Nixtamalized and extruded flours from quality protein maize (QPM, V‐537C) and tortillas made from them were evaluated for some technological and nutritional properties and compared with the commercial brand MASECA. Both QPM flours showed higher (P < 0.05) protein content, total color difference, pH, available lysine, and lower (P < 0.05) total starch content, Hunter L value, water absorption index, gelatinization enthalpy, resistant starch, and retrograded resistant starch than nixtamalized MASECA flour. Tortillas from nixtamalized and extruded QPM flours had higher contents of essential amino acids than tortillas from MASECA flour, except for leucine. Tortillas from processed QPM flours also showed higher (P < 0.05) values of the nutritional indicators calculated protein efficiency ratio (C‐PER 1.80–1.85 vs. 1.04), apparent and true in vivo protein digestibility (78.4‐79.1 vs. 75.6% and 76.4–77.4 vs. 74.2%, respectively), PER (2.30–2.43 vs. 1.31), net protein retention (NPR; 2.88–2.89 vs. 2.11), and protein digestibility corrected amino acid score (PDCAAS; 54–55 vs. 29% based on preschool children and 100 vs. 85% based on adults) than MASECA flour. The use of QPM for flour and tortilla preparation may have a positive effect on the nutritional status of people from countries where these products are widely consumed.  相似文献

A strong recovery response occurs in cantaloupe (Cucumis melo) and watermelon (Citrullus lanatus) infected with the bipartite begomovirus Cucurbit leaf crumple virus (CuLCrV). This response is characterized by initially severe symptoms, which gradually become attenuated (almost symptomless). An inverse relationship was detected between viral DNA levels and recovery, indicating that recovered tissues had reduced viral titers. Recovered tissues also were resistant to reinfection with CuLCrV; i.e., recovered leaves reinoculated with the virus did not develop symptoms or have an increased level of viral DNA. In contrast, infection of CuLCrV-recovered leaves with the RNA virus, Cucumber mosaic virus (CMV), disrupted recovery, resulting in the development of severe disease symptoms (more severe than those induced by CMV or CuLCrV alone) and increased CuLCrV DNA levels. Small RNAs with homology to CuLCrV DNA were detected in recovered and nonrecovered tissues; as well as in phloem exudates from infected, but not uninfected plants. Levels of these small RNAs were positively correlated with viral titer; thus, recovered tissues had lower levels than symptomatic tissues. In addition, viral DNA from a host that undergoes strong recovery (watermelon) was more highly methylated compared with that from a host that undergoes limited recovery (zucchini). Furthermore, inoculation of CuLCrV-infected zucchini with a construct expressing an inverted repeat of the CuLCrV common region enhanced recovery and reduced viral symptoms and viral DNA levels in newly emerged leaves. Taken together, these results suggest that recovery from CuLCrV infection is an adaptive antiviral defense mechanism, most likely mediated by gene silencing.  相似文献

The main cellular responses of innate immunity are phagocytic activity and the respiratory burst, which produces a high amount of reactive oxygen species. Natural killer enhancing factor (NKEF) belongs to the peroxiredoxin family that has an antioxidant function and enhances cytotoxic cell activity. This molecule may play a key role in macrophage and cytotoxic cell communication during the innate immune response of fish against pathogens. In fish, the NKEF gene has been characterized in some species as showing an up-regulation in infected fish, suggesting a trigger effect upon NK-like cells. To detect and localize this molecule in salmonids at protein level, a monospecific polyclonal antibody was generated. A probable NKEF-like protein epitope region was identified and characterized using bioinformatic tools, and the sequence was chemically synthesized using Fmoc strategy, analysed by RP-HPLC and its molecular weight confirmed by mass spectrometry. The synthetic peptide was immunized and antibodies from ascitic fluid were obtained. The resulting antibody is a versatile tool for detecting NKEF by different immune techniques such as ELISA, Western blotting and immunohistochemistry. Analysis of NKEF-like protein is a useful method for characterizing immune properties of this molecule in fish during response to pathogens.  相似文献

