Grapevine leafroll disease is associated with several species of phloem-limited grapevine leafroll-associated viruses (GLRaV), some of which are transmitted by mealybugs and scale insects. The grape phylloxera, Daktulosphaira vitifoliae (Fitch) Biotype A (Hemiptera: Phylloxeridae), is a common vineyard pest that feeds on the phloem of vine roots. There is concern that these insects may transmit one or more GLRaV species, particularly GLRaV-2, a species in the genus Closterovirus. A field survey was performed in vineyards with a high incidence of grapevine leafroll disease and D. vitifoliae was assessed for acquisition of GLRaV. In greenhouse experiments, the ability of D. vitifoliae to transmit GLRaV from infected root sections or vines to co-planted virus-free recipient vines was tested. There were no GLRaV-positive D. vitifoliae in the field survey, nor did D. vitifoliae transmit GLRaV-1, ?2, ?3, or -4LV in greenhouse transmission experiments. Some insects tested positive for GLRaV after feeding on infected source vines in the greenhouse, however there was no evidence of virus transmission to healthy plants. These findings, in combination with the sedentary behaviour of the soil biotype of D. vitifoliae, make it unlikely that D. vitifoliae is a vector of any GLRaV. 相似文献

A standard area diagram set (SADs) with eight severity values ranging from 0.5 to 32% was evaluated as a tool to improve the accuracy of the estimates of Phomopsis leaf blight severity on eggplant by ten inexperienced raters. A first assessment in a 50-leaf dataset of digital images was performed unaided (UN, no SAD). Two further aided (A) assessments, conducted two (A1) and four weeks after the first (A2) unaided one were performed. The precision of the estimates, as indicated by the Pearson’s correlation coefficient (r), improved significantly when using the aid (A1: r = 0.96 and A2: r = 0.96) compared to unaided (UN: r = 0.79). However, the generalized bias (C b) was not significantly affected because the unaided estimates were already quite accurate (C b = 0.93). The overall concordance (ρ c) was significantly improved due to the large gains in precision when using the SADs. The raters’ estimates were more uniform (ρ c > 0.9) when using the aid, with progressive gains in the reliability of the estimates among them, indicated by the concordance correlation coefficient statistics. The SADs proposed in this study will be useful in severity estimation during field work involving multiple raters, especially as use of the SADs results in less variable estimates, improving accuracy and reliability among raters. 相似文献

The aim of this study was to evaluate the genotoxic and mutagenic potential of contaminated soil diluted in acidic solutions and not acidic, in the offspring of rats exposed during pregnancy and neonatal periods. To this end, a comet assay and micronucleus test were performed. Soil samples were solubilized in the following three solvents: distilled water (control group), acid solvent at pH 5.2, and acid solvent at pH 3.6. Soil and solvent were mixed in a rate of 1:2 in g/mL, and hydrofluoric acid was used in the acid solvents. In the comet assay, the tail length, percentage of DNA within the tail and tail moment was analyzed in the whole blood of the pups that were studied. The number of micronuclei found in the bone marrow cells was counted in the micronucleus test. In all parameters evaluated in the comet assay, the group exposed to the lowest pH value when associated with contaminated soil (p < 0.05) had the most damage. However, the micronucleus test showed differences between all exposed groups and the control group (p < 0.05). These results reaffirm the health risks related to the exposure to soil contaminants. 相似文献

Most Japanese plum-type cultivars are self-incompatible and cross pollination is necessary to ensure fruit set. In this study, the S -RNase genotype and the incompatibility group of 68 Japanese plum-type cultivars were determined by PCR amplification of the S-RNase gene. The S -RNase genotype of 50 cultivars is first reported here and five new Japanese plum S -RNase alleles ( So , Sp , Sq , Sr , Ss ) were identified. The results obtained, together with information compiled from previous studies, allowed describing 12 new incompatibility groups (VIII–XIX). The self-incompatibility of several cultivars and the cross-compatibility among different incompatibility groups were verified by self- and cross-pollination experiments followed by observation of pollen tube growth. Five cultivars behaved as self-compatible, but two of them do not have the Se allele, which has been correlated with self-compatibility. Thus, additional sources of self-compatibility different from Se appear to be involved in Japanese plum self-compatibility. 相似文献

BACKGROUND: In Brazil, dengue vector control is hampered by the resistance of Aedes aegypti L. populations to organophosphates (OPs). Insect growth regulators (IGRs) are a promising alternative, as their mechanisms of action are different from those of conventional insecticides. The authors analysed the effect of the IGR triflumuron, a chitin synthesis inhibitor, on the Ae. aegypti insecticide-susceptible strain Rockefeller, as well as on field populations both susceptible (TemS) and resistant (TemR) to the OP temephos. RESULTS: Triflumuron arrested development and inhibited adult emergence of the Rockefeller strain in a dose-dependent way (EI(50) and EI(90) of 0.8 and 1.8 microg L(-1) respectively). A direct relationship between triflumuron concentration and the precocity of its effects was evident. TemS and TemR temephos resistance ratios (RR(90)) were 4.5 and 13.8, triflumuron RR(90) being 1.0 and 1.3 respectively. CONCLUSION: The IGR triflumuron exhibited a dose-dependent effect against the reference Ae. aegypti Rockefeller strain. It was also effective against two field populations, regardless of their OP resistance status. The present results are discussed in the context of utilization of chitin synthesis inhibitors as potential alternatives in the control of Ae. aegypti in Brazil. 相似文献

