We characterized differences in cell wall material and polysaccharide structures, due to the quantitative trait loci associated with higher flesh firmness in a nonclimacteric near-isogenic line (NIL) SC7-2, and with the climacteric behavior of the NIL SC3-5-1, using their nonclimacteric inbred parentals, "Piel de Sapo" (PS) and PI 161375 (SC). PS was firmer and had a higher ripening index and greater hemicellulosic content than SC, with its lower wall material yield, and uronic acid, neutral sugar, cellulose and free sugar content and higher pectic content. SC3-5-1 showed lower uronic acid values, a higher soluble solid content, and similar flesh firmness to PS. SC3-5-1 yielded mainly high molecular weight polysaccharides in the imidazole-soluble fraction than PS. SC7-2 showed greater flesh firmness, a higher neutral sugar (especially galactose and mannose) and uronic acid content, together with a larger cellulose and α-cellulose residue than PS. SC7-2 also contained more polysaccharides of low molecular weight in the first pectic fraction and shifted toward higher molecular weights in the main peak of the 4 M potassium-soluble fraction compared with PS.  相似文献

Under semiarid Mediterranean climatic conditions, soils typically have low organic matter content and weak structure resulting in low infiltration rates. Aggregate stability is a quality indicator directly related to soil organic matter, which can be redistributed within soil by tillage. Long-term effects (1983–1996) of tillage systems on water stability of pre-wetted and air dried aggregates, soil organic carbon (SOC) stratification and crop production were studied in a Vertic Luvisol with a loam texture. Tillage treatments included conventional tillage (CT), minimum tillage (MT) and zero tillage (ZT) under winter wheat (Triticum aestivum L.) and vetch (Vicia sativa L.) rotation (W–V), and under continuous monoculture of winter wheat or winter barley (Hordeum vulgare L.) (CM). Aggregate stability of soil at a depth of 0–5 cm was much greater when 1–2 mm aggregates were vacuum wetted prior to sieving (83%) than when slaked (6%). However, slaking resulted in tillage effects that were consistent with changes in SOC. Aggregate stability of slaked aggregates was greater under ZT than under CT or MT in both crop rotations (i.e., 11% vs. 3%, respectively).

SOC under ZT tended to accumulate in the surface soil layer (0–5 and 5–10 cm) at the expense of deeper ones. At depths of 10–20 and 20–30 cm no differences in SOC were encountered among tillage systems, but CT exhibited the highest concentration at 30–40 cm depth. Nevertheless, when comparisons were made on mass basis (Mg ha−1), significant differences in stocked SOC were observed at depths of 0–10 and 0–20 cm, where ZT had the highest SOC content in both rotations. The stock of SOC to a depth of 40 cm, averaged across crop rotations, was greater under ZT (43 Mg ha−1) than under CT (41 Mg ha−1) and MT (40 Mg ha−1) although these figures were not significantly different. Likewise, no significant differences were encountered in the stock of SOC to a depth of 40 cm among crop rotations (i.e., 42 Mg ha−1 for W–V vs. 40 Mg ha−1 for CM).

Crop production with wheat–vetch and continuous cereal showed no differences among tillage systems. Yields were strongly limited by the environmental conditions, particularly the amount of rainfall received in the crop growth season and its distribution. Similar yield and improved soil properties under ZT suggests that it is a more sustainable system for the semiarid Mediterranean region of Spain.  相似文献

Free-living cells of the social amoebae Dictyostelium discoideum can aggregate and develop into multicellular fruiting bodies in which many die altruistically as they become stalk cells that support the surviving spores. Dictyostelium cells exhibit kin discrimination--a potential defense against cheaters, which sporulate without contributing to the stalk. Kin discrimination depends on strain relatedness, and the polymorphic genes tgrB1 and tgrC1 are potential components of that mechanism. Here, we demonstrate a direct role for these genes in kin discrimination. We show that a matching pair of tgrB1 and tgrC1 alleles is necessary and sufficient for attractive self-recognition, which is mediated by differential cell-cell adhesion. We propose that TgrB1 and TgrC1 proteins mediate this adhesion through direct binding. This system is a genetically tractable ancient model of eukaryotic self-recognition.  相似文献

Biomass monitoring is one of the main pillars of precision farm management as it involves deeper knowledge about pest and weed status, soil quality, water stress, and yield prediction, among others. This research focuses on estimating crop biomass from high-resolution red, green, blue imaging obtained with an unmanned aerial vehicle. Onion, as one of the most cultivated vegetables, was studied for two seasons under non-controlled conditions in two commercial plots. Green canopy cover, crop height, and canopy volume (Vcanopy) were the predictor variables extracted from the geomatic products. Strong relationships were found between Vcanopy and dry leaf biomass and dry bulb biomass. Adjusted coefficient of determination (\({\text{R}}_{\text{adj}}^2\)) values were 0.76 and 0.95, respectively. Nevertheless, crop management practices and leaf depletion at vegetative stages significantly affect the accuracy of the canopy model. These results suggested that obtaining biomass using aerial images are a good alternative to other sensors and platforms as they have high spatial and temporal resolution to perform high-quality biomass monitoring.  相似文献