Maslinic acid (MA) is a pentacyclic triterpene used, at experimental level, as an additive to stimulate growth, protein‐turnover rates and hyperplasia in fish. In this experiment we have studied the effects of feeding with MA at daily fixed ration of 10 g kg?1 of body mass, on growth, protein‐turnover rates and nucleic acid concentration in white muscle of gilthead sea bream grown in a local fish farm. The experimental groups contained 0 (control), 50 (MA50) and 100 (MA100) mg MA per kg of diet. Two groups were provided feed ad libitum (control AL and MA100AL), while three were given a fixed ration (control R, MA50R, and MA100R). We have determined the following: growth rate; nucleic acid and protein levels; relative and absolute protein‐accumulation (KG, AG), synthesis (KS, AS) and degradation (KD, AD) rate, protein‐synthesis capacity (CS), protein‐synthesis efficiency (KRNA), protein‐synthesis rate per DNA (KDNA) and protein‐retention efficiency. At the end of the experiment, higher body weights and muscle growth rates were found in both groups of fish fed with 100 mg MA per kg of diet. Feed‐efficiency rate, protein‐efficiency ratio and protein productive value, were higher in the MA100AL group than in control AL. Fractional and absolute protein‐synthesis and degradation rates in white muscle of MA100AL and MA100R fish were higher than in the control, resulting in a higher protein‐accumulation rate and tissue growth. Total DNA content, indicated cell hyperplasia, was higher in MA100AL and MA100R than its respective controls. Studies of optical and electronic microscopy corroborated these results. These findings indicate that MA added to the diet can stimulate the growth, white‐muscle protein‐turnover rates and tissue hyperplasia in gilthead sea bream.  相似文献

Fourteen different silages were prepared using mixtures of Moringa (Moringa oleifera), Elephant grass (Pennisetum purpureum cv Taiwan) or sugar cane (Saccharum officinarum). Molasses from sugar cane was used in the amounts of either 10 or 50 g kg?1 fresh matter (FM) in treatments without sugar cane. A completely randomized design with three replicates of each treatment was used. The silages were prepared in 1800 mL micro silos and opened after 120 d. The presence of Moringa and Elephant grass in the silage changed the pH by ?0·8 and +0·7, respectively (P < 0·001), indicating a favourable effect of Moringa on silage pH. Overall differences were found among treatments for dry matter content, crude protein and acetic acid concentrations, weight loss, CO2 production and silage pH after spoilage (P < 0·001). Weight loss was proportionately 0·034 and 0·014 in silages with and without sugar cane respectively (P < 0·001). Overall, differences (P < 0·05) were also found for neutral‐detergent fibre and lactic acid concentrations, lactic acid bacteria counts, clostridial counts and time to spoilage of the silages. Treatments containing Moringa had higher lactic acid concentrations (+16 g kg?1 DM; P < 0·01) compared to treatments without but the presence of Moringa decreased time to spoilage by 67 h (P < 0·05). No differences were found in propionic acid concentration or fungal growth of the silages. It is concluded that Moringa can be used as a component of high quality silages which also contain high concentrations of crude protein.  相似文献

