Cancer relapse following chemotherapy has been attributed in part to the presence of cancer stem cells (CSC), which drive tumour growth and metastasis and are highly resistant to the effects of cytotoxic chemotherapy. As a result, treatment with cytotoxic chemotherapy selects for drug‐resistant CSC populations that eventually drive tumour recurrence. Little is known currently regarding the role of CSC in dogs with lymphoma, nor the impact of chemotherapy on CSC populations. Therefore, we prospectively quantitated CSC populations in dogs with B‐cell (BCL) and T‐cell lymphoma (TCL), using tumour aspirates and flow cytometric analysis with a panel of CSC markers. In addition, in vitro studies were carried out to determine the impact of chemotherapy resistance on the stem cell phenotype and stem cell properties of lymphoma cells. We found that the percentages of tumour cells expressing CSC markers were significantly increased in dogs with BCL, compared with B cells from normal lymph nodes. Similar findings were observed in dogs with TCL. In vitro studies revealed that lymphoma cells selected for resistance to CHOP chemotherapy had significantly upregulated expression of CSC markers, formed spheroids in culture more readily, and expressed significantly greater aldehyde dehydrogenase activity compared with chemotherapy‐sensitive tumour cells. Similar results were observed in tumour samples dogs with relapsed BCL. These findings suggest that cytotoxic chemotherapy can lead to a relative enrichment of tumour cells with CSC properties, which may be associated with lymphoma recurrence. 相似文献
Biodiversity in turbid aquatic environments is commonly assessed using extractive sampling methods that damage the seabed. Underwater cameras equipped with clear liquid optical chambers (CLOCs) for the assessment of seabed habitats and species are a non-extractive alternative and have been applied in turbid environments globally. A CLOC is a body of clear liquid positioned in front of a camera to reduce the scattering of light that would otherwise occur when passing through the turbid water it displaces. Here, we test and quantify the effectiveness of a CLOC for marine benthic biodiversity assessments over gradients of increasing turbidity.
The addition of a CLOC to a conventional benthic camera system significantly enhanced the quality of information gathered. Images acquired using the CLOC system consistently recorded statistically higher values of image quality (49% increase, based on the clarity of the image), seabed visible within the drop-down frame (34% increase), and European Nature Information System habitat level identification (49% increase). Furthermore, it was found that the ‘annotation success’ of taxa (classification of a specimen to family level or higher) was found to increase between individual experts in the presence of a CLOC. A reduced sampling effort was also identified when using a CLOC. Taxonomic richness increased by 27% when comparing the same number of image stills collected with and without the CLOC.
By reducing the limitations of underwater visibility previously attributed to underwater cameras, this concept extends the potential for use of non-destructive survey techniques and allows for future users to collect robust information of an area, making better informed management decisions.
Well-being is increasingly being promoted and used to describe social progress. However, tension exists between framings that focus on enhancing individual well-being (living well) and societal or collective framings of well-being (living well together). Well-being is central to Aotearoa New Zealand's COVID-19 response and recovery. The COVID-19 pandemic reopened debates about what kind of society people want to live in. Our research explored the ‘shared typical’ or commonality of experiences of the first wave of COVID-19 response in Aotearoa New Zealand. Semi-structured interviews provided insights into a wide range of concerns participants faced and what that meant for their well-being and the well-being of Aotearoa New Zealand. We found that well-being is both multidimensional and hierarchical, and while people talked about their own well-being, it was often in the context of broader social well-being. These findings support research showing that well-being is relational. We suggest that Indigenous models of well-being are well placed to inform policy strategies enabling holistic well-being, but this needs to be done in ways that pair Indigenous and Western knowledge, rather than integrating or assimilating this knowledge into Western science approaches. 相似文献
