of multiple pathosystems vary in space and time because of their inherent structure of relationships, and also in response to management. Examples of variation in multiple pathosystems are given, of groundnut in Côte d’Ivoire, of wheat in Brittany, and of upland rice in northern Laos. Variation in the yield-reducing effects of multiple pathosystems is discussed, including interactions among disease elements, relationships with attainable performances, and linkages with production situations. Progress has been achieved in understanding the links between injury profiles, production situations, and attainable performances. Questions about the functioning and consequences of multiple pathosystems are central to defining the scientific bases for, the design of, and the implementing of IPM. The complexity of multiple pathosystems, however, remains a deterrent, not a challenge, to many plant pathologists. Progress achieved in designing production systems for hardy wheat in France, however, is very promising, because of the multidisciplinary science it involves, and because of the promise to deliver it carries. The concepts of epidemiological guilds and of guilds of harmful agents are offered as perspectives to address and manage syndromes of production and syndromes of disease.  相似文献

Top ranking varieties are tested in multiple environments before and after registration in order to assess their value for cultivation and use (VCU testing). Recently, interest has increased in obtaining varieties specifically adapted to organic farming conditions. This raised the question if an independent system of trials may be required for this purpose. To help answering this question, through the exchange network of European cereal researchers SUSVAR (), a number of data sets of agronomic traits from barley, wheat and winter triticale, from trials performed in Denmark, Sweden, The Netherlands, France, Switzerland, UK and Germany, were made available and analysed using an approach based on mixed models involving parameters describing genetic correlation between the two types of experiments, i.e., organic and non-organic (high or low input). Estimated variance components and correlations were used to evaluate response to selection and index selection. The response to index selection was analysed as a function of the fraction of available trials assigned to the organic system. The genetic correlations were interpreted in terms of ranking agreement. We found high genetic correlations between both systems for most traits in all countries. Despite high genetic correlations, the chances of very good agreement in observed rankings were moderate. Combining information from both organic and non-organic systems is shown to be beneficial. Further, ignoring information from non-organic trials when making decisions regarding performance under organic conditions is a sub-optimal strategy.  相似文献
