Fisheries science uses quantitative methods to inform management decisions that reflect cultural preferences which, in turn, indirectly influence the states of ecosystems. To date, it has largely supported Eurocentric preferences for the commodification of marine organisms under the tenets of maximum sustainable yield, whereby abundances are intentionally maintained far below their historical baselines despite broader socio-ecological trade-offs. In contrast, Indigenous Knowledge Systems (IKS) adhere to the principle of “take only what you need and leave lots for the ecosystem,” implementing lower fishery removals to support socio-ecological resilience. Despite the power imbalance favouring Eurocentric preferences in decision-making, fisheries scientists increasingly recognize that the pairing of IKS and Western science, or Two-Eyed Seeing, would lead to more holistic management goals. For recognition to transcend tokenism, meaningful collaborations and co-governance structures underlying knowledge co-production must carry through to legislated policy changes. Using recent co-governance developments for fisheries management and spatial protections involving federal, provincial and Indigenous governments in Pacific Canada, we illustrate how the precautionary approach, including reference points and harvest control rules broadly applied in international fisheries, could be revised to make collaborative fisheries management compatible with IKS and improve biodiversity and fisheries protections. Our recommendations may create socio-economic trade-offs at different timescales for commercial fishers. Pre-empting that challenge, we discuss IKS-compatible economic approaches for addressing shorter term costs arising from reduced exploitation rates. Although our case study derives from Pacific Canada, the insights provided here are broadly applicable elsewhere in the world.  相似文献

Experts identified water quality, manure, good handling practices (including personal hygiene and equipment sanitation), and traceability as critical farm problem areas that, if addressed, are likely to decrease risk associated with microbial contamination of fresh produce from all scales of agriculture. However, the diverse nature of production strategies used by produce farmers presents multiple options for addressing foodborne illness issues while simultaneously creating potential complications. We use a mental models methodology to enhance our understanding of the underlying factors and assumptions of small, medium, and large produce growers that influence their decision-making processes for contamination prevention and control. This empirical evidence demonstrates how challenges and opportunities to food safety are related to the scale of production and marketing strategies. We believe that refining the development of standards and existing extension and outreach food safety programs are important to both consumer protection and supporting agricultural communities. Additionally, this approach will help develop and refine food safety programs that will result in empirically grounded recommendations based on identified grower information needs.  相似文献

Certain human pathogens avoid elimination by our immune system by rapidly mutating the surface protein sites targeted by antibody responses, and consequently they tend to be problematic for vaccine development. The behavior described is prominent for a subset of viruses--the highly antigenically diverse viruses--which include HIV, influenza, and hepatitis C viruses. However, these viruses do harbor highly conserved exposed sites, usually associated with function, which can be targeted by broadly neutralizing antibodies. Until recently, not many such antibodies were known, but advances in the field have enabled increasing numbers to be identified. Molecular characterizations of the antibodies and, most importantly, of the sites of vulnerability that they recognize give hope for the discovery of new vaccines and drugs.  相似文献

Under conditions of stress, such as elevated temperature, molecular chaperones stabilize proteins from unfolding, aggregating, and precipitating. We have investigated the chaperone activity of the major milk proteins alpha(S)-, beta-, and kappa-casein with reduced insulin and the milk whey proteins, alpha-lactalbumin and beta-lactoglobulin, and compared it with that of the mammalian small heat shock protein (sHsp), alpha-crystallin, and clusterin. alpha(S)-Casein exhibited different chaperone behavior under reduction and heat stresses, i.e., chaperone activity increased with increasing temperature (as observed with alpha-crystallin), but under reduction stress, its chaperone activity increased at lower temperatures. beta- and kappa-casein had comparable chaperone ability with each other but were less effective than alpha(S)-casein. Under molecular crowding conditions, precipitation of stressed protein was accelerated, and alpha(S)-casein was a poorer chaperone. Furthermore, at slightly alkaline pH values, alpha(S)-casein was a less effective chaperone than at neutral pH. Detailed fluorescence, size exclusion chromatography, and real-time NMR studies studies indicated that the casein proteins underwent conformational changes and stabilized the partially unfolded whey proteins prior to formation of high molecular weight soluble complexes. These results are consistent with casein proteins acting as molecular chaperones in a manner similar to sHsps and clusterin.  相似文献

A genetic fingerprinting technique (AFLP) was used to determine the relationships among Capparis spp. Genetic distances, based on AFLP data were estimated for 45 accessions of Capparis species, from Spain, Morocco and Syria. The results of this analysis support the differentiation of four of the five taxa involved. The group of plants recognised as C. spinosa on the basis of morphological characters, includes several cultivars and appears in an intermediate position between C. orientalis and C. sicula and overlaps with C. orientalis. The other two species C. aegyptia and C. ovata are separate from the rest. Capparis spinosa had a low number of unique bands in comparison with the other species. Although these results cannot confirm the hybrid origin of C. spinosa, the distribution of the bands supports this hypothesis, the most likely parental species being C. orientalis and C. sicula.  相似文献

The CCR5 co-receptor binds to the HIV-1 gp120 envelope glycoprotein and facilitates HIV-1 entry into cells. Its N terminus is tyrosine-sulfated, as are many antibodies that react with the co-receptor binding site on gp120. We applied nuclear magnetic resonance and crystallographic techniques to analyze the structure of the CCR5 N terminus and that of the tyrosine-sulfated antibody 412d in complex with gp120 and CD4. The conformations of tyrosine-sulfated regions of CCR5 (alpha-helix) and 412d (extended loop) are surprisingly different. Nonetheless, a critical sulfotyrosine on CCR5 and on 412d induces similar structural rearrangements in gp120. These results now provide a framework for understanding HIV-1 interactions with the CCR5 N terminus during viral entry and define a conserved site on gp120, whose recognition of sulfotyrosine engenders posttranslational mimicry by the immune system.  相似文献

Is it possible that the rush to replicate existing models, using ‘best practice’ principles developed far from the Paci?c, is curtailing careful market analysis and sti?ing experimentation and innovation in the provision of ?nancial services for the poor? Background: Regulatory T cells (Treg) have been shown to suppress antitumor immunity and often are increased in humans and rodents with cancer. However, Tregs have not been well studied in dogs with cancer and it is not known if certain tumor types are associated with increased Tregs.
Hypothesis: We hypothesized that Treg percentages would be increased in dogs with cancer and that Treg percentages would be higher in dogs with certain types of cancer.
Animals: The percentages and numbers of Tregs and nonregulatory T cells and B cells were assessed in 34 dogs with cancer and 9 age-matched control dogs. Dogs evaluated included 14 dogs with sarcoma, 7 dogs with carcinoma, 7 dogs with lymphoma, and 6 dogs with mast cell tumor.
Methods: Numbers and percentages of Tregs, CD4+, and CD8+ T cells and B cells were determined using flow cytometry and compared between control dogs and dogs with cancer.
Results: The percentage of Tregs was significantly increased overall in dogs with cancer compared with control dogs. When tumor types were compared, Treg percentages were significantly increased in dogs with carcinoma. The Treg/CD8 T cell ratio was significantly higher in dogs with cancer compared with control dogs and was also significantly increased in 2 dogs with T-cell lymphoma.
Conclusions: Treg percentages in blood were increased in dogs with cancer, particularly in dogs with carcinoma. The Treg/CD8 ratio also identified tumor-specific abnormalities in dogs with cancer. These findings indicate that tumor-specific factors may affect Tregs in dogs.  相似文献