Master‐planned residential development has proliferated as a new residential phenomenon in metropolitan areas globally. The trend, the new governance mechanisms it entails and resultant forms of urban development have been critically theorised as products and vectors of neoliberalisation and iconic spaces of the neoliberal city. However, tracing the emergence and enactment of master‐planned residential estate (MPRE) development in Sydney, Australia, this paper suggests that more contingent and contextualised theorisations of such spaces can reveal possibilities for animating a different politics of MPREs. Deploying theorisation sensitive to the multiple drivers, logics and political projects played out through MPRE development in situated contexts, the paper traces the political genesis of these developments in Sydney, outlines the multiple drivers and logics accounting for their growing popularity and points to the salience of the complex performance of land and housing markets in their production. The post‐structural political economy approach used here to investigate MPRE development can overcome the politically constraining effects of the dominant neoliberal critique. It does so, first, by opening analysis up to the importance of logics, actions and contexts that are irreducible to neoliberalism and, second, by gesturing towards the potential for an alternative politics to be animated through mechanisms, techniques and processes of MPRE development habitually associated with neoliberalism. 相似文献
Abstract The International Center for Living Aquatic Resources Management (ICLARM) has demonstrated that coastal village communities in Solomon Islands can successfully farm giant clams. The production technology is simple and does not require a large capital investment. The main inputs are clam seed, labour and time. Labour is used for activities such as planting, cleaning, thinning and harvesting. In this paper, a bioeconomic model is used to explore optimal farm management for two species of giant clam fanned for the aquarium and seafood markets. The theoretical basis for this analysis is found in the economic theory of optimal forestry exploitation. Optimal management involves finding the combination of the decision variables and the cycle‐length that maximises a stream of discounted profits. The decision variables considered here are husbandry which relates to cleaning, and the frequency with which thinning is undertaken. The optimal cycle‐length is determined for both a single‐clam harvest and multiple harvests for various management scenarios. The labour requirements for these management scenarios are identified for the multiple‐harvest case and input substitution between optimal combinations of labour and cycle‐length is investigated. Results indicate that profits are maximised for both species when husbandry is excellent and labour usage is most intensive. Thinning is only necessary for seafood clams for which the optimal cycle‐length is longer. Village farmers may not be profit maximisers however, and labour spent on giant‐clam farming takes them away from other activities. Rather than investing more labour and harvesting the clams earlier, a village farmer with other objectives may devote less labour and harvest the clams later, and spend more time on other activities. In general, these results are consistent with extension advice provided to village farmers by ICLARM. Optimal solutions were found to be very stable when incorporated into global optimisation routines and sensitivity analysis of a wide range of parameter values. 相似文献
Canine B‐cell lymphoma is a clinically heterogenous disease; however, it is generally treated as a single disease entity. The purpose of this clinical trial was to prospectively evaluate naïve canine B‐cell lymphoma patients using histopathology, flow cytometry (FC) and a standardized chemotherapy protocol to better define subsets of this disease that may respond differently to treatment. Sixty‐four dogs with naïve multicentric B‐cell lymphoma were treated with a standardized 19‐week CHOP (cyclophosphamide, doxorubicin, vincristine, and prednisone) chemotherapy protocol. Most of the dogs (84.3%) were diagnosed with diffuse large B‐cell lymphoma (DLBCL), followed by nodal marginal zone (7.8%), small B‐cell (4.7%), Burkitt‐like (1.6%) and follicular lymphoma (1.6%). FC confirmed the diagnosis of B‐cell lymphoma in all cases. There were no clear phenotyping differences between the subtypes of B‐cell lymphoma detectable by our FC panel. The histologic subtypes in this study exhibited a range of forward scatter values on flow cytometry, but all of the DLBCL cases were higher than a value of 469, while the only cases with a lower forward scatter value were follicular lymphoma and diffuse small B‐cell lymphoma. Dogs with DLBCL had a significantly better objective response rate to the CHOP protocol (96.3%) than the non‐DLBCL subtypes (70%, P = .024). The median progression‐free survival time for patients with DLBCL (233 days) was significantly longer than that of all other histopathologic subgroups combined (163 days, P = .0005). 相似文献
