The diffusion of two fluorescent dyes, Oregon Green 488 (Oregon Green) and Rhodamine B into the leaves of broad bean (Vicia faba L) plants was studied to simulate the foliar uptake process of pesticides. The uptake rate of these model xenobiotics into bean foliage was measured using a standard leaf surface wash-off method. Diffusion into leaf tissues was visualised in vivo by confocal laser scanning microscopy (CLSM). The moderately lipophilic dye (Rhodamine B) showed faster uptake than the hydrophilic one (Oregon Green), despite the former being a larger molecule. While no distinct channels or domains for preferential entry of any of the dyes could be detected in the cuticle layer by CLSM, two different diffusion patterns were identified for the movement of these two dyes after traversing the cuticle. Upon desorption from the cuticle, Rhodamine B diffused extensively into the vacuole of the epidermal cells. Further transport of this dye from the epidermal cells to the mesophyll cells was not observed. In contrast, Oregon Green was found in the epidermal cell walls and cytoplasm, and was also present in the mesophyll cells. Examination of the petioles of the treated leaves revealed that, once absorbed, Oregon Green moved readily out of the treated leaf, whereas Rhodamine B did not show any phloem translocation. It is proposed that these two different diffusion characters may be responsible for the contrasting phloem mobility of the two xenobiotics. The results are discussed in relation to the current knowledge on the uptake, translocation and efficacy of pesticides as influenced by their properties.  相似文献

Stock assessments are often used to provide management advice, such as a total allowable catch (TAC), to fishery managers. Many stocks are not assessed annually, and the TAC from the previous assessment is often maintained in years between assessments. We developed two interim management procedures (MPs) that update the estimate of current vulnerable biomass from a surveyed index of abundance to adjust the TAC from a previous assessment. These MPs differ in how they handle uncertainty in observed indices. Using closed‐loop simulation, we evaluated the two interim MPs (with 10‐ and 5‐year assessment intervals) against several “status quo” approaches: (1) an annual assessment, and (2) a stock assessment every 5 or 10 years with (a) fixed TACs or (b) projections between assessments. We evaluated performance across three life‐history types and six operating model scenarios. The interim MPs performed similarly to annual assessments in terms of trends in biomass and yield, regardless of the assessment interval of the interim MPs. The interim MPs often produced more yield than the Fixed TAC MP with 10‐year assessment intervals, for example, in depleted scenarios. The Fixed TAC MP performed more similarly to interim MPs when the assessment interval for the Fixed TAC MP was decreased to five years. The interim MPs can also perform well when circumstances arise that are not accounted for in the Projection MP. Our results show that interim MPs should be considered for infrequently assessed stocks or rebuilding stocks, and highlight potential cost savings of interim MPs over annual assessments.  相似文献

We examine the effects of matrix type on forest bird species richness in historically fragmented forests of the KwaZulu-Natal midlands, South Africa. Bird species-area relationships for forest fragments within natural grassland were compared with those surrounded by plantation forestry (Pinus spp.). While fragments in grasslands displayed a species-area effect, no such effect (slope ∼0) was detected for fragments in plantations. The critical fragment size to avoid an island effect on species richness was 302 ha for fragments in grasslands, and contained 51 of the 61 forest bird species encountered. Small forest fragments (<50 ha) within grasslands were less species-rich than those surrounded by plantation, while the reverse was true for forests larger than 50 ha. Bird density was significantly lower in larger forest patches within plantation when compared to those in grassland. However, a slight decline in species density with increasing species richness in a patch (i.e. weak density compensation) characterised only those bird assemblages surrounded by a grassland matrix, suggesting these are approaching species saturation. Commercial plantations may increase the likelihood of colonisation of, and immigration from, small forest fragments by birds, reducing the incidence of area-dependent extinction in small fragments, but may also result in lower species richness in larger fragments. The latter is likely a consequence of the promotion by plantations of a wider distribution of generalist species in, and the loss of some specialised or rare species from, large patches. Management options are to avoid planting plantations near large forest patches and to increase the size of small patches where possible. Where afforestation is unavoidable, placing plantations in the vicinity of small forest patches rather than large forest patches may be preferable.  相似文献

Summary Simulated segregating barley populations were screened for resistance to scald (Rhynchosporium secalis) in the field at commercial seeding rates. A reduction in infection on the susceptible component occurred with increasing proportions of resistant genotypes. Similar trends were seen in space planted experiments but the use of susceptible buffer rows counteracted the effect, enhanced the infection in susceptible plants and greatly improved discrimination between resistant and susceptible. These results have been applied to the routine testing of F2 populations in the barley breeding programme.  相似文献