Clonally propagated crop species are less adaptable to environmental changes than those propagating sexually. DNA studies have shown that in all countries where taro (Colocasia esculenta (L.) Schott) has been introduced clonally its genetic base is narrow. As genetic variation is the most important source of adaptive potential, it appears interesting to attempt to increase genetic and phenotypic diversity to strengthen smallholders’ capacity to adapt to climatic changes. A global experiment, involving 14 countries from America, Africa, Asia and the Pacific was conducted to test this approach. Every country received a set of 50 indexed genotypes in vitro assembling significant genetic diversity. After on-station agronomic evaluation trials, the best genotypes were distributed to farmers for participatory on-farm evaluation. Results indicated that hybrids tolerant to taro leaf blight (TLB, Phytophthora colocasiae Raciborski), developed by Hawaii, Papua New Guinea and Samoa breeding programmes outperformed local cultivars in most locations. However, several elite cultivars from SE Asia, also tolerant to TLB, outperformed improved hybrids in four countries and in one country none of the introductions performed better than the local cultivars. Introduced genotypes were successfully crossed (controlled crossing) with local cultivars and new hybrids were produced. For the first time in the history of Aroids research, seeds were exchanged internationally injecting tremendous allelic diversity in different countries. If climatic changes are going to cause the problems envisaged, then breeding crops with wide genetic diversity appears to be an appropriate approach to overcome the disasters that will otherwise ensue.  相似文献

Cultivated beets (Beta vulgaris ssp. vulgaris) are unable to form reproductive shoots during the first year of their life cycle. Flowering only occurs if plants get vernalized, that is, pass through the winter, and are subsequently exposed to an increasing day length (photoperiod) in spring. Here, we show that the regulation of flowering time in beets is controlled by the interplay of two paralogs of the FLOWERING LOCUS T (FT) gene in Arabidopsis that have evolved antagonistic functions. BvFT2 is functionally conserved with FT and essential for flowering. In contrast, BvFT1 represses flowering and its down-regulation is crucial for the vernalization response in beets. These data suggest that the beet has evolved a different strategy relative to Arabidopsis and cereals to regulate vernalization.  相似文献

Dry and wet deposition of atmospheric nitrogen species (NO2 and HNO3) coming from nitrogen oxides emissions in Buenos Aires city to surface waters of de la Plata River were estimated. Atmospheric dispersion models DAUMOD-RD (v.2) and CALPUFF were applied to area and point sources, respectively. These models were run considering 1 year of hourly meteorological data. Emission information included a typical diurnal variation of area source emissions. Annual atmospheric nitrogen (N–NO2?+?N–HNO3) deposition to 1,763 km2 of the river was 35,600 kg-N year?1. Dry deposition processes accounted for 89% of this value. The small contribution of wet deposition was a consequence of the very few cases (5%) of rain events during offshore wind conditions. Monthly dry deposition to 1,763 km2 of the river varied from 1,628 kg-N month?1 in February to 3,799 kg-N month?1 in December, following the monthly occurrence of offshore winds. Monthly wet deposition varied from 1 kg-N month?1 in June to 1,162 kg-N month?1 in February. These results came from the combination of favorable conditions for formation of HNO3 and the occurrence of precipitation during offshore wind situations. Spatial distribution of annual atmospheric N deposition showed a strong coastal gradient. Deposition values reached a maximum of 137.1 kg-N km?2 year?1 near the shoreline, which was reduced to the half at 4 km from the coast.  相似文献

The FeEDDHA [iron(3+) ethylenediamine di(o-hydroxyphenylacetic) acid] is one of the most efficient iron chelates employed in the correction of iron clorosis in calcareous soils. FeEDDHA presents different positional isomers: the ortho-ortho (o,o), the ortho-para (o,p), and the para-para (p,p). Of these isomers, the p,p cannot chelate Fe in soil solution in a wide range of pH values, while both o,o and o,p can. The objective of this work was to compare the efficiency of both isomers (o,o and o,p) to provide Fe to two Strategy I plants (tomato and peach) in nutrient solution (pH approximately 6.0), as well as in calcareous soil (pH approximately 8.4; CALCIXEREPT). For this, chelates of both o,o-EDDHA and o,p-EDDHA with 57Fe (a nonradioactive isotope of Fe) were used, where the 57Fe acts as a tracer. The results obtained showed that the o,o isomer is capable of providing sufficient Fe to plants in both nutrient solution and calcareous soil. However, the o,p isomer is capable of providing sufficient Fe to plants in nutrient solution but not in calcareous soil.  相似文献