In vitro assays used porcine or bovine trypsin as models of exogenous enzymes to determine functioning in the presence of enzymatic extracts from the digestive gland of whiteleg shrimp Penaeus vannamei. Using electrophoresis and zymograms, when enzymes from the shrimp were mixed in the absence of protein substrate, they hydrolysed the trypsin from bovine or porcine origin. Porcine or bovine trypsin, when mixed with shrimp enzymes in pH‐stat assays in the presence of shrimp commercial feed, fish meal, or casein, there was added activity to hydrolyse the protein substrate. Hydrolysis of protein substrate was twofold to threefold stronger if exogenous enzymes were added. Results suggest that porcine or bovine trypsin could be used as feed supplements for whiteleg shrimp P. vannamei to enhance hydrolysis of proteins in feeds, because the commercial enzymes contributed to the hydrolysis of the protein in the three substrates in the presence of shrimp enzymes.  相似文献

The bacteria Microbacterium sp. 8L and Exiguobacterium mexicanum 8N are known to improve the culture of Artemia franciscana using baker's yeast as food. Using spectrophotometry, sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS‐PAGE), substrate‐SDS‐PAGE and pH‐stat in vitro‐digestibility assays, the activity, molecular mass and hydrolysis on baker's yeast protein of proteases from extracellular polymeric substances (EPS) of the strains 8L and 8N along with the pathogenic strains Microbacterium sp. 8R and Vibrio parahaemolyticus 588 CECT (Vp) were studied. The EPSs of 8L and 8R showed one activity band, on which the serine inhibitor phenylmethylsulphonyl fluoride (PMSF) had no effect. The EPSs of 8N showed four bands; two were unaffected by PMSF, whereas one was affected, and the other was partially affected. The EPSs of Vp showed two bands, one partially inhibited by PMSF. No inhibitory effects from 1‐chloro‐3‐tosylamido‐7‐amino‐2‐heptanone (trypsin inhibitor) were observed in the protease bands of the studied bacteria. The EPSs of 8L and 8N showed a similar degree of hydrolysis (pH‐stat). The EPSs of 8L had the lowest Dice index of similarity of yeast protein profiles at 1 h of reaction. We conclude that the strain 8L could benefit A. franciscana by providing bacterial proteases for digestion of baker's yeast.  相似文献

Wheat streak mosaic virus (WSMV) has been newly documented in Australia. The vulnerability of contemporary Australian elite wheat germplasm prompted a survey for effective resistance against an Australian isolate, WSMV‐ACT. This study confirms the effectiveness of previously reported sources of resistance and shows that new sources of resistance also confer protection. The resistance derived from Thinopyrum intermedium (Wsm1) as a 4D translocation and a new 4A translocation, and two bread wheat resistances, Wsm2 and the new source c2652, were effective against WSMV‐ACT in glasshouse experiments. Wsm1 was effective at lower temperatures but ineffective above 20°C, a temperature sensitivity shared with many of the derivatives of Wsm2 except for one new selection which was effective at 26°C. True wheats c2652 and Wsm2 selection CA745, and amphiploids Zhong1, Zhong2, Zhong4, Zhong5, TAF46, Summer1, Ot38 and OK7211542 were uniformly resistant at 20, 25 and 28°C. New sources of resistance were identified in a Th. scirpeum‐wheat amphiploid, B84‐994, and in chromosome addition lines Z2, Z6 and TAi27, derived from wheat‐Th. intermedium partial amphiploids. Several new, tightly linked SSR, RAPD and EST‐ILP PCR markers were developed for tracking the various Th. intermedium translocations associated with Wsm1, including the smaller translocations on wheat chromosome 4AS and 4DS. This survey was conducted to determine the chronobiology and seroprevalence of nasal bot infestation (Oestrus ovis) in Spain and to identify the risk factors associated with this disease in caprine herds.

A total of 1590 sera from adult goats were collected at random on 175 farms in southwestern Spain. Sera were tested by ELISA, using crude protein from second stage larvae as antigen. The mean seroprevalence was 46.04% and mean percentage of optical densities was 41.83. These data indicate a high prevalence of this parasite in the investigated areas. The serological survey revealed that goats managed at higher altitudes, at meridians latitudes and on farms with small herds had a smaller probability of infestation. Eighty goat heads, obtained from abattoirs in the central region of Spain, were collected and examined for nasal botflies from February to October 2002. O. ovis larval stages were recovered from the nasal-sinus cavities of 23 goats, reaching a prevalence of 34.94%. The mean larval burden was 3.9 larvae per infested head. No first instars were found during February and March, when the second instar reached its larger count. The third instar was observed in very small number during the whole period of study, with one peak occurring in July–August. These data show the existence of a favourable period for the development of larval instars of O. ovis in goats that starts in February and finishes in September.  相似文献