To evaluate the impacts of fishing on marine ecosystems, the total extraction of fish must be known. Putting a figure on total extraction entails the difficult task of estimating, in addition to reported landings, discards, illegal and unmandated catches. Unreported catches cast various types of shadow, which may be tracked and estimated quantitatively. Some shadows of unreported catches are reviewed, for example, an innovative, well‐funded NGO publicizes illegal catch in the Southern Ocean. For various reasons, official figures often have the implicit but unacceptable assumption that such categories are null. We present an estimation procedure based on adjustment factors taken from observer reports, correspondents and published information that track changes in a regulatory regime, and hence reflect incentives and disincentives to misreport. Monte Carlo simulations address uncertainty using multiple sources of information to provide upper and lower estimates. Once in place, this method provides preliminary estimates that may be refined without disruption. The method is demonstrated for fisheries in Iceland and Morocco. We use a ‘by‐species’ approach for Icelandic cod and haddock, while the Moroccan catch is divided into demersal and pelagic categories. Results suggest that Icelandic cod catches may have been underestimated by between 1 and 14% at different times, and haddock by between 1 and 28%. Underestimation of Moroccan catches appears to have been as much as by 50%. These case studies show that it is possible to obtain estimates of misreporting, even when direct data are lacking. Our method encourages transparency because sources of information are presented so that uncertain values are easily identified, offering a basis for comment, collaboration and refinement in estimating illegal and unreported fishing.  相似文献

Commercial abalone growers and aquaculture researchers need to monitor and compare the growth rate of their stocks. Growth rates based on both length and weights are often calculated, but the process of disturbance for measurement will affect the results of most studies. In natural populations, constant (linear) growth in length has been supported, while logarithmic transformations for growth rates (length and weight) have been used for aquaculture studies. This study of growth of greenlip abalone (Haliotis laevigata, Leach) over a prolonged period (3 years) indicates that both data treatments are suitable. If weights are being used, then a growth model that allows for exponential growth, such as specific growth rate (SGR), must be used. If lengths are measured, then the calculation of simple differences in the lengths over time is sufficient. The apparent paradox in use of two forms of measure is comfortably rationalized as a result of the weight–length relationship and the von Bertalanffy relationship for this species. Thus, methods of minimal disturbance and stress, such as photographic estimation of length, could be used to accurately monitor growth trends in aquacultural studies as well as in industry.  相似文献

Species responses to climate change are often measured at broad spatiotemporal scales, which can miss the fine-scale changes that are most relevant to conservation and fisheries management. We develop a scaleable geostatistical approach to assess how juvenile and adult fish distributions have been shaped by changes in bottom temperature and dissolved oxygen over a recent decade of warming in the northeast Pacific. Across 38 demersal fishes, biomass trends were associated negatively with warming and positively with dissolved oxygen, but when trends in both biomass and climate were converted to velocities—the speed and direction a population would have to move to maintain consistent conditions—the effect of temperature change differed depending on local conditions. In the warmest locations, warming velocities were associated with negative biotic velocities for 19 of 69 species-maturity combinations, and yet were almost always associated with stable or positive biotic velocities in the coolest locations (64 of 69). These spatially consistent biomass declines (negative biotic velocities) in the warmest locations and increases in cooler locations suggest a redistribution of species with the potential for new ecological and fisheries interactions. After controlling for temperature, the more spatially consistent effects of dissolved oxygen were often negative, suggesting a mechanism other than hypoxia avoidance—potentially changes in primary production. Our approach identifies the species and locations that are most sensitive to observed changes in the environment at any scale, thus facilitating future vulnerability assessments.  相似文献

A 2.5-year-old captive female mandrill (Papio sphinx) died following a protracted course of intermittent abdominal bloat, diarrhea, and severe weight loss. Necropsy revealed emaciation and marked gastrointestinal distention with gas and ingesta. Histologic evaluation revealed severe diffuse granulomatous enterocolitis and mesenteric lymphadenitis with massive numbers of 1-2-microm acid-fast bacilli within macrophages. Additionally, there was moderate to severe multifocal myocardial and vascular amyloidosis, moderate multifocal pyogranulomatous interstitial pneumonia with no acid-fast bacteria, and moderate multifocal glossal candidiasis. Samples of feces, ileum, and colon were positive for Mycobacterium avium subsp. paratuberculosis by radiometric culture and a polymerase chain reaction-amplified DNA probe specific for the insertion sequence IS900 of this organism.  相似文献