We used stable isotopes of carbon and nitrogen to study the trophic niche of two species of insectivorous lizards, the Husab sand lizard Pedioplanis husabensis and Bradfield's Namib day gecko living sympatrically in the Namib Desert. We measured the δ13C and δ15N ratios in lizard blood tissues with different turnover times (whole blood, red blood cells and plasma) to investigate lizard diet in different seasons. We also measured the δ13C and δ15N ratios in available arthropod prey and plant tissues on the site, to identify the avenues of nutrient movement between lizards and their prey. Through the use of stable isotope mixing models, we found that the two lizard species relied on a largely non‐overlapping but seasonally variable array of arthropods: P. husabensis primarily fed on termites, beetles and wasps, while R. bradfieldi fed mainly on ants, wasps and hemipterans. Nutrients originating from C3 plants were proportionally higher for R. bradfieldi than for P. husabensis during autumn and late autumn/early winter, although not summer. Contrary to the few available data estimating the trophic transfer of nutrients in ectotherms in mixed C3 and C4/crassulacean acid metabolism (CAM) plant landscapes, we found that our lizard species primarily acquired nutrients that originated from C4/CAM plants. This work adds an important dimension to the general lack of studies using stable isotope analyses to estimate lizard niche partitioning and resource use. 相似文献
Poultry are important for many poor households in developing countries, but there are many constraints to poultry production, including disease. One of the most important diseases of chickens is Newcastle disease (ND). Even though there are effective vaccines against this disease available in most countries, uptake by small-scale poultry keepers is often low. In this study, two areas in Kenya and Tanzania were studied, where some villages had received additional support to get vaccination and other villages had not. In Kenya, 320 households from 10 villages were interviewed, of which half of the villages had active promotion of vaccination through village-based advisors. In Tanzania, 457 households were interviewed, of which 241 came from villages that have had active support through either a project or government extension services. Knowledge about vaccines and the attitudes towards vaccinating against ND was evaluated using mixed multivariable logistic models. Results indicate that in Kenya, the most important determinants for understanding the function of a vaccine were having had support in the village and to have knowledge about ND signs, while in Tanzania gender and previous vaccine use were important in addition to having had support. Attitudes towards vaccination were mainly determined by knowledge, where more knowledge about how vaccines work in general or about ND contributed to more positive attitudes. Among Kenyan farmers that had never used the vaccine before, the amount of birds they lost to disease and predators also influenced attitudes. In conclusion, this study supports the notion that knowledge is a very important component of extension support and that simply making vaccines available may not be sufficient for high levels of uptake.
The objective of this study was to determine if individual beef cows in a herd have an inherent ability to maintain their blood urea nitrogen (BUN) concentration when exposed to different levels of dietary nitrogen supplementation. Ten Hereford and 12 Nguni cows, aged between 2 and 16 years, were utilized in two crossover experiments. In the first experiment, cows were exposed to two diets: a balanced diet with a crude protein (CP) level of 7.9% and a modified diet with a CP level of 14%, formulated by adding 20 kg of feed grade urea per ton of the balanced diet. At the end of the first crossover experiment, cows received the balanced diet for 1 week. The second component utilized the same cows wherein they were fed the balanced diet in addition to another modified diet containing only 4.4% CP. Blood urea nitrogen concentration was measured 22 times (twice weekly) from each cow during both components of the study. A linear mixed-effects model was used to assess whether baseline BUN concentration (measured 1 week before onset of the study) was predictive of subsequent BUN concentration in individual cows. Breed, cow age, body condition score, and body mass were also evaluated for their effects on BUN concentrations. Albumin, beta hydroxybutyric acid (BHBA), glucose, and total serum protein (TSP) were compared between diets within each breed. Baseline BUN concentration was a significant predictor of subsequent BUN concentration in individual cows (P = 0.004) when evaluated over both components of the study. Breed (P = 0.033), the preceding diet (P < 0.001), current diet (P < 0.001), and the week during which sampling was performed (P < 0.001) were also associated with BUN concentration. Results suggest that beef cattle (within a herd) have an inherent ability to maintain their BUN concentration despite fluctuations in levels of available dietary nitrogen.