Previous reviews of stress, and the stress hormone cortisol, in fish have focussed on physiology, due to interest in impacts
on aquaculture production. Here, we discuss cortisol in relation to fish welfare. Cortisol is a readily measured component
of the primary (neuroendocrine) stress response and is relevant to fish welfare as it affects physiological and brain functions
and modifies behaviour. However, we argue that cortisol has little value if welfare is viewed purely from a functional (or
behavioural) perspective—the cortisol response itself is a natural, adaptive response and is not predictive of coping as downstream
impacts on function and behaviour are dose-, time- and context-dependent and not predictable. Nevertheless, we argue that
welfare should be considered in terms of mental health and feelings, and that stress in relation to welfare should be viewed
as psychological, rather than physiological. We contend that cortisol can be used (with caution) as a tractable indicator
of how fish perceive (and feel about) their environment, psychological stress and feelings in fish. Cortisol responses are
directly triggered by the brain and fish studies do indicate cortisol responses to psychological stressors, i.e., those with
no direct physicochemical action. We discuss the practicalities of using cortisol to ask the fish themselves how they feel
about husbandry practices and the culture environment. Single time point measurements of cortisol are of little value in assessing
the stress level of fish as studies need to account for diurnal and seasonal variations, and environmental and genetic factors.
Areas in need of greater clarity for the use of cortisol as an indicator of fish feelings are the separation of (physiological)
stress from (psychological) distress, the separation of chronic stress from acclimation, and the interactions between feelings,

Little research has been done in optimizing the nitrogenous fraction of the fish diets in order to minimize welfare problems.
The purpose of this review is to give an overview on how amino acid (AA) metabolism may be affected when fish are under stress
and the possible effects on fish welfare when sub-optimal dietary nitrogen formulations are used to feed fish. In addition,
it intends to evaluate the current possibilities, and future prospects, of using improved dietary nitrogen formulations to
help fish coping with predictable stressful periods. Both metabolomic and genomic evidence show that stressful husbandry conditions
affect AA metabolism in fish and may bring an increase in the requirement of indispensable AA. Supplementation in arginine
and leucine, but also eventually in lysine, methionine, threonine and glutamine, may have an important role in enhancing the
innate immune system. Tryptophan, as precursor for serotonin, modulates aggressive behaviour and feed intake in fish. Bioactive
peptides may bring important advances in immunocompetence, disease control and other aspects of welfare of cultured fish.
Fishmeal replacement may reduce immune competence, and the full nutritional potential of plant-protein ingredients is attained
only after the removal or inactivation of some antinutritional factors. This review shows that AA metabolism is affected when
fish are under stress, and this together with sub-optimal dietary nitrogen formulations may affect fish welfare. Furthermore,
improved dietary nitrogen formulations may help fish coping with predictable stressful events. 相似文献

Bio-floc shrimp culture systems have been investigated in an attempt to optimize water use and prevent the discharge of effluent into the environment. The importance of microalgae in maintaining water quality and nutrition of the shrimp is well known in conventional systems; however, its maintenance amid bio-flocs and its role in the shrimp performance in this system are still poorly understood. The aim of this study was to evaluate the contribution of diatoms in the performance of Litopenaeus vannamei reared during the nursery phase in intensive system with minimal water exchange. Shrimp (0.31?±?0.10?g) were reared among diatoms, bio-flocs and the combination of the two forming the mixture medium. The survival of shrimp was high in all treatments (90?C97%). However, the shrimp reared among diatoms showed higher weight gain (P?<?0.05) and feed conversion ratio significantly more efficient, reaching a value of 0.47. The results indicate the importance of diatoms in bio-floc culture systems and points out to future research in an attempt to maintain a constant presence of these microalgae in culture medium without requiring successive inoculations. 相似文献

The palm Euterpe edulis has high ecological and economic importance in Brazil. Currently, this species is being cultivated and managed for spontaneous
regeneration in banana plantations. However, there are no data comparing its plantation growth performance to its native forest
growth. We evaluated growth and mortality (M) of individuals of E. edulis planted in secondary dense ombrophilous forest and in banana plantations, as well as their relationships with site variables
(canopy opening, soil nutrient availability, density of existing E. edulis, and herbivory). Twelve banana plantation sites and 12 sites in secondary dense ombrophilous forests were selected. At each
site, 25 young individuals of E. edulis were planted in 2003. Annually until 2008, morphometric, herbivory, and M of the individuals were evaluated. In 2008, canopy
and soil variables were measured at each plot. E. edulis growth was five times higher in banana plots compared to forest plots; current annual increment on height reached 38.9 cm
in banana plots, compared to 7.3 cm in forest plots. M was relatively low and similar at both sites, presenting an intraspecific
density-dependence pattern. Significant correlations were found between morphometric variables, M, and herbivory of E. edulis and canopy and soil variables. Euterpe edulis presented plasticity that allows for its establishment in banana plantations, indicating high potential for management in
agroforestry consortia. Such management may be a useful conservation strategy for this and other shade-tolerant species. 相似文献
